The distribution of selected elements in individual fractions of organic matter from anthropogenically contaminated soils was investigated. The attention was paid especially at Hg. Furthermore, contents of S, Mg, Mn, Fe, Cu, Zn and Pb were also measured. The decomposition of organic matter to particular fractions was carried out by the resin DAX-8. Ten soil samples were collected, and the Advanced Mercury Analyzer (AMA-254) was used for the determination of the total Hg content. The two highest Hg values reached up to the concentration 10.5 mg kg?1, and in the highest one, it was almost 29 mg kg?1. In each extract, mercury was measured by inductively coupled plasma mass spectrometry (ICP-MS), for other elements, inductively coupled plasma optical emission spectrometry (ICP-OES) was applied. Results of the analysis show that the Hg content bound to the humic acids is inversely proportional to the content of Mg, Mn, Fe and Cu. However, this dependence was not confirmed by the samples with the mercury content above 10 mg kg?1. In the case of fulvic acids, the relationship between Hg and S was observed and has again an inverse character.  相似文献

We quantified the effect of water and nutrient availability on aboveground biomass and nitrogen accumulation and partitioning in four species from the southeastern United States, loblolly pine (Pinus taeda), slash pine (Pinus elliottii), sweetgum (Liquidambar styraciflua), and sycamore (Platanus occidentalis). The 6-year-old stands received five levels of resource input (control, irrigation with 3.05 cm water week−1, irrigation + 57 kg N ha−1 year−1, irrigation + 85 kg N ha−1 year−1, and irrigation + 114 kg N ha−1 year−1). Irrigation significantly increased foliage, stem, and branch biomass for sweetgum and sycamore, culminating in 103% and 238% increases in total aboveground biomass. Fertilization significantly increased aboveground components for all species resulting in 49, 58, 281, and 132% increases in total aboveground biomass for loblolly pine, slash pine, sweetgum, and sycamore, respectively. Standing total aboveground biomass of the fertilized treatments reached 79, 59, 48, and 54 Mg ha−1 for loblolly pine, slash pine, sweetgum, and sycamore, respectively. Fertilization increased foliar nitrogen concentration for loblolly pine, sweetgum, and sycamore foliage. Irrigation increased total stand nitrogen content by 6, 14, 93, and 161% for loblolly pine, slash pine, sweetgum, and sycamore, respectively. Fertilization increased total nitrogen content by 62, 53, 172, and 69% with maximum nitrogen contents of 267, 212, 237, and 203 kg ha−1 for loblolly pine, slash pine, sweetgum, and sycamore, respectively. Growth efficiency (stem growth per unit of leaf biomass) and nitrogen use efficiency (stem growth per unit of foliar nitrogen content) increased for the sycamore and sweetgum, but not the loblolly or slash pine.  相似文献

Nine of 74 American alligators (Alligator mississippiensis) from a captive Florida herd of 3-4-m-long, 200-350-kg, adult males greater than 30 yr of age died within a 10-day period during 1995. Nonspecific clinical signs included anorexia, lethargy, muscle weakness, paraparesis, bilateral white ocular discharge, and various degrees of periocular, facial, cervical, and limb edema. Pneumonia, pericarditis, and arthritis were found on postmortem evaluation of the spontaneously dead and euthanatized alligators. Rapidly growing mycoplasmas were identified by culture, and mycoplasma nucleotide sequences were identified by polymerase chain reaction testing of fresh lung and synovial fluid from an affected alligator. Culture of banked frozen lung from necropsy specimens and fresh lung and fresh synovial fluid from newly affected alligators confirmed the presence of a new mycoplasma species in seven of eight individuals. Oxytetracycline was administered, but related deaths continued for 6 mo until only 14 of the initial alligators remained. An enzyme-linked immunosorbent assay to detect antibody was developed, and the organism was transmitted experimentally to naive juvenile alligators, although the source of the organism, Mycoplasma sp. (ATCC 700619), has not been identified. The alligator isolate is a novel species in the mycoplasma family because its nucleotide sequence does not match those of over 75 characterized mycoplasma species. Such factors as population density, animal age, and mycoplasmal virulence likely contributed to the course of disease.  相似文献

