Few pine species develop a seedling grass stage; this growth phase, characterized by strong, carrot-like taproots and a stem-less nature, poses unique challenges during nursery production. Fertilization levels beyond optimum could result in excessive diameter growth that reduces seedling quality as measured by the root bound index (RBI). We grew longleaf pine (Pinus palustris), a grass stage species, in containers of four different volumes (60–336 ml) either coated with copper oxychloride or left untreated and fertilized at low, medium, or high levels of nitrogen (N). In general, N concentration of tissues rose as N rate increased, with larger changes in concentration occurring between low and medium levels than between medium and high levels. N rate influenced root tissue N concentration less than it did stems and needles. Subtle needle color differences caused by N rate were significant, suggesting its potential utility during nursery production. As expected, seedlings grew larger as container volume increased and as N rate increased. Copper treatment, which we posited could influence the RBI, tended to increase root-collar diameter and tap root biomass and decrease total root volume. Chlorophyll abundance was affected more by N rate than by container volume or copper treatment, but photosynthesis was affected more by copper treatment than N rate or container size. Although RBI was 25 % greater for seedlings grown in small containers with high N rates than those grown in large containers with low N rates, RBI ranged only from 11 to 15 %, well below the critical 27 % threshold.  相似文献

Cellulose fibers coated with an N-halamine precursor have been reported. Cellulose fibers were dipped and padded with m-aramid dissolved in N,N-dimethylacetamide followed by coagulation in distilled water. The morphology of the maramid-coated cellulose fibers was analyzed using scanning electron microscopy. The m-aramid on cellulose fibers was characterized using X-ray photoelectron spectroscopy (XPS) and the chlorination of m-aramid on the cellulose fibers was determined using Fourier transform infrared spectroscopy and XPS. The liquid and gas permeability of m-aramid coated fabrics were conducted. Wash fastness was performed to measure the durability of the m-aramid on the cellulose fibers. The chlorinated m-aramid-coated cellulose fibers preformed a 7-log reduction against Escherichia coli and Staphylococcus aureus. The chlorine lost after the bacterial inactivation could be regained after a simple re-chlorination process.  相似文献

In this study, biodegradable poly(D,L-lactic-co-glycolic acid) (PLGA) fibers were prepared by a melt-electrospinning and treated with plasma in the presence of either oxygen or ammonia gas to modify the surface of the fibers. The effects of processing parameters on the melt-electrospinning of PLGA were examined in terms of fiber morphology and diameter. Among the processing parameters, the spinning temperature and mass flow rate had a significant effect on the average fiber diameter and its distribution. The water contact angle of melt-electrospun PLGA fibers decreased significantly from 123 ° to 55 ° (oxygen plasma treatment) or to 0 ° (ammonia plasma treatment) by plasma treatment for 180 sec, while their water content increased significantly from 2.4 % to 123 % (oxygen plasma treatment) or to 189 % (ammonia plasma treatment). Ammonia gas-plasma enhanced the surface hydrophilicity of PLGA fibers more effectively compared to oxygen gas-plasma. X-ray photoelectron spectroscopy analysis supported that the number of polar groups, such as hydroxyl and amino groups, on the surface of PLGA fibers increased after plasma treatment. Overall, the microfibrous PLGA scaffolds with appropriate surface hydrophilicity and fiber diameter could be fabricated by melt electrospinning and subsequent plasma treatment, without a significant deterioration of fiber structure and dimensional stability. This approach of controlling the surface properties and structures of fibers could be useful in the design and tailoring of novel scaffolds for tissue engineering.  相似文献

The present study reports the preparation of a cellulose scaffold for tissue engineering directly from cellulose fiber using ionic liquid (IL) by the NaCl leaching method with bovine serum albumin (BSA), which is well known protein utilized for biomedical applications like degradation of polymer, cell attachment and proliferation on scaffold. The 1-n-allyl-3-methylimidazolium chloride (AmimCl) IL was used as a solvent for cellulose. The morphology of the scaffold was studied by scanning electron microscopy (SEM) and the images showed that the pore sizes of the scaffolds were about 200 µm. In addition, the water uptake (WU) and degree of degradation of the cellulose scaffold were measured. Meanwhile, the biocompatibility and bioactivity of the scaffold were determined via 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenytetrazolium bromide (MTT) assay and the Live/Dead viability test. The various results demonstrated the ability of the Mesenchymal stem cells (MSC) to attach to the surface of the scaffolds amplified as percentage of BSA increased in cellulose scaffold.  相似文献

