Analysis of glycoprotein I (gI) negative and aberrant pseudorabies viral diagnostic isolates.

Glycoprotein I (gI) phenotypes and genotypes of 4 pseudorabies viral diagnostic isolates were evaluated by use of in vitro DNA amplification, monoclonal antibody binding, gI-specific serodiagnostic responses, and in vivo virulence approaches. Three viruses were avirulent and did not elicit gI-specific serologic responses, react with gI-specific monoclonal antibodies, or contain gI epitope-encoding DNA sequences. The fourth virus was virulent and did elicit a gI-specific serodiagnostic response. Compared with reference virulent pseudorabies viruses, however, the fourth isolate had reduced reactivity with a group of gI monoclonal antibodies and had a single nucleotide sequence substitution with a corresponding putative amino acid change in the epitopically dominant portion of the gI molecule. Presumably, the first 3 isolates represented diagnostic recoveries of viruses derived from gI-deleted modified-live pseudorabies viral vaccines, whereas the fourth isolate was a virulent but gI-aberrant wild-type virus. Thoroughly assessing the gI status of pseudorabies viral diagnostic isolates was considered to be essential in evaluating the epidemiologic importance of these viruses and in monitoring the validity of gI-based vaccine companion tests now used worldwide in pseudorabies control and eradication programs.     

Pulmonary artery and aortic pressure changes during high intensity treadmill exercise in the horse: effect of frusemide and phentolamine.

Intravenous frusemide (1.0 mg/kg bwt) or phentolamine (0.33 mg/kg bwt) was given to 7 horses 1 h before exercise and their effects on pulmonary artery and aortic pressure changes during strenuous exercise were examined. Short-term near-maximal treadmill exercise (10 m/sec, 3 degrees incline) produced increases in heart rate, mean pulmonary artery pressure (PAP), mean aortic pressure (AP), and packed cell volume (PCV). Frusemide did not affect heart rate, PAP or PCV during exercise. Frusemide significantly decreased mean AP by 10 to 15 mmHg during exercise. Phentolamine produced an increase in heart rate relative to control only early in exercise but not during later, more strenuous, exercise. Phentolamine had no statistically significant effect on AP, PAP, or PCV, but a significant reduction was observed between 180 and 230 sec of exercise when PAP and AP were standardised against heart rate. Frusemide did not prevent horses from haemorrhaging during exercise in this study. Treatment with phentolamine did not sufficiently reduce the PAP and AP to test our hypothesis that a reduction in PAP and AP would eliminate EIPH.     

Large granular lymphoma in an FIV-positive and FeLV-negative cat

The clinical course of a feline leukaemia virus (FeLV)-negative and feline immunodeficiency virus (FIV)-positive cat affected with a large granular lymphocyte lymphoma is presented. Cyto-logical examination showed neoplastic cells in the pleural effusion and in two abdominal masses. Bone marrow and peripheral blood were moderately involved and chemotherapy was used to control the tumour. Cytochemistry, immunohis-tochemistry and ultrastructural studies were applied to define the cellular lineage; cytochemistry suggested a T-cell lineage.     

A recombinant-based feline immunodeficiency virus antibody enzyme-linked immunosorbent assay.

We have developed an antibody detection enzyme-linked immunosorbent assay (ELISA) for the identification of animals infected by feline immunodeficiency virus (FIV). The ELISA solid-phase antigen consists of recombinant FIV gag proteins expressed in bacteria. The proteins are purified from bacterial lysates as insoluble inclusion bodies. In the case of bacterially expressed p24gag, it is shown that all of the linear, sequential epitopes presented by viral p24 during infection are retained. Purified preparations can be substituted for solid-phase whole virus in the IDEXX PetChektm immunoassay. The antibody ELISA duplicates the sensitivity and specificity of the whole virus based PetChek plate assay.

选用１０２６只１周龄罗斯蛋用雏鸡为试验动物，研究不同日粮Ｓｅ水平对雏鸡组织Ｓｅ含量的影响。结果表明雏鸡的Ｓｅ缺乏或过量均可通过各组织Ｓｅ含量的变化反映出来，但反映的敏感性及临界指示值因组织及发育期不同而异。日粮Ｓｅ水平缺乏或过量时，各组织Ｓｅ含量相应降低或增加。在本试验条件下得出罗斯蛋用雏鸡的Ｓｅ需要量范围为０．１４５×１０￣（－６）～０．４９５×１０￣（－６），其中以０．１９５×１０￣（－６）最佳；日粮Ｓｅ含量≤０．０９５×１０￣（－６）或≥０．９９５×１０￣（－６），雏鸡出现Ｓｅ缺乏症或中毒症。     

Oscillating potentials on the B-wave of the ERG in the dog]

The oscillatory potentials (OP) on the b-wave of the canine ERG are characterized. Normal values for an OP-index, implicit times and periodicity are given. The OP-index increases during dark adaptation as well as with increasing stimulus intensities for white light flashes. Scotopic blue and red stimuli, although balanced for the maximum b-wave amplitude, surprisingly result in higher OP-indices for red flashes. Implicit times decrease for all OP with increasing stimulus intensities. Scotopic balanced red light results in OP with markedly reduced implicit times compared to blue light stimulation. The intervals between two OPs remain constant during dark adaptation, however, intervals between OPs with longer implicit times tend to be shorter. In contrast, the b-waves for scotopic balanced stimuli show rhythmic oscillations with constant intervals between them, the intervals being shorter for blue light than for red light. The intervals between OPs tend to become shorter with increasing stimulus intensities. This investigation indicates that OPs are influenced but not generated by photoreceptors. The examination of a dog with optic nerve hypoplasia indicates that the ganglion cells do not contribute to the biogenesis of OPs. The prognostic value of OPs in the early diagnosis for hereditary retinal degenerations in the dog is discussed.     

Serum amyloid A protein (SAA) in horses: objective measurement of the acute phase response

A sensitive and precise immunoassay for equine serum amyloid A protein (SAA) was established and used to determine, for the first time, the circulating concentration of this protein in health and disease. As in other species, equine SAA was present only at trace levels in healthy animals but behaved as an extremely sensitive and rapidly responding acute phase reactant following most forms of tissue injury, infection and inflammation, objectively reflecting the extent and activity of disease. Measurements of SAA should make a significant contribution to diagnosis and management of viral and bacterial infection in horses, and routine serial assays could provide an objective criterion for monitoring prospectively the general health of horses in training and racing.     

The oxygen uptake capacity of swine

Comprehensive studies into domestic pigs and wild boars together with literature data provided a basis for an assessment of aerobic metabolic capacity (VO2max) of swine. The values quoted, from 20 to 94 ml/min-1/kg, had been due to several factors of methodological approach, growth, training, and nutrition as well as to pathophysiological aspects. For full capacity utilisation of VO2max, untrained pigs can be challenged at belt velocities between 0.8 and 1.8 m/s-1 and trained animals at 5 m/s-1.     

Presumptive subcutaneous surgical transplantation of a urinary bladder transitional cell carcinoma in a dog

A urinary bladder mass in a 12-year-old spayed female West Highland White Terrier was diagnosed after exploratory surgery and biopsy as a transitional cell carcinoma. Four months later the dog presented with an ulcerated plaque-like cutaneous lesion at the previous surgical incision site; concurrent inguinal lymphadenopathy and recurrence of the urinary bladder mass were identified. Transitional cell carcinoma was diagnosed at all 3 sites. Although a definitive relationship cannot be established between the initial surgery for urinary bladder mass and the resultant subcutaneous lesion, surgical implantation should be considered as a source for the neoplastic cells.
