We established 9 cell lines from 63 tumor cases of enzootic bovine leukosis and studied their properties. Cells of all lines formed small clumps and floated in culture medium, indicating growth. Four of the 9 cell lines were surface immunoglobulin (SIg)-positive, but the remaining 5 line cells were negative for SIg or, if SIg was detected, the percentage of SIg-positive cells was very low. Tests for the properties of the cells with monoclonal antibodies to lymphocytes revealed that the established line cells are B-lymphocytes. Morphological observation also revealed that they had the morphology of B-lymphoblastic cell. The results of E and EAC rosette assay were negative, but 6 of 8 cell lines were positive for EA rosetting. All the 9 cell lines reacted with MoAb C-143, which recognizes the tumor-associated antigen (TAA) of the EBL tumor cell. All 9 cell lines produced bovine leukosis virus (BLV). These results suggest that the 9 cell lines are tumor cells derived from B-lymphocytes of EBL.  相似文献

A survey of Theileria parasite infection in cattle in Cambodia and Vietnam was carried out by using allele-specific polymerase chain reaction. A total of 137 blood samples from draught animals in Cambodia and 40 blood samples from dairy cattle in Vietnam were analyzed. In Cambodia, 69 out of 137(50.4%) samples were PCR-positive containing mainly the Thai and the C type parasites. In Vietnam, 11 (27.5%) samples were positive and all were of the Thai type parasite.  相似文献

Chronic lung injury resulting from a variety of different causes is frequently associated with the develop ment of pulmonary fibrosis in humans. Although the etiology of pulmonary fibrosis is generally unknown, several sources of evidence support the hypothesis that a number of environmental and occupational agents play an etiologic role in the pathogenesis of this disease. The agents discussed in this review include beryllium, nylon flock, textile printing aerosols, polyvinyl chloride and didecyldimethylammonium chloride. The authors also describe a variety of animal models, including genetically modified mice, in order to investigate the molecular mechanism of pulmonary fibrosis, focusing on chemokine receptors, regulatory T cells and transforming growth factor-β and bone morphogenetic protein signaling. Overall, we propose the concept of toxicological pulmonary fibrosis as a lung disease induced in response to environmental cues.  相似文献

The Glycine max (L.) Merr. cultivar Waseshiroge is highly resistant to several races of Phytophthora sojae in Japan. In order to determine which Rps gene might be present in Waseshiroge, 15 differential cultivars were challenged with 12 P. sojae isolates. None had a reaction pattern identical to that of Waseshiroge, indicating that Waseshiroge may contain a novel Rps gene. In order to characterize the inheritance of Waseshiroge resistance to P. sojae isolates, 98 F2 progeny and 94 F7:8 lines were produced from crosses between the susceptible cultivar Tanbakuro and Waseshiroge. Chi-square tests indicated that segregation fit a 3:1 ratio for resistance and susceptibility in two F2 sub-populations of 42 and 56 seedlings. This and a 46.27:1.46:46.27 (or 63:2:63) ratio for resistance: segregation: susceptibility among the 94 F7:8 lines indicated that resistance was controlled by a single dominant gene. DNA analyses were carried out on Tanbakuro, Waseshiroge and the 94 F7:8 lines, and a linkage map was constructed with 17 SSR markers and nine new primer pairs that amplify marker loci linked to Rps1 on soybean chromosome 3 (linkage group N). The closest markers, Satt009 and T000304487l, map to locations 0.9 and 1.6 cM on each side of the estimated position of the Rps gene, respectively. The results showed that the Rps gene in Waseshiroge is either allelic to Rps1, or resides at a tightly linked locus in a gene cluster. A three-way-contingency table analysis indicated that marker-assisted selection with the two flanking markers could be used in the development of new resistant cultivars.  相似文献

The main purpose of this study was to establish whether sugi lumber with an extremely low Young’s modulus, which is plentifully produced in southern Japan, can be practically used as laminae for glued laminated timber (glulam) from the viewpoint of long-term performance under loading. Bending creep tests were carried out on sugi (Cryptomeria japonica D. Don) glulam with extremely low Young’s modulus laminae (3–4 kN/mm2) for the inner layers, as were tests on hybrid glulam with Douglas-fir (Pseudotsuga menziesii Franco) laminae (14–15 kN/mm2) for the outermost layer and sugi laminae (including those with a Young’s modulus of 3–4 kN/mm2) for the inner layers. The specimens were eight glulam beams with different compositions that were 105 mm wide, 210 mm deep, and 3980 mm long. The term of the creep test was 4 years. The results are summarized as follows. First, there were no significant differences between the Young’s modulus or bending creep of glulam L30 (laminae with Young’s modulus of 3–4 kN/mm2) and that of glulam L50 (laminae with Young’s modulus of 5–6 kN/mm2) for the inner layers. Second, for asymmetric compositions, the behavior of increases and decreases of relative creep due to atmospheric changes showed opposite behavior for glulam loaded from the side of lower Young’s modulus and from the side of higher Young’s modulus. Third, the required experimental term for the creep test to estimate an accurate long-term curve is 1 or 2 years (with data for the first 6 months excluded) when the power law is used for the estimation. Fourth, the values of relative creep in 50 years obtained from the experimental term were much lower than 2, which is the standard value set by Notification No. 1459 of the Ministry of Construction in Japan, and these values were not affected by the composition of the laminae.  相似文献

