Hemostatic biomarkers in dogs with chronic congestive heart failure   总被引:1,自引:0,他引:1
BACKGROUND: Chronic congestive heart failure (CHF) in humans is associated with abnormal hemostasis, and abnormalities in hemostatic biomarkers carry a poor prognosis. Alterations in hemostatic pathways can be involved in the pathogenesis of CHF in dogs, and microthrombosis in the myocardium could contribute to increased mortality. HYPOTHESIS: That plasma concentration or activity of hemostatic biomarkers is altered in dogs with CHF and that these factors predict mortality. ANIMALS: Thirty-four dogs with CHF caused by either dilated cardiomyopathy (DCM, n=14) or degenerative valvular disease (CDVD, n=20) compared with 23 healthy age-matched control dogs were included in this study. Dogs with CHF were recruited from 2 referral cardiology clinics, and control dogs were owned by friends or colleagues of the investigators. METHODS: Clinical examination and echocardiography were performed in all dogs. Plasma fibrinogen and D-dimer concentrations, antithrombin and protein C activity, and thrombin-antithrombin complex (TAT) were measured in all dogs. RESULTS: Dogs with CHF had significantly higher fibrinogen (P = .04), D-dimer (P = .002), and TAT concentration (P < .0001), lower antithrombin (P < .0001) and protein C activity (P < .001) compared with control dogs. None of the hemostatic biomarkers were associated with risk of death. CONCLUSIONS AND CLINICAL IMPORTANCE: There is evidence of a procoagulant state in dogs with CHF. The lack of predictive value for survival might be due to the small number of dogs examined. Further studies are necessary to investigate the presence and importance of microthrombosis in dogs with CHF.  相似文献

Populations of the marble trout (Salmo marmoratus) are critically declining due to introgression by brown trout (S. trutta) strains. Hybrids between the two forms are fertile and presently predominant in most rivers of the species’ range. The involved hybridization mechanisms have been studied through two large scale in-stream experiments (Driselpoh and Stopnikarca) as 50% of each species have been stocked at the age of one year in fishless streams, each fish being individually marked. Diagnostic molecular markers were applied to test a partial reproductive isolation between the two species. Emphasis was put on survival and growth patterns of stocked fish (parental generation) as well as on fish hatched within the stream (F1 generation). No evidence of a partial reproductive barrier between the two species was observed. Survival of the parental generation depended on the year in both streams, as well as on the species in Stopnikarca, but was identical for both species in Driselpoh. In both streams survival of instream hatched individuals measured from 0+ to 2+ was lower for brown trout. In Driselpoh, F1 0+, 1+ and 2+ hybrids were larger than pure individuals. Larger hybrids were only observed in Stopnikarca when analyses focused on individuals in inter-specific competition suggesting that heterosis and stress effects may explain the observed size differences. Our results point out important ecological differences between marble and brown trout and have shown that hybridization can easily take place. The findings indicate that high F1 hybrid presence, survival and heterosis effects may impede marble trout rehabilitation in the area.  相似文献

In this study we quantify the partitioning of recent assimilates to above- and below-ground carbon (C) pools in two subarctic mire ecosystems - wet minerotrophic and semiwet ombrotrophic mire - using in situ 14C pulse-labelling. Ecosystem C partitioning to rhizomes, coarse roots, fine roots, dissolved organic carbon (DOC) and microbes were quantified twice  during the growing season at three different soil depths. Finally the 14C-partitioning data from this and a previous study were combined to estimate the overall C partitioning of the three main vegetation types of a Scandinavian subarctic mire in early and late summer. The semiwet ombrotrophic ecosystem hosted a much larger root biomass on an area basis compared to the wet minerotrophic ecosystem which might be due to differences in the soil nutrient level. Microbial C was found to be the largest C-pool in both ecosystems. Ecosystem 14C partitioning was poorly related to plant biomass for the semiwet and the wet ecosystem. Overall a higher partitioning of recent assimilates to below-ground compartments was apparent in August-September compared to June-July, while the opposite was found for the above-ground C-pools. In the semiwet ecosystem twice as much 14C was found in DOC compared to the wet ecosystem, where root density, litter and above-ground biomass were important controls of the 14C-recovery in DOC. Plant-derived DOC was estimated to be 15.4 versus 12.9 mg C m−2 d−1 in the semiwet and wet ecosystem, respectively.Graminoid dominated and dwarf shrub dominated vegetation types of the subarctic mire Stordalen differ with respect to the relative amount of recently assimilated C partitioned to C-pools with “slow” versus “fast” decomposition rate. The capacity for sequestration of recently fixed C within “slow” C-pools might affect the ecosystem C balance (NEE) and C-storage. The potential for vegetation changes might therefore be an important factor to consider in studies of response of ecosystem C-dynamics to global change factors in subarctic mires.  相似文献

Forest management faces growing uncertainty concerning environmental conditions and demand for ecosystem services. To help forest managers consider uncertainty, we applied a robust and multi-criteria approach to select the optimal composition of a forest enterprise from 12 stand types. In our simulation, the forest enterprise strives for either financial return or a multi-criteria forest management considering financial return, carbon storage and forest ecosystem stability. To quantify the influence of climate change on these decision criteria, we used the concept of analogous climate zones. Our results provide recommendations for long-term strategies for tree species selection in a Southeast German forest enterprise. The results show that considering both uncertainty and multifunctionality in forest management led to more diversified forest compositions. However, robust and multi-criteria optimisation required the forest enterprise to pay a premium in terms of lower income. Financial returns decreased when forest composition accounted for uncertainty or multiple objectives. We also found that adaptation measures could only partly financially compensate the effects of climate change. As the study is limited to two tree species, including additional tree species, variants of mixing proportions and further silvicultural strategies in the optimisation appears a promising avenue for future research.

