Canine leishmaniasis caused by Leishmania infantum is enzootic in the North American foxhound population. Currently available chemotherapy for canine leishmaniasis is not completely effective and relapses are common in treated dogs. Pentamidine and related aromatic diamidines possess broad spectrum antiprotozoal activity. The in vitro antileishmanial activities of 35 aromatic cationic molecules were determined, using pentamidine as the reference drug. The compounds were examined for activity against promastigotes of L. infantum isolated from a foxhound from Virginia. The compounds most active against Leishmania parasites were reversed amidines. Compound 9, a reversed amidine, exhibited the highest activity against L. infantum, with a 50% inhibitory concentration (IC(50)) of 0.0042 microM compared with 14.2 microM for pentamidine. Antileishmanial activities of nine compounds were at least 1000-fold higher relative to the reference drug. Results from this study indicate that several pentamidine-related compounds warrant further investigation as possible new agents for the treatment of canine leishmaniasis.  相似文献

Chitinous material was extracted from mycelia of Aspergillus niger and Mucor rouxii grown in yeast peptone dextrose broth for 15 and 21 days, respectively. The extracted material was characterized for purity, degree of acetylation, and crystallinity and tested for antibacterial and eliciting properties. The maximum glucosamine level determined in the mycelium of A. niger was 11.10% dw and in the mycelium of M. rouxii was 20.13% dw. On the basis of the stepwise extraction of freeze-dried mycelia, it appeared that M. rouxii mycelia contained both chitin and chitosan, whereas A. niger contained only chitin. The yields of crude chitin from A. niger and M. rouxii were 24.01 and 13.25%, respectively, and the yield of chitosan from M. rouxii was 12.49%. Significant amounts (7.42-39.81%) of glucan were associated with chitinous compounds from both species and could not be eliminated by the extraction method used. The degrees of acetylation were determined to be 76.53 and 50.07% for chitin from A. niger and M. rouxii, respectively, and 19.5% for M. rouxii chitosan. The crystallinity of fungal chitin and chitosan was estimated to be less intense than in corresponding materials from shrimp shells. The extracted chitin and chitosan in a concentration of 0.1% reduced Salmonella Typhimurium DT104 2576 counts by 0.5-1.5 logs during a 4 day incubation in tryptic soy broth at 25 degrees C. Furthermore, all tested chitinous materials from fungal sources significantly reduced lesions caused by Botrytis cinerea and Penicillium expansum in harvested apples.  相似文献

Accumulation of chitinous material in Agaricus bisporus stalks was determined during postharvest storage at 4 and 25 degrees C. The chitinous material was extracted after alkali treatment and acid reflux of alkali insoluble material and analyzed for yield, purity, degree of acetylation (DA), and crystallinity. The total glucosamine content in mushroom stalks increased from 7.14% dry weight (DW) at harvest (day 0) to 11.00% DW and 19.02% DW after 15 days of storage at 4 degrees C and 5 days of storage at 25 degrees C, respectively. The yield of crude chitin isolated from stalks stored at 25 degrees C for 5 days was 27.00% DW and consisted of 46.08% glucosamine and 20.94% neutral polysaccharides. The DA of fungal chitin was from 75.8 to 87.6%, which is similar to commercially available crustacean chitin. The yield of crude fungal chitin of 0.65-1.15% on a fresh basis indicates the potential for the utilization of these mushroom byproducts.  相似文献

The influence of the Gumigel bacterial-humus preparation on the biological activity of soils was investigated. The activity was assessed by the respiration intensity of the soil microorganisms, the total number of bacteria, and the structure of the saprotrophic bacterial complex. It was demonstrated that the microorganisms were preserved in the preparation for a long time. As this preparation was kept at 4°C, the number of microorganisms was preserved at a permanent level during no less than half a year. The preparation did not have a significant effect on the biological activity of the unpolluted soil, but it intensified the biological activity in the case of the polluted soil, which was seen from an increase in the number of microorganisms and their respiration intensity.  相似文献

