Objective

To characterise the minimum dose of intramuscular alfaxalone required to facilitate intubation for mechanical ventilation, and to investigate the impact of cranial versus caudal injection on anaesthetic depth.

Study design

Randomised crossover study.

Animals

Six healthy juvenile ball pythons (Python regius).

Methods

Three dosages (10, 20 and 30 mg kg–1) of alfaxalone were administered to each python in a caudal location with a minimum 2 weeks washout. Induction and recovery were monitored by assessing muscle tone, righting reflex, response to a noxious stimulus and the ability to intubate. A subsequent experiment assessed the influence of injection site by comparing administration of 20 mg kg–1 alfaxalone in a cranial location (1 cm cranial to the heart) with the caudal site. Respiration rate was monitored throughout, and when intubation was possible, snakes were mechanically ventilated.

Results

Regardless of dose and injection site, maximum effect was reached within 10.0 ± 2.7 minutes. When administered at the caudal injection site, intubation was only successful after a dosage of 30 mg kg-1, which is higher than in previous reports for other reptiles. However, intubation was possible in all cases after 7.2 ± 1.6 minutes upon cranial administration of 20 mg kg–1, and anaesthetic duration was significantly lengthened (p < 0.001). Both 30 mg kg–1 at the caudal site and 20 mg kg–1 at the cranial site led to apnoea approximately 10 minutes post-injection, at which time the snakes were intubated and mechanically ventilated.

Conclusions and clinical relevance

Alfaxalone provided rapid, smooth induction when administered intramuscularly to pythons, and may serve as a useful induction agent prior to provision of volatile anaesthetics. The same dosage injected in the cranial site led to deeper anaesthesia than when injected caudally, suggesting that shunting to the liver and first-pass metabolism of alfaxalone occur when injected caudally, via the renal portal system.  相似文献

Europe’s woodland and savanna rangelands, often part of silvopastoral systems known as wood-pastures, are deteriorating because of abandonment that leads to return to a forested state or lack of tree regeneration from overgrazing or tree and shrub removal. Despite numerous local studies, there has been no broader survey of the stand structure of European wood-pastures showing which systems are at risk of losing their semiopen character. This overview aims to 1) show some of the differences and similarities in wood-pastures from landscapes across Europe and 2) identify which of these wood-pastures are at risk of losing their semiopen character. We collated a dataset of 13 693 trees from 390 plots in wood-pastures from eight different European regions (western Estonia, eastern Greece, northern Germany, Hungary, northern Italy, southern Portugal, central Romania, and southern Sweden), including tree diameters at breast height, tree density, management type, and tree species composition. On the basis of their structural characteristics, we classified wood-pastures using principal component analysis (PCA) and cluster analysis. The PCA showed a gradient from dense wood-pastures with high levels of regeneration (e.g., in Estonia) to sparse wood-pastures with large trees but a lack of regeneration (e.g., in Romania). Along this gradient, we identified three main groups of wood-pastures: 1) sparse wood-pastures with mostly big trees; 2) dense wood-pastures composed of small trees, and 3) wood-pastures containing a wide range of tree ages. Our results show a large structural gradient in European wood-pastures, as well as regeneration problems varying in their severity, highlighting the importance of social-ecological context for wood-pasture conditions. To maintain the ecological and cultural integrity of European wood-pastures, we suggest 1) more comprehensively considering them in European policies such as the Common Agricultural Policy and EU Habitats Directive, while 2) taking into account their structural characteristics and social-ecological backgrounds.  相似文献

Climate change is already impacting species around the world. Although most focus has been on the effect of temperature, changes in climatic variables other than temperature are also expected to drive biological change. Current models suggest that ectotherms, such as reptiles, will be strongly affected by climate change; however, data from natural populations are rare. Here, we use extensive data from 2 populations of a viviparous lizard (Niveoscincus ocellatus Gray, 1845) at the climatic extreme of the species distribution. We examine the effects of climate at a local, a regional and a global scale (thus, integrating a suite of variables at different spatial and temporal scales) on 2 key life history traits: offspring date of birth and size at birth. Overall, our results show that across 9 years of study, local temperature had strong effects on the offspring date of birth but not on the size at birth. Therefore, a rapid increase in local temperature throughout the species range (as predicted under global warming scenarios) is likely to affect phenological processes with potential concomitant effects on offspring fitness and survival.  相似文献

Purpose

The importance of the bioaccessible fractions of particle-bound contaminants is a key feature for the assessment of their likely risks to the aquatic environment. The present study investigated the extractability and potential toxicity of polycyclic aromatic hydrocarbons (PAHs) in river sediments. This study combined chemical with bioanalytical tools to establish a more in-depth insight into the relationship between the extractability of sediment contaminants, the chemodynamic properties of each extraction approach, and resulting toxicity.

Materials and methods

Sediment samples were treated with different extraction methods, namely Soxhlet extraction with acetone (SOX), membrane dialysis extraction (MDE) with n-hexane, ultrasonic extraction with acetone (USE), and extraction with (2-hydroxpropyl)-??-cyclodextrin (HBCD). The extracts were analyzed for PAHs using gas chromatography/mass spectrometry and tested using the neutral red retention assay and the ethoxyresorufin-O-deethylase (EROD) induction assay with the permanent fish cell line RTL-W1.

