Estimation of the plant-available water capacity (PAWC) of soils at a regional scale helps in adopting better land use planning, developing suitable irrigation schedules for crops, and optimizing the use of scarce water resources. In the current study, 72 soil profiles were sampled from the Barossa region of South Australia to estimate pedo-transfer functions deduced from easily estimated soil properties. These functions were then used to estimate the fixed (10 and 33 kPa) and dynamic pressure head (hfc) water contents at field capacity (FC) for minimum drainage flux (0.01 and 0.001 cm d-1), which serves as the upper boundary for plant-available water in soils. The estimated residual water content was corrected for subsoil constraints, especially the exchangeable sodium percentage (ESP). The results showed that the mean values of hfc in sand-dominated light and medium textured soils (i.e., sand, loamy sand, sandy loam, and loam) varied in a narrow range (15.8–18.2 kPa), whereas those in the clay-dominated heavy textured soils (i.e., clay loam) showed a wide range (11.3–49.3 kPa). There were large differences in PAWC for dynamic FC (PAWCfc) and fixed FC at 10 kPa (PAWC10), 33 kPa (PAWC33), and a mix of 10 and 33 kPa (PAWC10,33) pressure heads depending on soil texture. Normally, the difference between PAWC at 10 kPa and hfc (∆PAWC10) was positive, whereas that between 33 kPa and hfc (∆PAWC33) was negative across all sites. Nevertheless, the estimation of PAWC assuming a fixed FC at 10 and 33 kPa pressures (i.e., PAWC10,33) for sandy, clay, and silty soils reduced the difference between fixed and dynamic pressure PAWCs to < 10% across the region. The estimation of PAWC was improved by incorporating the impact of subsoil constraints, such as high ESP, which was more pronounced for clay and silty soils. These findings demonstrate the inherent inconsistencies between static pressure and flux-based dynamic FC estimations in soils. Soil heterogeneity, intra-texture variability, subsoil constraints, and swell-shrink clays can have great impacts on the water retention capacity in response to dynamic and fixed pressure FC values. 相似文献

SummaryA dot blot assay, based on the internal transcribed spacer of the ribosomal region, was developed for rapid diagnosis and identification of Botrytis cinerea, the causal agent of grey mould of greenhouse tomatoes. In this assay, DNA from fungal cultures and diseased plant tissue were amplified by PCR using primers specific to septate fungi, fixed to nylon membranes, and hybridized with digoxigenin-d-UTP labelled oligonucleotides specific to B. cinerea. The hybridized probes were detected by chemiluminescence. Six isolates of B. cinerea, four isolates of Botrytis species, one isolate each of B. fabae, B. squamosa, B. streptothrix, B. allii, B. porri, and B. tulipae and some common greenhouse pathogens were tested using this assay. Of four probes screened, probe Bot1 gave a positive reaction with all isolates of B. cinerea and with fresh or frozen plant tissue affected with grey mould from research centres and commercial greenhouses. All other greenhouse fungi tested negative with probe Bot1. This dot blot diagnoses infection by B. cinerea in large numbers of samples rapidly and reliably and distinguishes isolates of B. cinerea from other epiphyte fungi found on greenhouse crops. 相似文献

Helicoverpa armigera (Hübner) (Noctuidae: Lepidopetra) is a polyphagous pest of major crops grown in India. To prevent the damage caused by H. armigera farmers rely heavily on insecticides of diverse groups on a regular basis which is not a benign practice, environmentally and economically. To provide more efficient and accurate information on timely application of insecticides, this research was aimed to develop a forecast model to predict population dynamics of pod borer using Autoregressive Integrated Moving Average (ARIMA) and Artificial Neural Networks (ANN). The data used in this study were collected from the randomly installed sex pheromone traps at International Crops Research Institute for the Semi-arid Tropics (ICRISAT), Patancheru, Hyderabad. Several ARIMA (p, d, q) (P, D, Q) and ANN models were developed using the historical trap catch data. ARIMA model (1,0,1), (1,0,2) with minimal BIC, RMSE, MAPE, MAE, and MASE values and higher R2 value (0.53) was selected as the best ARIMA fit model, and neural network (7-30-1) was found to be the best fit to predict the catches of male moths of pod borer from September 2021 to August 2023. A comparative analysis performed between the ARIMA and ANN, shows that the ANN based on feed forward neural networks is best suited for effective pest prediction. With the developed ARIMA model, it would be easier to predict H. armigera adult population dynamics round the year and timely intervention of control measures can be followed by appropriate decision-making schedule for insecticide application. 相似文献