In this study, we reasoned that if we challenged rainbow trout with the causative agents of enteric redmouth disease (ERM), rainbow trout fry syndrome (RTFS), and viral haemorrhagic septicaemia (VHS), we would: 1) detect additive genetic variation for resistance to ERM, RTFS, and VHS; and 2) find that resistance of the trout to ERM and RTFS are favourably correlated genetically, while resistance to VHS is unfavourably correlated with resistance to ERM and RTFS. We tested these premises by challenging 63 full-sib families of rainbow trout (50 sires, 38 dams) with Yersinia ruckeri, Flavobacterium psychrophilum, and VHS virus, the causative agents of ERM, RTFS, and VHS. Resistance to each disease was assessed as both a binary trait (i.e., died/survived) and a longitudinal trait (i.e., time until death following challenge). Additive genetic variation and genetic correlations for resistance to ERM, RTFS, and VHS were estimated by fitting a threshold liability model to resistance assessed as a binary trait. As a longitudinal trait, additive genetic variation and genetic correlations were estimated by fitting a Weibull frailty model to the times until death. Our findings support the first of our premises as we detected additive genetic variation for resistance to ERM, RTFS, and VHS. The heritability for resistance to ERM, RTFS, and VHS ranged between 0.42 and 0.57 on the underlying liability scale when resistance was assessed as a binary trait. As a longitudinal trait, the heritabilities ranged between 0.07 and 0.21 for time until death on the logarithmic-time scale. We were, however, unable to support our second premise as we found that resistance to each of the diseases tended to be weakly correlated genetically. The genetic correlations between the resistances ranged between −0.11 and 0.15 when resistance was assessed as a binary trait, and between −0.23 and 0.16 when resistance was assessed as a longitudinal trait. These findings are encouraging for commercial trout production. The additive genetic variation detected for resistance demonstrates that selectively breeding trout for resistance to ERM, RTFS, and VHS will be successful, providing a complementary approach to control these diseases. The weak genetic correlations suggest that it should be relatively easy to improve resistance to each of the diseases simultaneously.  相似文献

Lipase-catalyzed interesterification of butterfat blended with rapeseed oil (70/30, w/w) was investigated both in batch and in continuous reactions. Six commercially available immobilized lipases were screened in batch experiments, and the lipases, Lipozyme TL IM and Lipozyme RM IM, were chosen for further studies in a continuous packed bed reactor. TL IM gave a fast reaction and had almost reached equilibrium with a residence time of 30 min, whereas RM IM required 60 min. The effect of reaction temperature was more pronounced for RM IM. TL IM showed little effect on the interesterification degree when the temperature was raised from 60 degrees C to 90 degrees C, whereas RM IM had a positive effect when the temperature was increased from 40 degrees C to 80 degrees C. Even though TL IM is an sn-1,3 specific lipase, small changes in the sn-2 position of the triacylglycerol could be seen. The tendency was toward a reduction of the saturated fatty acid C14:0 and C16:0 and an increase of the long-chain saturated and unsaturated fatty acids (C18:0 and C18:1), especially at longer residence times (90 min). In prolonged continuous operation the activity of TL IM was high for the first 5 days, whereafter it dramatically decreased over the next 10 days to an activity level of 40%. In general, the study shows no significant difference for butterfat interesterification in terms of enzyme behavior from normal vegetable oils and fats even though it contains short-chain fatty acids and cholesterol. However, the release of short-chain fatty acids from enzymatic reactions makes the sensory quality unacceptable for direct edible applications.  相似文献

Earthworms in pastureland may be exposed to residues of veterinary medicines excreted in the faeces of treated cattle. Possible sub-lethal effects of these drugs on life history characteristics are critical to earthworm populations, but there are no internationally accepted guidelines for the laboratory testing that is essential as a supplement to field studies. In the laboratory, Lumbricus terrestris were kept in artificial soil and offered dung collected from cattle with sustained-release boluses of ivermectin or fenbendazole, or from untreated cattle (control). Earthworms were therefore exposed to drug residues in concentrations excreted naturally by treated cattle. Furthermore, worms were exposed to the drug in the natural way, i.e. by ingestion. A matrix model based on survival and reproductive rates as functions of life-stages was used to quantify the population dynamics of L. terrestris. There were no adverse effects of either ivermectin or fenbendazole on the individual survival and growth of L. terrestris. Mature worms produced on average 2-3 cocoons per worm per month irrespective of drug treatment, and about 50% of these cocoons hatched successfully. The median time to hatching (incubation time) depended on the age of the parent worms and was significantly lower for cocoons in the ivermectin group compared to controls, as the worms became older. The incubation time varied considerably in all groups; even within a batch of cocoons produced during 1 month by one pair of worms, the period sometimes varied by more than 1 year. The intrinsic rate of natural increase (r≈0.0044 d−1) was not affected by either drug treatment. The methods and the model developed in this study are suitable for long-term studies on L. terrestris and give ecologically relevant information on population dynamics. There were no negative effects of ivermectin and fenbendazole on life history characteristics of L. terrestris and it is unlikely that earthworm populations will be affected in the field following normal use of sustained-release boluses in cattle. Finally, the matrix model points to adult survival rate and cocoon viability as the most important variables to be included in future ecotoxicological tests on L. terrestris.  相似文献