Results and discussion

The SOX and MDE approaches were comparable in their extraction power regarding PAHs and their cytotoxicity. However, the EROD activity differed between SOX and MDE, possibly due to retardation effects of the polyethylene membrane used in MDE. Thus, macromolecular components of the sediment were not extracted and therefore did not contribute to toxicity. HBCD extraction provided 3.4?% of the total PAH content in the sediments and might be an appropriate approach to predict the bioaccessible fraction. USE showed an extraction power between the HBCD approach and the MDE as well as SOX and MDE method. Hence, USE is neither appropriate for exhaustive extraction nor for biomimetic extraction.

Conclusions

MDE and SOX have been approved for the exhaustive extraction of PAHs from sediments for a worst case assessment of effect potentials. The influence of the low-density polyethylene membrane and the experimental conditions on the diffusion of compounds with broad physical?Cchemical properties, and the establishment of models such as quantitative-structure relationships for the prediction of diffusion behavior of a broad range of compounds require further assessment. HBCD was confirmed as a method providing a certain, putatively bioaccessible, fraction. Mechanistic research is required regarding desorption and uptake kinetics of particularly bound compounds with different physical?Cchemical properties in the solid phase?Cwater?CHBCD system to get more insight in the underlying partitioning processes.  相似文献

Elemental concentrations in leaf tissue can identify limiting conditions in crops and can be useful in managing and restoring marshes. Coastal management and restoration plans frequently include Spartina spp. because they are common plants in coastal wetlands across North America. Researchers frequently compare results of stoichiometric studies among Spartina spp. to corroborate results, although their stoichiometry may not be comparable. We compare the stoichiometry of paired samples of Spartina patens and Spartina alterniflora collected across Louisiana. Overall differences in stoichiometry between species, seasonal changes, and effects of porewater chemistry were quantified. Manganese (Mn) concentrations and calcium (Ca) concentrations were higher in S. alterniflora and the difference in [Ca] increased seasonally. Sodium concentrations were similar, except during prolonged inundation. Short flooding durations decreased carbon (C):nitrogen (N) without increasing [Mn] or flooding stress in both species.  相似文献

European agroecosystems host a variety of farm woodlands that act as primary determinants of biodiversity and ecosystem services. While woodland areas have been in decline worldwide, they have regionally increased, for example, in Eastern Germany. This study performs a quantitative and spatially explicit assessment of differences in species richness, diversity, and evenness as well as forest physiognomy and structure among Eastern German farm woodlands established during (1) the presocialist era (until 1945), (2) the socialist era (1945–1990), and (3) the postsocialist era (after 1990). Aerial imagery was used to allocate woodlands to one of the three eras, after which a forest inventory of 120 woodlands was performed. The results show substantial differences in forest composition and structure. Presocialist-era woodlands are composed of native (mean 96 %), deciduous (mean 94 %) tree species. Mean diameters and species richness values are high. Typical socialist woodland species are nonnative (mean 35 %) and/or coniferous (mean 51 %). Stands have a uniform, even-aged stand structure. Species richness/diversity indices are generally low. Postsocialist woodlands exhibit a high degree of variability. Percentages of nonnative (7 %) and coniferous (10 %) individuals are low. The findings suggest that socialist and postsocialist farmland and forest policies translated into distinct land-use legacies in the newly established farm woodlands, which differ considerably from the composition and structure of presocialist woodlands. We argue that forest conservation planning should actively consider land-use legacies, which are of particular relevance in the landscapes of Central and Eastern Europe, as these have undergone multiple, abrupt, and severe land-use transitions.  相似文献

Clinical outbreaks due to Actinobacillus pleuropneumoniae occur recurrently, despite the wide-scale use of antimicrobials or vaccination. Therefore, new approaches for the prevention and control of these outbreaks are necessary. For the development of alternative measures, more insight into the transmission of the bacterium on farms is necessary. The aim of this cohort study was to quantify transmission of A. pleuropneumoniae amongst weaned piglets on farms. We investigated three possible transmission routes: (i) indirect transmission by infected piglets within the same compartment, (ii) transmission by infected pigs in adjacent pens and (iii) transmission by direct contact within pens. Additionally, we evaluated the effect of independent litter characteristics on the probability of infection. Two farms participated in our study. Serum and tonsil brush samples were collected from sows pre-farrowing. Serum was analysed for antibodies against Apx toxins and Omp. Subsequently, tonsil brush samples were collected from all piglets from these dams (N = 542) in three cohorts, 3 days before weaning and 6 weeks later. Tonsil samples were analysed by qPCR for the presence of the apxIVA gene of A. pleuropneumoniae. Before weaning, 25% of the piglets tested positive; 6 weeks later 47% tested positive. Regression and stochastic transmission models were used to assess the contribution of each of the three transmission routes and to estimate transmission rates. Transmission between piglets in adjacent pens did not differ significantly from that between non-adjacent pens. The transmission rate across pens was estimated to be 0.0058 day−1 (95% CI: 0.0030–0.010), whereas the transmission rate within pens was ten times higher 0.059 day−1 (95% CI: 0.048–0.072). Subsequently, the effects of parity and serological response of the dam and litter age at weaning on the probability of infection of pigs were evaluated by including these into the regression model. A higher dam ApxII antibody level was associated with a lower probability of infection of the pig after weaning; age at weaning was associated with a higher probability of infection of the pig after weaning. Finally, transmission rate estimates were used in a scenario study in which the litters within a compartment were mixed across pens at weaning instead of raising litter mates together in a pen. The results showed that the proportion of infected piglets increased to 69% if litters were mixed at weaning, indicating that farm management measures may affect spread of A. pleuropneumoniae.  相似文献
