The aim of this study was to determine the frequency of carriage of methicillin-resistant Staphylococcus pseudintermedius (MRSP) among dogs admitted to a small animal hospital during a 17-month period, to characterize these isolates and to initially screen for possible factors associated with MRSP carriage. Swabs were taken from the nose/pharynx and the perineum as well as from wounds and skin infections (if present) of 814 dogs before entering the small animal hospital. A questionnaire for background information was completed. The staphylococcal species and methicillin resistance were confirmed pheno- and genotypically. The identified MRSP isolates were characterized by SCCmec typing, testing for susceptibility to 25 antimicrobial agents and SmaI-directed pulsed-field gel electrophoresis. A first screening for possible risk factors for MRSP carriage was performed by means of unifactorial contingency tables and CART analysis. Sixty (7.4%) dogs were positive for MRSP. All MRSP isolates harboured a type II-III SCCmec cassette and showed extended resistance to antimicrobial agents. Fifteen different SmaI patterns were observed. The major factors that clustered with MRSP carriage were former hospitalization and antibiotic treatment within the last six months before sampling. This study showed that only a minor part of the sampled dogs carried multi-resistant MRSP isolates. The facts that prior hospitalization and/or antibiotic therapy are potential associated factors for MRSP carriage underline the necessity of a judicious use of antibiotics in small animal medicine.  相似文献

Human activity has induced a multitude of global changes that are likely to affect the functioning of ecosystems. Although these changes act in concert, studies on interactive effects are scarce. Here, we conducted a laboratory microcosm experiment to explore the impacts of temperature (9, 12 and 15 °C), changes in soil humidity (moist, dry) and plant diversity (1, 4, 16 species) on soil microbial activity and litter decomposition.We found that changes in litter decomposition did not mirror impacts on microbial measures indicating that the duration of the experiment (22 weeks) may not have been sufficient to determine the full magnitude of global change effects. However and notably, changes in temperature, humidity and plant litter diversity/composition affected in a non-additive way the microbial parameters investigated. For instance, microbial metabolic efficiency increased with plant diversity in the high moisture treatment but remained unaffected in low moisture treatment suggesting that climate changes may mask beneficial effects of biodiversity on ecosystem functioning. Moreover, litter decomposition was unaffected by plant litter diversity/composition but increased with increasing temperature in the high moisture treatment, and decreased with increasing temperature in the low moisture treatment.We conclude that it is inevitable to perform complex experiments considering multiple global change agents in order to realistically predict future changes in ecosystem functioning. Non-additive interactions highlight the context-dependency of impacts of single global change agents.  相似文献

Termites are estimated to contribute between <5 and 19% of the global methane (CH4) emissions. These estimates have large uncertainties because of the limited number of field-based studies and species studied, as well as issues of diurnal and seasonal variations. We measured CH4 fluxes from four common mound-building termite species (Microcerotermes nervosus, M. serratus, Tumulitermes pastinator and Amitermes darwini) diurnally and seasonally in tropical savannas in the Northern Territory, Australia. Our results showed that there were significant diel and seasonal variations of CH4 emissions from termite mounds and we observed large species specific differences. On a diurnal basis, CH4 fluxes were least at the coolest time of the day (∼07.00 h) and greatest at the warmest (∼15.00 h) for all species for both wet and dry seasons. We observed a strong and significant positive correlation between CH4 flux and mound temperature for all species. A mound excavation experiment demonstrated that the positive temperature effect on CH4 emissions was not related to termite movement in and out of a mound but probably a direct effect of temperature on methanogenesis in the termite gut. Fluxes in the wet season were 5-26-fold greater than those in the dry season. A multiple stepwise regression model including mound temperature and mound water content described 70-99% of the seasonal variations in CH4 fluxes for different species. CH4 fluxes from M. nervosus, which was the most abundant mound-building termite species at our sites, had significantly lower fluxes than the other three species measured. Our data demonstrate that CH4 flux estimates could result in large under- or over-estimation of CH4 emissions from termites if the diurnal, seasonal and species specific variations are not accounted for, especially when flux data are extrapolated to landscape scales.  相似文献

Tropical savanna ecosystems are a major contributor to global CO2, CH4 and N2O greenhouse gas exchange. Savanna fire events represent large, discrete C emissions but the importance of ongoing soil-atmosphere gas exchange is less well understood. Seasonal rainfall and fire events are likely to impact upon savanna soil microbial processes involved in N2O and CH4 exchange. We measured soil CO2, CH4 and N2O fluxes in savanna woodland (Eucalyptus tetrodonta/Eucalyptus miniata trees above sorghum grass) at Howard Springs, Australia over a 16 month period from October 2007 to January 2009 using manual chambers and a field-based gas chromatograph connected to automated chambers. The effect of fire on soil gas exchange was investigated through two controlled burns and protected unburnt areas. Fire is a frequent natural and management action in these savanna (every 1-2 years). There was no seasonal change and no fire effect upon soil N2O exchange. Soil N2O fluxes were very low, generally between −1.0 and 1.0 μg N m−2 h−1, and often below the minimum detection limit. There was an increase in soil NH4+ in the months after the 2008 fire event, but no change in soil NO3. There was considerable nitrification in the early wet season but minimal nitrification at all other times.Savanna soil was generally a net CH4 sink that equated to between −2.0 and −1.6 kg CH4 ha−1 y−1 with no clear seasonal pattern in response to changing soil moisture conditions. Irrigation in the dry season significantly reduced soil gas diffusion and as a consequence soil CH4 uptake. There were short periods of soil CH4 emission, up to 20 μg C m−2 h−1, likely to have been caused by termite activity in, or beneath, automated chambers. Soil CO2 fluxes showed a strong bimodal seasonal pattern, increasing fivefold from the dry into the wet season. Soil moisture showed a weak relationship with soil CH4 fluxes, but a much stronger relationship with soil CO2 fluxes, explaining up to 70% of the variation in unburnt treatments. Australian savanna soils are a small N2O source, and possibly even a sink. Annual soil CH4 flux measurements suggest that the 1.9 million km2 of Australian savanna soils may provide a C sink of between −7.7 and −9.4 Tg CO2-e per year. This sink estimate would offset potentially 10% of Australian transport related CO2-e emissions. This CH4 sink estimate does not include concurrent CH4 emissions from termite mounds or ephemeral wetlands in Australian savannas.  相似文献

