The fertility was compared between ejaculated and cauda epididymal sperm sensitized with prostatic fluid in dog after freeze-thawing using the fertility of ova from the contralateral ovary after injection (2 × 10(8) sperm) into dog uterus on the unilateral ovariectomized side, on the basis of the presence or absence of conception. No significant difference was observed in sperm quality after freeze-thawing between the two groups and conception rates were equivalent and low. Therefore, to achieve a high fertility by intrauterine insemination of canine frozen-thawed ejaculated and cauda epididymal sperm, intrauterine insemination on both sides is recommended, rather than insemination with a lot of sperm of the uterine horn on one side. 相似文献

Piglets of eight sows vaccinated by different routes with the attenuated TGE mutant coronavirus, Nouzilly (N) strain, and piglets from two field seropositive sows were challenged with a virulent TGE strain. On the day of challenge and 10 days after challenge, milk and serum samples from sows were analysed for their level of neutralizing antibodies, total immunoglobulin classes and TGE antibody classes by an ELISA. No direct relationship was seen between the level of protection of the litters and the titres of the different antibody classes on the day of challenge. The quantity of 140S particles in inactivated foot-and-mouth disease virus (FMDV) vaccine samples produced in Foot-and-Mouth Disease Vaccine Production Center (FMD Vaccine Production Center) in Thailand was estimated by the sucrose gradient ultracentrifugation and optical density analysis by using the computer applying system. The soft ware; Chromato Data System (CDS) (Nihon Chromato Works Co., Ltd. Japan) which is prepared for the analysis of chromatography, was applied for the estimation of 140S particles in FMDV vaccine. The quantity of 140S particles in each vaccine sample measured by CDS was mostly ranged from 2-4 micrograms/ml and this quantity was consistent with the results of the other reports. This method is considered to be the available method for estimation of 140S particles in FMDV vaccine as routine assay. 相似文献

Recently, we established the GEEP (“gene editing by electroporation of Cas9 protein”) method, in which the CRISPR/Cas9 system, consisting of a Cas9 protein and single guide RNA (sgRNA), is introduced into pig zygotes by electroporation and thus induces highly efficient targeted gene disruption. In this study, we examined the effects of sgRNA on the blastocyst formation of porcine embryos and evaluated their genome‐editing efficiency. To produce an animal model for diabetes, we targeted PDX‐1 (pancreas duodenum homeobox 1), a gene that is crucial for pancreas development during the fetal period and whose monoallelic disruption impairs insulin secretion. First, Cas9 protein with different sgRNAs that targeted distinct sites in the PDX‐1 exon 1 was introduced into in vitro‐fertilized zygotes by the GEEP method. Of the six sgRNAs tested, three sgRNAs (sgRNA1, 2, and 3) successfully modified PDX‐1 gene. The blastocyst formation rate of zygotes edited with sgRNA3 was significantly (p < 0.05) lower than that of control zygotes without the electroporation treatment. Our study indicates that the GEEP method can be successfully used to generate PDX‐1 mutant blastocysts, but the development and the efficiency of editing the genome of zygotes may be affected by the sgRNA used for CRISPR/Cas9 system. 相似文献

The objectives of this study were to estimate and evaluate potassium (K) and magnesium (Mg) budgets and flows of animal production in the basin of Dianchi Lake, China. Feed sampling and farmer interviews were conducted in field surveys. The supplies of K and Mg from local and external feeds and the retention, production and excretion of animals were calculated individually for dairy cows, fattening pigs, breeding sows, and broilers and laying hens. The K and Mg flows on a regional level were estimated using the individual budgets. At the individual level, in dairy cattle, the K and Mg supplied from local feeds accounted for large parts of the total nutrient intakes, whereas in the other animal categories most of the K and Mg in the feeds depended on external resources. Our findings also suggested that excessive Mg intake resulted in high Mg excretion and low use efficiency in dairy cattle and fattening pigs. At the regional level, the K and Mg amounts of manure produced and applied in the area (K: 339 and Mg: 143 t/year) exceeded those used as local feeds. Our results imply the animal production potentially increased the K and Mg loads in the regional agriculture system. 相似文献