Bluetongue (BT), a serious disease of sheep and some wild ruminants, is caused by bluetongue virus (BTV), a member of the family, Reoviridae. The current research thrust for controlling BT is on development of efficient vaccines, necessitating clear understanding of ovine immunology. At present, comparative studies on cytokine gene expression profiles of na?ve and BTV-sensitized peripheral blood mononuclear cells (PBMC) in sheep have not been clearly understood. In the present study, PBMC from na?ve and BEI-inactivated-saponin-adjuvanted BTV-1 vaccinated sheep were stimulated in vitro with heterologous BTV-23. At various intervals, RT-qPCR was carried out to estimate cytokine (interferon-gamma, interleukin-12 and interleukin-2) mRNA expressions that are linked to cell-mediated immunity. The results showed that PBMC cytokine profiles were relatively increased both temporally and quantitatively in immunized sheep PBMC compared to na?ve ones, suggesting that BTV-1 vaccination may prime immune system that can cross-react with BTV-23 antigens. 相似文献

Drought stress is presently a major productivity limiting factor in wheat. This study developed five wheat lines with inbuilt tolerance to drought stress using marker‐assisted backcross breeding (MABB) approach employing three linked quantitative trait loci (QTLs) in an initial population of 516 BC1F1 plants. The high‐yielding wheat cultivar ‘HD2733’ grown over last few years extensively in the eastern plains of India is largely sensitive to drought and is used as the recurrent parent. ‘HI1500’ released for water‐limiting conditions and carrying drought‐tolerant QTLs was used as donor parent. MABB lines were advanced using foreground and background selection, coupled with stringent phenotyping. We identified 29 lines that were homozygous for targeted QTLs in different combinations with background recovery range of 89.2%–95.4%. Further evaluation of selected lines for physiological traits and distinctness, uniformity and stability (DUS) characters under rainfed condition identified five potential varieties for national varietal evaluation programme in the zone. Iron deficiency chlorosis (IDC) causes a significant reduction in yield of groundnut grown in calcareous and alkaline soils in India. The main aim of the study was to assess genotypic differences for morpho-physiological parameters associated with IDC resistance across different stages and their effect on yield and its related traits. The factorial pot experiment was comprised of two major factors, i) soil-Fe status [normal-Fe, deficit-Fe], and ii) genotypes [five] with differential IDC response, constituting 10 treatments. They were assessed for five morpho-physiological parameters associated with IDC resistance across five crop growth stages and also yield and its related traits. Associations between these traits were also estimated. Under deficit-Fe conditions, IDC resistant genotypes recorded significantly lower visual chlorosis rating (VCR), higher SPAD values, active Fe, chlorophyll content, peroxidase activity, and high yield compared to susceptible ones. Between normal- to deficit-Fe soils, resistant compared to susceptible genotypes showed no change in VCR scores; a lower reduction in SPAD, chlorophyll, active Fe, peroxidase activity, and pod yield. Under deficit-Fe conditions, high yield among resistant genotypes could be attributed to higher seed weight, number of pods and haulm yield, while contrasting reduction in main stem height and number of primaries. The results indicate that for initial large-scale screening of groundnut genotypes for IDC resistance, SPAD values are most ideal while active Fe could be utilized for confirmation of identified lines. 相似文献

Insulin and glucose concentrations were measured by radioimmunoassay and by the oxygen rate method, respectively. Data were analyzed with anova and Bonferroni post hoc (5% significance) and are shown as mean ± SD.ResultsPlasma insulin and glucose concentrations were similar between WT and CD38?/? mice under control conditions. As compared to controls, medetomidine administration produced a statistically significant decrease in plasma insulin concentrations in the WT mice whereas the decrease in the CD38?/? mice was not statistically significant. Two experiments were conducted to study the effect of methionine- and lysine-supplemented raw and hydrothermically processed full-fat soybean as a dietary protein source on growth performance, digestibility, and digestive enzyme activity in grey mullet, Mugil cephalus (mean BW 0.55 g), and milkfish, Chanos chanos (mean BW 3.83 g). Eight isocaloric (gross energy about 18.27 kj g?1) and isonitrogenous (crude protein about 40%) diets (1–4 = raw soybean based; 5–8 = processed full-fat soybean based) were formulated. A fishmeal-based diet was used as the reference/control diet. Growth performance, nutrient retention, feed conversion ratio (FCR), and protein digestibility in fishes correlated well with the trypsin inhibitor activity levels of the diets. Carcass phosphorous levels were significantly (P < 0.05) higher in fish fed the fishmeal control diet. Irrespective of the fish species, significantly (P < 0.05) lower levels of total ammonia excretion and reactive phosphate production (mg kg?1 BW d?1) were recorded where the fish were fed processed full-fat soybean diets compared with the fish fed fishmeal or raw soybean diets. Among these diets, the best growth performance and the lowest levels of metabolic waste were obtained in fishes fed the methionine- and lysine-supplemented processed soybean-based diets containing no fishmeal. 相似文献
