Kernel pericarp wax of the corn breeding population GT-MAS:gk has been associated with resistance to Aspergillus flavus infection and aflatoxin production. GT-MAS:gk wax, previously compared to waxes of three susceptible genotypes, was presently compared to wax of a different, and more numerous, group of susceptible lines. Wax separation by TLC confirmed previous findings, demonstrating a unique GT-MAS:gk band and a unique "susceptible" band. Only GT-MAS:gk wax inhibited the growth of A. flavus; however, no association was established, as before, between kernel wax abundance and resistance. Gas chromatography-mass spectroscopy (GC-MS) analysis of kernel whole wax showed a higher percentage of phenol-like compounds in wax from GT-MAS:gk than in waxes from the susceptible lines. The GT-MAS:gk unique band contained phenol-like compounds and ethyl-hexadecanoate; butyl-hexadecanoate was preeminent in most of the "susceptible bands". Alkylresorcinol (phenolic compounds) content was dramatically higher in GT-MAS:gk wax than in the wax of susceptible lines. An alkylresorcinol, 5-methylresorcinol, also inhibited in vitro growth of A. flavus. These and other phenolic compounds may contribute to kernel wax inhibition of A. flavus infection/aflatoxin production. Further investigation is needed to confirm a role for them in GT-MAS:gk resistance.  相似文献

Increased root exudation and a related stimulation of rhizosphere-microbial growth have been hypothesised as possible explanations for a lower nitrogen- (N-) nutritional status of plants grown under elevated atmospheric CO2 concentrations, due to enhanced plant-microbial N competition in the rhizosphere. Leguminous plants may be able to counterbalance the enhanced N requirement by increased symbiotic N2 fixation. Only limited information is available about the factors determining the stimulation of symbiotic N2 fixation in response to elevated CO2.In this study, short-term effects of elevated CO2 on quality and quantity of root exudation, and on carbon supply to the nodules were assessed in Phaseolus vulgaris, grown in soil culture with limited (30 mg N kg−1 soil) and sufficient N supply (200 mg N kg−1 soil), at ambient (400 μmol mol−1) and elevated (800 μmol mol−1) atmospheric CO2 concentrations.Elevated CO2 reduced N tissue concentrations in both N treatments, accelerated the expression of N deficiency symptoms in the N-limited variant, but did not affect plant biomass production. 14CO2 pulse-chase labelling revealed no indication for a general increase in root exudation with subsequent stimulation of rhizosphere microbial growth, resulting in increased N-competition in the rhizosphere at elevated CO2. However, a CO2-induced stimulation in root exudation of sugars and malate as a chemo-attractant for rhizobia was detected in 0.5-1.5 cm apical root zones as potential infection sites. Particularly in nodules, elevated CO2 increased the accumulation of malate as a major carbon source for the microsymbiont and of malonate with essential functions for nodule development. Nodule number, biomass and the proportion of leghaemoglobin-producing nodules were also enhanced. The release of nod-gene-inducing flavonoids (genistein, daidzein and coumestrol) was stimulated under elevated CO2, independent of the N supply, and was already detectable at early stages of seedling development at 6 days after sowing.  相似文献

The effect of Zn deficiency in wheat (Triticum aestivum L. cv. Ares) on the release of Zn mobilizing root exudates was studied in nutrient solution. Compared to Zn sufficient plants, Zn deficient plants had higher root and lower shoot dry weights. After visual Zn deficiency symptoms in leaves appeared (15–17 day old plants) there was a severalfold increase in the release of root exudates efficient at mobilizing Zn from either a selective cation exchanger (Zn-chelite) or a calcareous soil. The release of these root exudates by Zn deficient plants followed a distinct diurnal rhythm with a maximum between 2 and 8 h after the onset of light. Re-supply of Zn to deficient plants depressed the release of Zn mobilizing root exudates within 12 h to about 50%-, and after 72 h to the level of the control plants (Zn sufficient plants). The root exudates of Zn deficient wheat plants were equally effective at mobilizing Fe from freshly precipitated FeIII hydroxide as Zn from Zn-chelite. Furthermore, root exudates from Fe deficient wheat plants mobilized Zn from Zn-chelite, as well as Fe from FeIII hydroxide. Purification of the root exudates and identification by HPLC indicated that under Zn as well as under Fe deficiency, wheat roots of the cv. Ares released the phytosiderophore 2′-deoxymugineic acid. Additional experiments with barley (Hordeum vulgare L. cv. Europa) showed that in this species another phytosiderophore (epi-3-hydroxymugineic acid) was released under both Zn and Fe deficiencies. These results demonstrate that the enhanced release of phytosiderophores by roots of grasses is not a response mechanism specific for Fe deficiency, but also occurs under Zn deficiency. The ecological relevance of enhanced release of phytosiderophore also under Zn deficiency is discussed.  相似文献