The aim of this study is to assess the effect of CO2 gas stunning which has not been conducted until now in comparison with captive bolt stunning on the meat quality of cattle. A total of 40 steers were slaughtered following two stunning processes: CO2 gas stunning (n=20), exposure at 70% CO2 gas atmosphere for 140 sec; and captive bolt stunning (CBS, n=20). The slaughtered steers were classified into Group A (620–710 kg) and Group B (720–790 kg) by their live weight. CO2 gas stunning decreases pH value (p<0.05) but increases lightness (p<0.001) and sarcomere length (p<0.001 and p<0.01 for Group A and Group B, respectively) in all live weight groups. Drip loss was increased with CO2 gas stunning in Group A (620–710 kg of live weight) (p<0.05), whereas WBSF was decreased with CO2 gas stunning in Group B (720–790 kg of live weight) (p<0.001). Therefore, CO2 gas stunning negatively affects muscle pH and water-holding capacity in steers but CO2 gas stunning can improve the tenderness and lightness compared with captive bolt stunning in cattle.  相似文献

Fruit size and seed moisture content were measured and seed production and dispersal were monitored to understand the seed biology of the Korean ash (Fraxinus rhynchophylla Hance) in two Korean ash forests. A combination of four ground treatments and four crown closure levels were created in a natural forest to understand the site conditions needed for seedling emergence in natural stands and to determine practical regeneration methods for direct seeding. The seed size of Korean ash increased until late May and its moisture content decreased rapidly in early November. Prolific seed bearing occurred every 3 yr. The distance of seed dispersal by wind was about 30 m, but more than 90% of the seeds dispersed within 10 m from the seed trees. Twenty-five seed trees per hectare provided for successful natural regeneration. The seedling emergence in the natural stands was best with scarification treatment and 25~50% of crown closure.  相似文献

In this study, we investigated diabetic stage dependent cyclooxygenase-2 (COX-2) immunoreactivity in the dentate gyrus in streptozotocin (STZ)-induced type 1 diabetic rats. The animals were sacrificed at 2, 3 and 4 weeks after STZ treatment. Blood glucose levels were increased after STZ treatment. COX-2 immunoreactivity in dentate gyrus was significantly increased in these regions 3 weeks after STZ treatment and restored to its basal level to 4 weeks after STZ treatment. In contrast, COX-2 immunoreactivity was not changed in CA3 region in all groups. These results suggest that STZ-induced type 1 diabetes transiently, but not permanently, decreased synaptic transmission and plasticity 3 weeks after STZ treatment in the dentate gyrus.  相似文献

Several types of cells, including blastoderm cells, primordial germ cells, and embryonic germ cells were injected into early-stage recipient embryos to produce chimera avians and to gain insights into cell development. However, a limited number of studies of avian adult stem cells have also been conducted. This study is, to the best of our knowledge, the first to evaluate chicken bone marrow cells' (chBMC) ability to differentiate into multiple cell lineages and capability to generate chimera chicks. We induced random differentiation of chBMCs in vitro and injected immunologically selected pluripotent cells in chBMCs into the blastoderms of recipient eggs. The multipotency of BMCs from the barred Plymouth rock (BPR) was confirmed via AP staining, RT-PCR, immunocytochemistry, and FACS using specific markers, such as Oct-4 and SSEA-1, 3 and 4. Isolated chBMCs were found to be able to induce in vitro differentiation to multiple cell lineages. Approximately 5,000 chBMCs were injected into the blastoderms of white leghorn (WL) recipients and proved able to contribute to the generation of somatic chimera chicks with a frequency of 2.7% (2 of 73). Confirmation of chimerism in hatched chicks was achieved via PCR analysis using D-loop-specific primers of BPR and WL. Our study demonstrated the successful production of chimera chicks using chBMC. Therefore, we propose that the use of adult chBMCs may constitute a new possible approach to the production of chimera poultry, and may provide helpful studies in avian developmental biology.  相似文献