We examined fermentation capacity of fecal microbial inocula of Przewalski horse (Equus ferus przewalskii), Asian wild ass - kulan (Equus hemionus hemionus), and Chapman zebra (Equus quagga chapmani) in vitro. Interactions of the substrates (amorphous cellulose, wheat straw, meadow hay, xylan from oat spelt, and ground barley grain) and type of fecal inocula in the gas volume and in vitro dry matter digestibility were detected in all substrates after 72 hours of fermentation in five replicates for each substrate and type of inocula. No effects of fecal inocula sources were detected on total short-chain fatty acids concentrations. No live fecal ciliate population was present in kulan feces. Complex ciliate populations in zebra feces and the number and genera resembled ciliates from the colon of horses. Fresh feces of kulan and zebra were fractionated by galvanotaxis and centrifugation to separate fecal ciliates and bacteria. Specific activities (μmol of reducing equivalents/mL min mg protein) of carboxymethyl cellulase (CM-cellulase), xylanase, α-amylase, and inulinase were measured in crude cell-free extract of fecal ciliates (zebra), fecal bacteria (zebra and kulan), and total fecal preparation (zebra and kulan). All examined specific enzymatic activities were present in zebra fecal samples. We were unable to measure the inulinase activity and CM-cellulase activity in kulan fecal samples. Zebra ciliates are actively involved in the digestion of plant storage (α-amylase, 0.53 ± 0.02; inulinase, 1.77 ± 0.01, specific activities) and structural polysaccharides (CM-cellulase, 0.4 ± 0.15; xylanase, 0.26 ± 0.06). Accumulating data clearly indicate that the induction of apoptosis by nontoxic natural compounds is a potent defense against the development and progression of many malignancies, including colon cancer. Resveratrol and the fucoidans have been shown to possess potent anti-tumor activity in vitro and in vivo. The aim of the present study was to examine whether the combination of a fucoidan from the brown alga Saccharina cichorioides Miyabe and resveratrol would be an effective preventive and/or therapeutic strategy against colon cancer. Based on NMR spectroscopy and MALDI-TOF analysis, the fucoidan isolated and purified from Saccharina cichorioides Miyabe was (1→3)-α-L-fucan with sulfate groups at C2 and C4 of the α-L-fucopyranose residues. The fucoidan enhanced the antiproliferative activity of resveratrol at nontoxic doses and facilitated resveratrol-induced apoptosis in the HCT 116 human colon cancer cell line. Apoptosis was realized by the activation of initiator caspase-9 and effector caspase-7 and -3, followed by the cleavage of PARP. Furthermore, significant inhibition of HCT 116 colony formation was associated with the sensitization of cells to resveratrol by the fucoidan. Taken together, these results demonstrate that the combination of the algal fucoidan with resveratrol may provide a potential therapy against human colon cancer.  相似文献

Three different fucoidan fractions were isolated and purified from the brown alga, Sargassum mcclurei. The SmF1 and SmF2 fucoidans are sulfated heteropolysaccharides that contain fucose, galactose, mannose, xylose and glucose. The SmF3 fucoidan is highly sulfated (35%) galactofucan, and the main chain of the polysaccharide contains a →3)-α-l-Fucp(2,4SO3)-(1→3)-α-l-Fucp(2,4SO3)-(1→ motif with 1,4-linked 3-sulfated α-l-Fucp inserts and 6-linked galactose on reducing end. Possible branching points include the 1,2,6- or 1,3,6-linked galactose and/or 1,3,4-linked fucose residues that could be glycosylated with terminal β-d-Galp residues or chains of alternating sulfated 1,3-linked α-l-Fucp and 1,4-linked β-d-Galp residues, which have been identified in galactofucans for the first time. Both α-l-Fucp and β-d-Galp residues are sulfated at C-2 and/or C-4 (and some C-6 of β-d-Galp) and potentially the C-3 of terminal β-d-Galp, 1,4-linked β-d-Galp and 1,4-linked α-l-Fucp residues. All fucoidans fractions were less cytotoxic and displayed colony formation inhibition in colon cancer DLD-1 cells. Therefore, these fucoidan fractions are potential antitumor agents.  相似文献

Neoskrjabinolepis (Neoskrjabinolepidoides) merkushevae sp. n. is described from the shrew Sorex minutus L. (type host) from Belarus. The new species is characterised by rostellar hooks 35-37 microm long, provided with small epiphyseal thickening of the handle; short (35-40 microm) cirrus consisting of a basal part with claw-shaped spines, a parabasal part with thin needle-shaped spines and an unarmed distal part; cirrus-sac slightly passing into median field; and 12-16 eggs per gravid uterus. In addition to the type host and the type locality, the host range of the new species includes Sorex araneus L. and there are geographical records from Bulgaria, Lithuania and North-East Altay (Russia).  相似文献

The gut microbiota is composed of a diverse population of obligate and facultative anaerobic microorganisms which are shown to influence host metabolism and immune homeostasis. This study investigated the effects of virgin coconut oil on the weekly fasting glycaemia, daily food and water intake and weekly body mass gain over 16 weeks, as well as the changes in composition of gut microbiota in both non-diabetic and alloxan-induced diabetic rats. Although the intake of virgin coconut oil did not decrease the diabetes-induced hyperglycemia, it affected the secondary parameters, such as food and water intake and average body mass gain. Furthermore, its potential to positively affect the fecal microbiome was proved, since it significantly increased the abundance of probiotic bacteria, such as Lactobacillus, Allobaculum and Bifidobacterium species.  相似文献
