A linear programming model was developed to determine management policy for a yearly planning horizon on a typical 1320 acre (535 ha) Southern Colorado mountain ranch. Income producing activities (selling cattle and hay and leasing land) and cost activities (buying cattle, borrowing capital, feeding supplements, purchasing fertiliser and fuel, hiring labour and leasing land) were defined for three land types through four seasons.The purpose of the modelling effort was to determine the level of each activity (cow herd size, yearling herd size and area of meadow to be harvested for hay) that resulted in maximum net return for the ranch subject to resource limitation on land, labour and capital. In addition, sensitivity and shadow price analyses revealed expansion options that would be profitable during existing economic conditions.The optimal management plan, using 3 April, 1975 prices, called for grazing 199 head of 450 lb (204 kg) steers and 52 head of 530 lb (240 kg) steers. The plan also called for purchasing and spreading 42 tons (38 MT) of nitrogen and 10 tons (9 MT) of phosphorus on Land I and for growing, harvesting and selling 742 tons (674 MT) of hay. This optimal management plan produced a net return of $27,642. In addition, the optimal plan showed that hay production began replacing beef production when the price of hay was $35·48/ton (39·08/MT) and that the maximum amount of hay was produced with the price increased to $50·00/ton ($55·00/MT).  相似文献

Factors influencing C2H4 production in a silt loam were investigated in an effort to determine the source of this gas in soil. Air-dried samples of soil in glass vials were moistened to about ?10 kPa, sealed with rubber septa, and incubated at 30 or 35°C with an original atmosphere of air or O2-free N2. C2H4 concentrations in the vials were determined by gas chromatography.Addition of the antibacterial agents chloramphenicol or novobiocin to the soil inhibited C2H4 production, whereas the antifungal agent cycloheximide had no effect. Sodium azide and sodium cyanide also reduced C2H4 production. Treatment of the soil with moist heat (i.e. passing a steam-air mixture through it) at 80°C for 30 min failed to reduce the ability of the soil to produce C2H4 during subsequent incubation at 30°C, but autoclaving it twice at 121°C prevented C2H4 production. As with nonheated soil, C2H4 production from soil treated at 80°C was prevented by novobiocin but not by cycloheximide. Only about 10% of the bacteria isolated from nontreated soil were spore-formers. In contrast, 95–98% and possibly more of the bacteria isolated from heat-treated soil were spore-formers, including those in soil which was heat-treated and then incubated moist at 30°C for an additional 3 days before dilution plating. Addition of methionine had no effect on the production of C2H4 in anaerobic soil, whereas ethionine, chlorogenic acid, and ethylenediaminetetraacetic acid (EDTA) all enhanced C2H4 production. Ethionine, but not chlorogenic acid or EDTA, also resulted in considerable C2H4 accumulation in autoclaved soil; the C2H4 detected in ethionine-amended soil was apparently nonmicrobial in origin. Soil samples incubated at constant temperatures of 30, 50, or 70°C all produced C2H4.The results collectively indicate that C2H4 in soil is most likely produced by facultative or strictly anaerobic bacteria which are probably spore-formers and may also be thermophilic. Several isolates of spore-forming bacteria were inoculated into autoclaved soil, but none produced appreciable amounts of C2H4 under the test conditions.  相似文献

Footrot is a mixed bacterial infection of the hooves of sheep. The gram-negative anaerobic bacterium Dichelobacter nodosus is the principal causative agent, with different strains causing diseases of different severity, ranging from benign to virulent. In Australia, in the state of New South Wales (NSW), only virulent footrot is subject to regulatory action, including quarantine. However, it is often difficult to distinguish benign footrot from virulent footrot in the initial stages of infection, or under adverse climatic conditions. The gelatin gel test, which measures the thermostability of secreted bacterial proteases, is the laboratory test most widely used in Australia to aid in the differential diagnosis of footrot. The proteases of virulent strains are, in general, more thermostable than the proteases of benign strains. However, there are some false positives in the gelatin gel test, which may lead to unnecessary quarantine procedures. We used Southern blot analysis on 595 isolates of D. nodosus from 124 farms on which sheep had benign or virulent footrot to test for the presence of the intA gene. We found that for D. nodosus strains which are stable in the gelatin gel test, there is a high correlation between the presence of the intA gene and the ability of the strain to cause virulent footrot. We also developed a PCR-based assay for the rapid detection of intA, which can be used to test DNA extracted from colonies grown on plates, or DNA extracted from cotton swabs of culture plates.  相似文献

Reference ranges for five parameters of blood coagulation were established in clinically normal farmed fallow deer (Dama dama) and red deer (Cervus elaphus). Storage of plasma at -15 degrees C resulted in small increases in activated partial thromboplastin time (APTT) for both species, and in the one stage prothrombin time (OSPT) of fallow deer, between days 1 and 30. These times were then stable between days 30 and 60. The fibrinogen levels of fallow deer plasma showed a small apparent increase over 60 days. These storage artefacts were not of sufficient magnitude to preclude the use of such plasma for diagnostic purposes although they could limit its use in research. Levels of fibrin/fibrinogen degradation products (FDP) were not affected by storage at -15 degrees C. Rabbit brain thromboplastin appeared adequate for OSPT determination in both species. The activated clotting time (ACT) is recommended as a field test for screening the intrinsic and common pathways of blood coagulation in deer.  相似文献

