To explore adaptation of avian influenza virus to mice we previously performed serial lung-to-lung passages of the influenza A/Chicken/Jiangsu/7/2002 (H9N2) strain, resulting in the isolation of a variant influenza strain lethal for mice. We now report that virulence correlates with improved growth characteristics on mammalian cells and extended tissue tropism in vivo. Sequencing of the complete genomes of the wild-type and mouse-adapted viruses revealed 25 amino acid substitutions. Some were found to reiterate known substitutions in human and swine H9N2 influenza isolates. Functions affected include nuclear localization signals and sites of protein and RNA interaction, while others are known determinants of pathogenicity and host specificity such as the viral polymerase PB2 E627K substitution. These observations suggest that enhanced growth characteristics and modified cell tropism may contribute to increased virulence in mice. We conclude that multiple amino acid substitutions are likely to be involved in the adaptation of H9N2 avian influenza virus to mice.  相似文献

The pharmacokinetic properties of drugs are closely related to their pharmacological efficacy. The kinetics of ivermectin are characterised, in general terms, by a slow absorption process, a broad distribution in the organism, low metabolism, and slow excretion. The kinetics vary according to the route of administration, formulation, animal species, body condition, age, and physiological status, all of which contribute to differences in drug efficacy. Characterisation of ivermectin kinetics can be used to predict and optimise the value of the parasiticide effects and to design programmes for parasite control. This article reviews the pharmacokinetics of ivermectin in several domestic animal species.  相似文献

为了解掌握养殖场中环境微生物对养殖业和公共卫生造成的影响,初步对我国养殖场环境微生物进行了研究。对吉林、湖北等省份的部分养猪场和养鸡场环境采样,并进行大肠杆菌和沙门氏菌分离鉴定,同时对从鸡舍和猪舍内分离大肠杆菌和沙门氏菌进行动物致病性实验及对13种抗生素的耐药性实验。实验结果表明,所有实验的沙门氏菌和90.7%的大肠杆菌菌株对小白鼠有致病性;所有分离菌株对磺胺类药物的耐药性非常严重,其次是对四环素,对其他药物也有不同程度的耐药性。  相似文献

本研究旨在探索意大利蜜蜂工蜂成蜂饲粮中铜的添加水平对蜜蜂生长发育、生理生化及抗氧化性能的影响。从蜂群中随机取3000只刚出房的蜜蜂,随机平均分成6组,分别命名为A、B、C、D、E、F,每组5个重复,每个重复100只蜜蜂;6组工蜂幼虫分别饲喂铜的添加水平为0.00、2.00、4.00、6.00、8.00、10.00μg/g的糖水,饲养至全部死亡。分别取成蜂测定体成分、血淋巴生化指标、抗氧化指标、相关基因表达量、制作中肠切片,并测定寿命。结果表明:(1)铜对蜜蜂寿命的影响显著,从蜜蜂存活率曲线图来看,当饲粮中铜的添加水平为2.00μg/g时对蜜蜂中肠食膜的结构最为有利。(2)与对照组相比,铜的添加水平为2.00~10.00μg/g组9日龄蜜蜂体组织铜含量显著升高(P<0.05),蜜蜂血淋巴中总蛋白(TP)和总胆固醇(TCHO)含量显著降低(P<0.05)。(3)9日龄蜜蜂蜂体超氧化物歧化酶(Cu/Zn-SOD)活性随着铜添加水平的提高出现先升高后降低的趋势,在2.00μg/g时活性最高(P<0.05)。(4)当饲粮中铜的添加水平在0.00~10.00μg/g的范围内不断升高时,CAT、Ctr1基因相对表达量显著增加(P<0.05)。由此得出饲粮中铜的添加水平对蜜蜂的生理机能影响显著。  相似文献

1. Apparent MEn and oil digestibility of hulled sunflower seed (SFS) and the products derived from the oil extraction process: press extracted SFS (PESFS), sunflower seed meal (SFSM), and press and solvent oils (PO and SO) were determined with 198 cockerels. Recombined products (mix of meal and oil) were also evaluated to study the effect of the 2 consecutive oil extraction steps. Each foodstuff was included in a basal diet, according to the proportions resulting from processing, at 100, 200, and 300 g SFS/kg. 2. Dietary energy value and digestible fat content were linearly related to rate of inclusion of test ingredients. Extrapolation values for AMEn (MJ/kg DM) were: SFS, 16.20; PESFS, 9.46; SFSM, 7.62. A decreasing quadratic trend was also found in the AMEn of PESFS, with interpolation values ranging from 11.77 to 9.33 MJ/kg DM between 70 and 210 g/kg DM of inclusion. No differences were observed between PO and SO. The AMEn of of sunflower oil, calculated from its digestibility, was 33.70 MJ/kg DM. 3. The oil extraction process affected the nutritional value of sunflower products. Recombined materials showed greater values than original foodstuffs: R-SFS, 17.47; R-PESFS, 11.49 MJ/kg DM. The increase in oil digestibility (from 0.814 to 0.862 g/kg DM in SFS; from 0.778 to 0.892 in PESFS) accounted for most of the increase observed in AMEn values. 4. As the form in which oil is incorporated in diets (released or within SFS or PESFS) affects the utilisation of sunflower products, their nutritional value is less than maximal and should not be calculated from their ingredients. Solvent oil seems to be the less available fraction of sunflower oil within SFS as the effect of the 2nd extraction proved to be greater than that of the previous press extraction.  相似文献

H. parasuis isolates (n=112) from pigs were tested for antimicrobial susceptibility against 23 antimicrobial agents by the disk diffusion method. All isolates were sensitive to Florfenico and most strains were sensitive to Cefotaxime (103/112; 92%), Ceftazidime (99/112; 88.4%), Chloramphenicol (90/112; 80.4%) and Gentamicin (85/112; 75.9%). High resistance levels to Nalidixic acid (84.8%), TMP (67.9%) and Trimethoprim+Sulfamethoxazole (58%) were observed. Genomic DNA extracted from 52 isolates resistant to at least seven antimicrobial agents was analyzed by PFGE and 46 distinct PFGE patterns identified. Diverse variation was observed between the drug-resistant H. parasuis isolates examined, suggesting that resistance traits were acquired independently by the respective isolates.  相似文献

Background: Upon binding luteinizing hormone in the ovary, the luteinizing hormone/choriogonadotropin receptor (LHCGR) is necessary for follicular maturation and ovulation, as well as luteal function. We detected mutations in the LHCGR gene and evaluated their association with superovulation. Methods: Using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and DNA sequencing, we examined polymorphisms in LHCGR and the genotypes associated with superovulation traits in 127 Chinese Holstein heifers. Results: A G/T polymorphism (ss52050737) in exon 11 was significantly associated with the total number of ova and the number of transferable embryos. Conclusions: LHCGR may be a new predictor for superovulation in Chinese Holstein heifers.  相似文献
