This study aimed to describe successful cryopreservation of sperm from maned wolves (Chrysocyon brachyurus). Three ejaculates from 2 maned wolves were collected by digital manipulation of the penis and evaluated subjectively, centrifuged and frozen in BotuCrio® (Botupharma, Botucatu, Brazil) or Tris–yolk egg extender. Spermatozoa were thawed at 37ºC/30s or 70ºC/4s and evaluated for kinetics, morphology, plasma and acrosome membrane integrity, mitochondrial potential, hydrogen peroxide, superoxide anion and lipid peroxidation. From 5 thawed samples, two had sperm total motility >55% (56.0% and 64.0%) and progressive motility ~35% (35% and 40%), both frozen with Tris–yolk egg. Plasma and acrosome membrane integrity decreased and percentage of sperm defects increased post-thawing. We concluded that is possible to freeze spermatozoa from maned wolves using semen collection and processing methods applied for domestic dogs.  相似文献

Grooming is a natural hygienic behaviour of cats that favours the formation of hairballs. Increased fibre concentration in the diets is a strategy to minimize hairball formation, but it is not fully effective. Because cat hair is formed mostly by keratin, the addition of keratinases in the diets might be an alternative for hairball control. Thus, the objective was to evaluate the effect of the combined use of sugarcane fibre and a protease complex to reduce the hairball excretion in cats. Twenty-four adult cats were divided into four treatment groups (n = 6 per treatment) in a 2 × 2 factorial arrangement. Treatments were as follows: control diet (Control; containing low concentration of insoluble fibre, 5.34% of total dietary fibre), control diet plus enzyme (Co-e), high-fibre diet (HF; containing high amounts of insoluble fibre, 17.8% of total dietary fibre), and HF diet plus enzyme (HF-e). Proteases from Bacillus licheniformis PWD-1 were administered orally (5 mg/day) as gastro-resistant capsules. Total collection of faeces was carried out to determine the number of excreted hairballs and the coefficient of total tract apparent digestibilities (CTTAD) of the diets. Separate addition of insoluble fibre (HF; p = .5947) or enzyme complex (Co-e; p = .3633) had no effect on the hairballs excreted in the faeces. However, the combined use of insoluble fibre and enzymes (HF-e) reduced (p = .0344) the total number of hairballs excreted. The size distribution of hairballs (small, medium, or large) was not affected by treatments (p = .3763). The CTTAD of crude protein was not affected by protease addition (p = .781) but was reduced by HF and HF-e treatments. Sugarcane fibre associated to keratinolytic enzymes reduces the hairball excretion in cats. The aim of this study was to generate models to predict the salinity of the soils in the geothermal zone which is located in Michoacán, México, over space and time. Systematic sampling was performed at locations 400 m apart with a sampling period every 48 days. At each site, two soil samples, from depths of 0–30 cm and 30–60 cm, were collected and analysed in laboratory. Regression-Kriging was used to make spatial predictions of the electrical conductivity of soil (ECs). Spherical and Gaussian theoretical models fit the experimental semivariograms. The results showed that the soil samples ranged from slightly saline (2.1–4 dS m?1) to strongly saline (8.1–16 dS m?1). In addition, the ECs was greater at a depth of 30–60 cm than it was at a depth of 0–30 cm. The ECs decreased from more than 8.0 dS m?1 to less than 4.0 dS m?1 after the first sampling period.  相似文献