Osmoregulatory actions of growth hormone (GH) and its mode of action in salmonids are reviewed. We present evidence suggesting
that insulin-like growth factor I (IGF-I) mediates some of the actions of GH on seawater acclimation. Plasma concentration
and turnover of GH rise following exposure to seawater. Exogenous GH (in vivo) increases gill Na+,K+-ATPase activity and the number of gill chloride cells, and inhibits an increase in plasma osmolarity and ions following transfer
of fish to seawater. A single class of high affinity GH receptors is present in the liver, gill, intestine, and kidney. The
levels of IGF-I mRNA in the liver, gill and kidney increased after GH-injection. After transfer to seawater, IGF-I mRNA increased
in the gill and kidney following the rise in plasma GH, although no significant change was seen in the liver. Injection of
IGF-I improved the ability of the fish to maintain plasma sodium levels after transfer to seawater. GH treatment also sensitizes
the interrenal to adrenocorticotropin (ACTH), increasing cortisol secretion. Both cortisol and IGF-I may be involved in mediating
the action of GH in seawater adaptation, although studies on the effect of GH on osmoregulatory physiology of non-salmonid
species are limited. An integrated model of the osmoregulatory actions of GH is presented, and areas in need of research are
outlined.
Résumé Cet article est une revue des effets osmorégulateurs de l'hormone de croissance et de son mode d'action. Nous présentons des résultats qui suggèrent que le facteur de croissance de type insuline (IGF-I) est un médiateur de certaines des actions de la GH sur l'adaptation à l'eau de mer. Les concentrations plasmatiques et le renouvellement de la GH augmentent après transfert en eau de mer. La GH exogène stimule (in vivo) l'activité Na+,K+-ATPase et le nombre de cellules à chlorure branchialeset inhibe les augmentations de l'osmolarité et des concentrations ioniques du plasma observées après transfert en eau de mer. Une seule classe de récepteurs à haute affinité pour la GH est présent dans le foie, les branchies, l'intestin et le rein. Les niveaux d'ARNm d'IGF dans le foie, les branchies et le rein augmentent après injection de la GH. Après transfert en eau de mer, les ARNm de l'IGF augmentent dans les branchies et dans le rein en suivant l'augmentation de GH plasmatique, bien qu'aucune modification ne soit observée au niveau du foie. L'injection d'IGF augmente la capacité du poisson à maintenir ses niveaux de sodium plasmatique après transfert en eau de mer. Le traitement à la GH augmente la sensibilité à l'adrenocorticotropine (ACTH) et stimule donc les niveaux de cortisol. A la fois le cortisol et l'IGF-I semblent impliqués comme médiateurs des effets de la GH dans l'adaptation à l'eau de mer, bien que les études sur les effets de la GH sur la physiologie de l'osmorégulation chez les espèces non-salmonidés restent encore limitées. This study was conducted to determine the correlation between reproductive hormones and musth in a male African elephant. Changes in circulating luteinizing hormone (LH), follicle stimulating hormone (FSH), testosterone and immunoreactive (ir-) inhibin and the degree of musth were evaluated for 4 years. LH increased 4 weeks before musth began. The highest concentrations of testosterone and ir-inhibin were observed from April to October. There were positive correlations among testosterone, ir-inhibin and musth behavior. These findings suggested that the surge-like LH in the pre-musth period might stimulate secretion of testosterone and ir-inhibin and thus initiate the musth behavior. This study also suggested that the high LH level before musth might be a useful biomarker for the beginning of the musth season. 相似文献

γ-Aminobutyric acid (GABA)-producing lactic acid bacteria (LAB) were isolated from four types of Myanmar traditional fermented
fishery products with boiled rice. All of them belonged to the genus Lactobacillus, and comparison of the effects of these representatives on GABA accumulation in fermented fishery products with boiled rice
revealed that Lactobacillus farciminis D323 is the most effective strain as a starter culture. These results may contribute to the development of traditional fermented
fishery products with functional properties. In addition, this study is the first to show in detail the distribution of GABA-producing