Surveys were made at the end of the 1990 and 1991 growing seasons for root-inhabiting fungi in the genera Fusarium, Cylindrocarpon and Pythium from the roots of one year-old container-grown Douglas-fir and spruce seedlings grown under greenhouse conditions. In the 1990 survey of four nurseries, it was found that 61–97% of both Douglas-fir and spruce roots were colonized with Fusarium, Cylindrocarpon or Pythium. There were significantly (p0.05) more Douglas-fir roots than spruce roots colonized by Fusarium at all nurseries, however, there were significantly (p0.05) more spruce roots than Douglas-fir roots colonized by Cylindrocarpon and Pythium. Root colonization of Douglas-fir and spruce by the three fungal genera during 1991 varied from 0–82% at three nurseries, however, only at a south coastal nursery was there significantly (p0.05) more spruce than Douglas-fir roots colonized by Cylindrocarpon. Significantly more seedlings were infected in 1990 than in 1991. In 1991, there were few significant differences between Douglas-fir and spruce, in the percentage of seedlings with colonized roots and in the percentage of growth medium colonized by the fungi. An acute stress response can be provoked by abrupt social, nutritional, and environmental changes associated with weaning, and this may disrupt homeostasis and thus compromise well-being. Manipulating environmental factors, such as photoperiod, might provide a simple way to reduce the physiological consequences that piglets experience due to weaning stress. The objective of this study was to evaluate the impacts of photoperiod manipulation across various weaning ages on leukocyte populations, lymphocyte proliferation, natural killer cytotoxicity (NK), chemotaxis, phagocytosis, and immunoglobulin G, cortisol, and BW of piglets during the nursery phase. Sixty-eight crossbred piglets were obtained from sows kept on a short-day (8 h of light/d) photoperiod from d 90 of gestation until weaning. Piglets were weaned at 14, 21, or 28 d of age and kept on a short or long (16 h of light/d) photoperiod until 10 wk of age. Piglet BW and blood samples were collected at weaning and at 6, 8, and 10 wk of age. Pigs weaned at 28 d had reduced neutrophil counts (P < 0.001), phagocytosis (P < 0.001), and lymphocyte proliferation (P < 0.05) at weaning compared with those weaned at 14 and 21 d. Pigs weaned at 21 d tended to have lower (P = 0.08) lymphocyte counts than did pigs weaned at 14 or 28 d. Pigs weaned at 14 d had reduced (P < 0.01) NK relative to those weaned at 21 or 28 d. Photoperiod also influenced pig BW and immune status. Generally, those pigs on the long-day photoperiod and weaned at 28 d were heavier (P < 0.001) than their counterparts weaned at 14 or 21 d. At 6 wk of age, NK was greater (P = 0.002) in pigs kept on a long day and weaned at 14 or 21 d than in pigs weaned at 28 d. Phagocytosis was less (P = 0.005) at 6 wk of age, but was greater at 8 wk, in piglets kept on the long day and weaned at 28 d than in long-day pigs weaned at 14 or 21 d. These results suggest that photoperiod differentially influences immune responses in piglets weaned at different ages and indicate an inverse relationship between growth and immune status. Here, weaning at 28 d and a long-day photoperiod was the treatment combination that was most physiologically beneficial to piglets, whereas a 14-d weaning and short-day photoperiod was least physiologically beneficial.  相似文献

Accurate quantification with real-time PCR requires the use of stable endogenous controls. Recently, there has been much debate concerning the stability of commonly used reference or housekeeping genes. To address this concern, a number of statistical approaches have been designed to analyse data and assist in determining the most appropriate reference genes for experimental comparisons. In this study, three programs, BestKeeper, Norm Finder, and geNorm were used to assess four candidate reference genes: 18S rRNA, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), acidic ribosomal protein large (RPLP0) and beta-actin, for use in expression profiling of individuals from divergent cattle genotypes subject to parasitic challenge with the cattle tick Boophilus microplus. Results demonstrated beta-actin and GAPDH were the most suitable reference genes in blood and could be used either individually or combined as an index to normalise data. RPLP0 was identified as the least stable gene, while 18S rRNA was omitted as being too highly expressed. As the recommendations on the most suitable reference genes varied between the programs, it is recommended that more than one should be utilised, to ensure the most robust experimental tools are selected.  相似文献

Summary About 1000 Brassica oleracea accessions were evaluated in glasshouse tests for response to Plasmodiophora brassicae (clubroot). Resistance was confirmed in some north and west European kales and cabbage. A new source of resistance in cabbage, from Eire, is reported. Most other accessions were highly susceptible but lower levels of susceptibility were observed in open pollinated Brussels sprouts and forms of south European cabbage, cauliflower and broccoli. Modern breeding (as in the production of hybrid cultivars) appears to have resulted in increased susceptibility in several crop types. The implications of these results for the exploitation of germplasm are discussed.  相似文献
