The median infectious dose (ID(50)) of porcine reproductive and respiratory syndrome (PRRS) virus isolate MN-184 was determined for aerosol exposure. In 7 replicates, 3-week-old pigs (n=58) respired 10l of airborne PRRS virus from a dynamic aerosol toroid (DAT) maintained at -4°C. Thereafter, pigs were housed in isolation and monitored for evidence of infection. Infection occurred at virus concentrations too low to quantify by microinfectivity assays. Therefore, exposure dose was determined using two indirect methods ("calculated" and "theoretical"). "Calculated" virus dose was derived from the concentration of rhodamine B monitored over the exposure sequence. "Theoretical" virus dose was based on the continuous stirred-tank reactor model. The ID(50) estimate was modeled on the proportion of pigs that became infected using the probit and logit link functions for both "calculated" and "theoretical" exposure doses. Based on "calculated" doses, the probit and logit ID(50) estimates were 1 × 10(-0.13)TCID(50) and 1 × 10(-0.14)TCID(50), respectively. Based on "theoretical" doses, the probit and logit ID(50) were 1 × 10(0.26)TCID(50) and 1 × 10(0.24)TCID(50), respectively. For each point estimate, the 95% confidence interval included the other three point estimates. The results indicated that MN-184 was far more infectious than PRRS virus isolate VR-2332, the only other PRRS virus isolate for which ID(50) has been estimated for airborne exposure. Since aerosol ID(50) estimates are available for only these two isolates, it is uncertain whether one or both of these isolates represent the normal range of PRRS virus infectivity by this route. 相似文献

This study was designed to determine the ability of tibial plateau leveling osteotomy (TPLO) to eliminate cranial tibial translation (CTT) through a loaded range of motion. Twenty-four large-breed canine cadaver limbs were compared. Each limb was placed in a custom designed jig at 120° of stifle extension under an axial load of 20% body weight. A force of approximately 10 N/s mimiced the action of the quadriceps muscle and allowed the limb to move from 120° to maximal extension. Positional data were acquired using electromagnetic motion-tracking sensors. Each limb was tested under normal, cranial cruciate ligament (CrCL)-deficient, and TPLO-treated conditions. Cranial tibial translation significantly increased after transection of the CrCL. The TPLO failed to normalize CTT within the CrCL deficient stifle; however, values trended towards intact values throughout the range of motion. The TPLO was more effective at higher angles of flexion. These altered biomechanics may help explain the continued progression of osteoarthritis in TPLO repaired stifles. Understanding how comets work--what drives their activity--is crucial to the use of comets in studying the early solar system. EPOXI (Extrasolar Planet Observation and Deep Impact Extended Investigation) flew past comet 103P/Hartley 2, one with an unusually small but very active nucleus, taking both images and spectra. Unlike large, relatively inactive nuclei, this nucleus is outgassing primarily because of CO(2), which drags chunks of ice out of the nucleus. To understand fully the risk of biological invasions, it is necessary to quantify propagule pressure along all introduction pathways. In the Antarctic region, importation of fresh produce is a potentially high risk, but as yet unquantified pathway. To address this knowledge gap, >11,250 fruit and vegetables sent to nine research stations in Antarctica and the sub-Antarctic islands, were examined for associated soil, invertebrates and microbial decomposition. Fifty-one food types were sourced from c. 130 locations dispersed across all six of the Earth’s inhabited continents. On average, 12% of food items had soil on their surface, 28% showed microbial infection resulting in rot and more than 56 invertebrates were recorded, mainly from leafy produce. Approximately 30% of identified fungi sampled from infected foods were not recorded previously from within the Antarctic region, although this may reflect limited knowledge of Antarctic fungal diversity. The number of non-native flying invertebrates caught within the Rothera Research Station food storage area was linked closely with the level of fresh food resupply by ship and aircraft. The ion-mediated enhancement of xylem hydraulic conductivity in angiosperms is thought to be controlled by the pectin chemistry of intervessel pit membranes. However, there is little or no direct evidence on the ultrastructure and chemical nature of pit membranes in species that show an 'ionic effect'. The potential link between the magnitude of the ionic effect and pectin composition in intervessel pit membranes of four Lauraceae species (Laurus nobilis, Lindera megaphylla, Litsea sericea and Umbellularia californica) that show rather similar vessel and pit dimensions was studied using transmission electron microscopy (TEM). The TEM observations confirmed the presence of a pectic matrix associated with intervessel pit membranes, indicating that the relative abundance of acidic versus methylesterified pectins was closely related to the ionic effect. The two species examined with a high ionic effect ~20%, i.e. Laurus nobilis and Umbellularia californica) showed relatively high levels of acidic pectins, whereas methylesterified pectins were abundant in Lindera megaphylla and Litsea sericea, which showed a low ionic effect (~10%). Variation in the ionic effect is strongly associated with the chemical nature of pit membrane pectins in the species studied. Our findings support the current interpretation of the ionic effect due to dynamic swelling and shrinking behaviour of pit membrane pectins. 相似文献