Large sized biopores (diameter >2 mm) in the subsoil can be created by tap roots, which leave voids after their decay, or by the burrowing activity of anecic earthworms which may benefit from the temporary lack in tillage in perennial cropping systems. However, the interactions between root growth and earthworm activity in the process of biopore formation during perennial ley cropping are not well understood. The aim of this field study was to quantify the development of the abundance of the anecic earthworm Lumbricus terrestris and the biopore density during the cultivation of lucerne (Medicago sativa L.), chicory (Cichorium intybus L.) and tall fescue (Festuca arundinacea Schreb.) grown for either one, two or three years. An increased abundance of L. terrestris was already recorded after two years of continuous ley when compared with one year cultivation. The ley crop species had only minor influence on the abundance of L. terrestris. Biopore densities of both diameter classes under study (2–5 mm and >5 mm) were not significantly affected by the duration of ley cropping. In contrast, biopore densities were influenced by ley crop species. More biopores of diameter class 2–5 mm were recorded after chicory than after fescue. Lucerne cropping resulted in intermediate biopore density. Additionally, in an incubation experiment under field conditions, we quantified whether L. terrestris preferentially created new biopores or colonized abandoned, previously existing ones. After three weeks of incubation, one third of the adult individuals incubated in the experimental area created new biopores at 0.4 m soil depth. A similar percentage of individuals colonized previously existing biopores, partially widening the lumen of smaller sized biopores. The remaining individuals remained in the topsoil. Sub-adult individuals rarely formed new pores. Half of the introduced sub-adults remained in the topsoil. We conclude that in crop rotations new biopores can be generated during perennial ley cropping by taproot systems of ley crops, but that a two to three- year period without tillage is not sufficient for populations of anecic earthworms to make a marked contribution to biopore density in the subsoil. The relevance of anecic earthworms for altering physical and chemical properties of biopores during ley cropping still needs further investigation.  相似文献

Flowers of Ziziphus spina christi are known to be attractive for parasitoids and predators. In Y-tube olfactometer experiments, the dried flowers attracted significantly (p < 0.001) the female parasitoids Aphelinus abdominalis. The flower volatile compounds were analyzed to understand which compounds could be specifically responsible for this attractiveness. The volatile compounds of Ziziphus flowers were extracted by closed-loop-stripping-analysis (CLSA) and also by solid phase microextraction (SPME) followed by gas chromatography-mass spectrometry (GC-MS) analysis. The main chemical classes of the volatile compounds are aldehydes, monoterpene-alcohols, ketones and hydrocarbons. Flower extract and some specific compounds will be further tested for their responsiveness to predators and parasitoids in behavioural and electrophysiological experiments.  相似文献

Crop protection seldom takes into account soil heterogeneity at the field scale. Yet, variable site characteristics affect the incidence of pests as well as the efficacy and fate of pesticides in soil. This article reviews crucial starting points for incorporating soil information into precision crop protection (PCP). At present, the lack of adequate field maps is a major drawback. Conventional soil analyses are too expensive to capture soil heterogeneity at the field scale with the required spatial resolution. Therefore, we discuss alternative procedures exemplified by our own results concerning (i) minimally and non-invasive sensor techniques for the estimation of soil properties, (ii) the evidence of soil heterogeneity with respect to PCP, and (iii) current possibilities for incorporation of high resolution soil information into crop protection decisions. Soil organic carbon (SOC) and soil texture are extremely interesting for PCP. Their determination with minimally invasive techniques requires the sampling of soils, because the sensors must be used in the laboratory. However, this technique delivers precise information at low cost. We accurately determined SOC in the near-infrared. In the mid-infrared, texture and lime content were also exactly quantified. Non-invasive sensors require less effort. The airborne HyMap sensor was suitable for the detection of variability in SOC at high resolution, thus promising further progress regarding SOC data acquisition from bare soil. The apparent electrical conductivity as measured by an EM38 sensor was shown to be a suitable proxy for soil texture and layering. A survey of arable fields near Bonn (Germany) revealed widespread within-field heterogeneity of texture-related ECa, SOC and other characteristics. Maps of herbicide sorption and application rate were derived from sensor data, showing that optimal herbicide dosage is strongly governed by soil variability. A phytoassay with isoproturon confirmed the reliability of spatially varied herbicide application rates. Mapping areas with an enhanced leaching risk within fields allows them to be kept free of pesticides with related regulatory restrictions. We conclude that the use of information on soil heterogeneity within the concept of PCP is beneficial, both economically and ecologically.  相似文献