Using an automated rapid headspace solid phase microextraction (SPME) method for volatile extraction in cantaloupes, 86 compounds already reported for muskmelons were recovered and an additional 53 compounds not previously reported were identified or tentatively identified. The SPME method extracted a copious number of volatiles that can be analyzed to clearly differentiate between variety, growth stage, and stage of harvest ripeness. Most of the newly reported compounds in cantaloupe were esters and aldehydes that have already been demonstrated as flavor-related compounds in other products. All esters believed to have flavor impact increased progressively after pollination, and this trend continued with increasing harvest maturity. However, compound recovery often decreased when fruits were harvested over-ripe. Most aldehydes increased during early growth stages and then tapered off with increasing harvest maturity. The SPME method suitably recovered most compounds reported to impart characteristic flavor/aroma in muskmelons. SPME offers experimental flexibility and the ability to discover more compounds and address flavor quality changes in fresh-cut cantaloupe.  相似文献

The aim of this study was to produce in vitro oxidation products of lycopene, which could be possible in vivo metabolites. An oxidation of lycopene with potassium permanganate gave a range of lycopene degradation compounds resulting from the oxidative cleavage of one or two carbon-carbon double bonds. Eleven apo-lycopenals/ones and six apo-carotendials were obtained and tentatively characterized by HPLC-DAD-MS. Apo-11-lycopenal and apo-8,6'-carotendial were isolated and characterized by (1)H NMR for the first time. Lycopene was submitted to an oxidation by atmospheric oxygen catalyzed by a metalloporphyrin, a model system of the active center of cytochrome P450 enzymes. (Z)-Isomers, monoxides, and cleavage compounds of (E)-lycopene were formed. We propose a mechanism of oxidation of lycopene by this system.  相似文献

The aim of this study was to examine genetic variation in desiccation tolerance in cocoons of the parthenogenetically reproducing earthworm Dendrobaena octaedra by comparing populations originating from different geographic regions (Denmark, Norway and Finland), representing large differences in precipitation and temperature. In one experiment, the tolerance of the three populations to increasing desiccation stress in the range from 100 to 91.6% relative humidity (RH) was examined, aiming to represent ecologically relevant RH values. In a second experiment, the effect of cocoon size on desiccation tolerance was investigated at 92.3% RH in the same three populations. There were highly significant differences in desiccation tolerance between populations, indicating a high genetic differentiation of this trait in D. octaedra. Cocoons from Denmark were much more sensitive (71±14% mortality at 91.6% RH) than cocoons from Norway (21±4% mortality) and Finland (4±5% mortality). Cocoons of worms from Finland and Norway were significantly larger than cocoons produced by worms from Denmark suggesting that cocoons from Denmark lost water at a higher rate when subjected to low humidity. Assuming that slow dehydration is necessary for physiologically based protection mechanisms it may be expected that desiccation tolerance is positively correlated with cocoon size. However, within each of the populations cocoon fresh weight did not have any significant impact on desiccation tolerance. When all populations were pooled there was a significant positive effect of cocoon fresh weight on desiccation tolerance, explaining about 20% of the total variation (linear regression). It seems therefore that genetic variation of desiccation tolerance in D. octaedra cocoons is related to variation in both cocoon size and other, physiologically based tolerance mechanisms.  相似文献

In 50 domestic pigeons, two circular pieces were removed from the skin to the right and left of the crista sterni, and the wounds were treated in different ways: A. with a collagen membrane dampened with a sodium chloride solution, B. with a fibrin glue, C. with a combination of A) and B). The wounds of group D were left untreated. Scab formation could be seen in all cases, also under the transparent membrane. The collagen membrane had no protective function in the experiment, however, it contributed to the acceleration of re-epithelialisation and the reduction of the wound diameter. The additional application of fibrin glue improved wound healing even further.  相似文献

Anaesthesia in birds is ordered by law and is also necessary for various operations and manipulations. Anaesthesia by injection of Ketamin, which in special cases may be combined with Diazepam, has been found useful. Anaesthesia by inhalation with Halothan, Methoxyfluran or Isofluran is the most careful method. Local anaesthesia has few indications.  相似文献