Mediterranean mountain landscapes have high ecological and cultural values. However, since 1950, a large number of these landscapes have been subject to degradation as a consequence of land abandonment. Some regional administrations have enacted measures that favour the regeneration of pasturelands through scrubland clearing. The aim of this study was to evaluate the consequences of clearing and the regeneration of pasturelands on landscape structure and the recovery of extensive livestock in La Rioja. The results indicate that between 1986 and 2011 an area of 29,659 ha was cleared in the mountains of La Rioja, although 62·6% of this was cleared in the period 2000–2011. In 1956, the landscape was dominated by cultivated fields, but by 2001, much of the agricultural area had been abandoned and replaced by scrubland, natural woods and reforested areas. Furthermore, the landscape was more fragmented in 1956 than in 2001. The landscape structure varied following clearing, with a doubling in the number of patches and an increase in heterogeneity. The consequences of clearing have included an increase in the area of pastureland and the number of large animals. In addition, the number of farms has decreased, but their mean size has increased. Clearing is an appropriate way for spatial reorganization in marginal mountain areas. It does not require excessive investment but favours the reintegration of abandoned fields and an increase in pasturelands. The results of this study contribute to the debate about reclaiming mountain landscapes affected by human activities and subject to intense degradation processes. Drought and salinity are the main limiting environmental factors that restrict the establishment of soya bean plants. In order to recommend genotypes for cultivation under adverse drought and saline stress conditions, multi-environment trials (MET) are needed. However, MET analysis is usually performed considering a single trait, which provides lower reliability in recommending genotypes when compared to multi-trait analysis. Thus, this study was carried out to investigate the stability of multi-trait stability index (MTSI) in 46 soya bean cultivars under the effects of drought and saline stress on seed germination and initial seedling growth. Drought and saline stresses were imposed by seed exposure to −0.20 MPa iso-osmotic solutions with polyethylene glycol—PEG 6000 (119.6 g/L) or NaCl (2.36 g/L) for 12 days at 25°C. The germination rate, seedling length and seedling dry matter were measured. We showed here how genotypic stability can be quantified by MTSI when comparing drought and salinity conditions in relation to non-stressful environment (control) and how this index can be employed under different conditions. When considering the index for multiple environments, we can select as the most stable genotypes TMG 716 RR, FPS Antares RR, AS 3610 IPRO, NS 7300 IPRO and FPS Solar IPRO among the 46 tested genotypes. Owing to high stability and gains with selection verified for these genotypes under salinity and drought conditions, they can be used as genitors in breeding programs aimed at obtaining offspring with higher resistance to antibiotic stresses.  相似文献

Soil contamination may contribute to forest decline, by altering nutrient cycling and acquisition by plants. This may hamper the establishment of a woody plant cover in contaminated areas, thus limiting the success of a restoration program. We studied the nutritional status of planted saplings of Holm oak (Quercus ilex subsp. ballota (Desf.) Samp.), white poplar (Populus alba L.), and wild olive tree (Olea europaea var. sylvestris Brot.) in the Guadiamar Green Corridor (SW Spain) and compared it with established adult trees. Soils in this area were affected by a mine-spill in 1998 and a subsequent restoration program. The spill resulted in soil acidification, due to pyrite oxidation, and deposited high concentrations of some trace elements. In some sites, we detected a phosphorus deficiency in the leaves of Q. ilex and O. europaea saplings, as indicated by a high N:P ratio (>16). For O. europaea, soil contamination explained 40% of the variability in leaf P and was negatively related to chlorophyll content. Soil pH was a significant factor predicting the variability of several nutrients, including Mg, P, and S. The uptake of Mg and S by P. alba was greater in acidic soils. The monitoring of soil pH is recommended since long-term effects of soil acidification may negatively affect the nutritional status of the trees.  相似文献

Malathion is an insecticide widely used in agriculture and in public health programs that when used indiscriminately in large amounts can cause environmental pollution and risk to human health. However, it is possible that during the metabolism of malathion, reactive oxygen species can be generated, and malathion may produce oxidative stress in intoxicated rats that can be responsible for alterations in DNA molecules related in some studies. As a result, the present study aimed to investigate the DNA damage of cerebral tissue and peripheral blood in rats after acute and chronic malathion exposure. We used single cell gel electrophoresis (Comet assay) to measure early damage in hippocampus and peripheral blood and the Micronucleus test in total erythrocytes samples. Malathion was administered intraperitoneally once a day for one day (acute) or for 28 days (chronic) protocols (in both protocols, malathion was administered at 25, 50, 100, and 150 mg/kg). Our results showed that malathion (100 and 150 mg/kg) increased the DNA damage index in the peripheral blood and in the hippocampus after both chronic and acute treatment. Malathion increased the frequency of micronuclei only in chronic treatment at 150 mg/kg dose, and induced a cytotoxic dose-dependent decrease in the frequency of polychromatic erythrocytes in the peripheral blood of rats. In conclusion, since malathion increased both the peripheral blood and hippocampus DNA damage index using the Comet assay and increased the frequency of micronuclei in the total peripheral blood, it can be regarded as a potential mutagen/carcinogenic agent.  相似文献