We studied the relationships between the shear-force value and physical and structural properties of the intramuscular connective tissue (IMCT) in six classes of porcine skeletal muscle to elucidate the contribution of IMCT to toughness of raw pork. The shear-force value of raw pork correlated significantly with that of the IMCT model prepared from each class of skeletal muscle ( P  < 0.05). The correlation suggested that the variable toughness of pork was caused by the mechanical strength of the endomysium and perimysium. The thickness of the secondary perimysium correlated significantly with the shear-force value of raw pork ( P  < 0.05) and with that of the IMCT model ( P  < 0.05). The shear-force value of raw pork correlated significantly with the total amount of collagen ( P  < 0.05) but not with the heat-solubility of collagen. We concluded therefore that the thickness of the secondary perimysium determines the mechanical strength of IMCT and contributes to toughness in raw pork.  相似文献

Daily matrix flow at 1-m depth in a volcanic ash soil was calculated during a period of one year using Darcy's law. Unsaturated hydraulic conductivity of undisturbed core samples could be expressed as a unique function of the soil water content. Hydraulic gradient obtained from soil water suction by a tensiometer installed at 90- and 110-cm depths, and hydraulic conductivity converted from the soil water content by time domain reflectometry (TDR) were monitored every 30 min throughout a year in a maize (Zea mays L.) Chinese cabbage (Brassica pekinensis Rupr.) field. The matrix flow obtained by this method was substituted for the water balance equation to estimate the bypass flow, and monthly and annual evapotranspiration. Annual rainfall in 1997 was 989 mm and evapotranspiration was estimated to be 730 mm. Net matrix flow at 1-m depth was 164 mm downward even though upward matrix flow occurred during half of the year. Downward flow determined by the water balance method exceeded the downward matrix flow during two heavy rain events in the year and the difference between the two flows was 63 mm, which was considered to correspond to a bypass flow. The bypass flow accounted for only 6.4% of the annual rainfall. Matrix flow was well monitored by the application of unsaturated Darcy's law in a field, and monthly evapotranspiration and bypass flow could be quantified by the introduction of the water balance equation.  相似文献

ABSTRACT:   For pearl production, pearl oyster seeds from foreign pearl oysters as well as hybrids between native and such foreign pearl oysters are produced in Japanese hatcheries. However, it is very difficult to identify these pearl oysters and hybrids based on morphological measurements. Thus, a molecular identification method for distinguishing Atlantic pearl oysters Pinctada imbricata from the Indian-Pacific pearl oyster group including P. martensii and P. fucata , was developed. The polymerase chain reaction (PCR) products of the partial intergenic spacer (IGS) of nuclear ribosomal RNA (rRNA) genes exhibited length polymorphism between P. imbricata (590 bp) and the other two species (427 bp). The restriction fragment length polymorphism analysis of the PCR products (PCR-RFLP) cleaved with Mse  I observed in the IGS of nuclear rRNA genes also gave different profiles between P. imbricata and the other two species. The difference in PCR-RFLP using Alu  I was also detected in the mitochondrial 16S rRNA gene regions between P. imbricata and the other two species. Thus, the method developed enables the distinction of P. imbricata from P. martensii and P. fucata .  相似文献

Erianthus, one of the genus in Saccharum complex, is important genetic resources for sugarcane improvement. The morphology and flowering phenology of 108 accessions belonging to Erianthus procerus and three types of E. arundinaceus collected from throughout Thailand were compared. PCA analysis based on 22 characteristics clearly supported the separation of Type II and Type III of E. arundinaceus from E. procerus and Type I of E. arundinaceus according to their morphological characteristics, particularly their bud size, and the development of root primordia, which greatly affected axis I of the PCA analysis. E. procerus and Type I showed overlapping in many of their characteristics including their flowering periods. Flower characteristics such as rachis joint length, which were used for previous taxonomic classifications, were not available for the classification of the Thai samples because of the wide variation and overlapping among them. Most of these phenotype similarities and differences are considered to have developed convergently as a result of niche adaptation. Type II and III inhabit riverbanks and streambeds where floods occur frequently, while E. procerus and Type I mainly grow in non-flooding areas such as mountainous grassland, the edge of forests, and beside fields. All Thai Erianthus show unique geographic distributions in Thailand. In particular, the biogeographic boundary between Type II and Type III appeared to be located at the Isthmus of Kra. Although some types showed morphological similarities, reproductive isolation among the four groups seemed to be maintained by differentiation in geographic distribution, habitat preference, and flowering timing.  相似文献