Pools of macro-nutrients in soil and vegetation were studied in an old fertilization experiment with a large previous input of N. Different doses of N, in the form of urea, had been added four times during a 20-year period. In total, between 480 and 2400 kg N ha−1 had been given. The experiment was established in a relatively productive Norway spruce stand and the expectation was that the large N input would cause an accelerated leaching of N, especially nitrate, accompanied by soil acidification and losses of several nutrients. The aim was to test for possible residual effects. Thirteen years after the last N addition, samples from the aboveground part of trees, field layer, S-layer, humus layer and mineral soil (0–10 cm) were analyzed for concentrations of most major nutrients. Nutrient pools were calculated. In the humus layer, the concentration of N increased and the C/N-ratio decreased with increasing N dose. The calculated recovery of added N in soil including ground vegetation was complete for the lowest N dose, while it was 25–50% for higher doses. The amount of N retained was unaffected by the N dose. The amount of extractable P in the upper part of the mineral soil was negatively correlated with N dose, as was also the concentration of total P in the S-layer. Neither soil pH, nor concentrations or amounts of Ca, Mg and K were affected by the previous fertilization. The calculated total soil-plant pool was only influenced by N dose in the case of P, which was 20% lower at the highest N dose compared with unfertilized conditions. Despite the large extra N input, the nutritional changes in plants and soil of the actual study site seemed surprisingly small.  相似文献

Twenty-one adult cats, allotted into 2 groups, were given gentamicin sulfate at dosages of either 5.0 mg/kg of body weight or 2.5 mg/kg as a single IM injection. During a 24-hour period, serum concentrations of gentamicin were measured serially, using a fluorescence immunoassay. The mean peak serum concentration of gentamicin in cats given 5.0 mg/kg was 23.1 micrograms/ml at postinjection hour (PIH) 0.5; thereafter, the mean serum concentration steadily decreased to 2.0 micrograms/ml at PIH 24. The mean peak serum concentration for cats administered 2.5 mg/kg was 9.1 micrograms/ml at PIH 0.5; thereafter, the mean serum concentration steadily decreased to 1.3 micrograms/ml at PIH 12. Serum therapeutic concentrations, without exceeding toxic concentrations, were attained at the 2.5 mg/kg dosage.  相似文献

Roots of wheat and oat seedlings were treated with diclofop-methyl (methyl 2-[4-(2′,4′-dichlorophenoxy)phenoxy]propanoate) in a specially designed Plexiglas treatment apparatus. Diclofopmethyl severely inhibited the root growth of susceptible oat seedlings but roots of resistant wheat seedlings were unaffected. Diclofop-methyl at 0.3 μM reduced the growth of oat roots to 50% of the control. Direct contact between diclofop-methyl and the inhibited root zone was necessary for growth inhibition since other parts of the seedling (roots and shoots) isolated from contact with diclofop-methyl solution by a physical barrier were unaffected. Diclofop (2-[4-(2′,4′-dichlorophenoxy)phenoxy]propionic acid), the free acid metabolite of diclofop-methyl, was somewhat more phytotoxic than the parent compound. The herbicide, 2,4-dichlorophenoxyacetic acid (2,4-D), which engenders auxin responses, slightly enhanced the inhibition of oat root growth by diclofop-methyl. The primary wheat metabolite, ring-hydroxylated diclofop, was nonphytotoxic to oat root growth, whereas the acetylated derivative of the primary water-soluble oat metabolite (neutral glucose ester of diclofop) inhibited oat root growth to the same extent as diclofop-methyl. These results support the hypothesis that the basis for selectivity between resistant wheat and susceptible oat is the metabolism of diclofop-methyl by aryl hydroxylation and conjugation but not glucose ester conjugation. Translocation is also not an important factor in the phytotoxic activity of diclofop-methyl.  相似文献

A group of SPF cats were moderately susceptible to the causal organism of contagious equine metritis (CEM) following intra-uterine or intrapreputial challenge with an Irish streptomycin resistant strain isolated from a clinically infected mare. Subclinical infections were established in only 50% of the cats, none of which became long-term carriers of the organism. Cytological examination of vaginal smears was of no diagnostic value in confirming infection in inapparently infected cats. Bacteriological responses after primary or secondary challenge with the CEM organism were essentially similar, with one exception, a female cat in which there was possible evidence of local immunity persisting after the primary infection. Efforts to reactivate shedding subsequent to the immediate post-challenge period were unsuccessful. Throughout the experimental period, the cats remained sero-negative to the complement-fixation test, and they failed to develop any significant increase in the levels of antibody activity as measured by the kinetics-based ELISA or KELA system. On day 89 after primary challenge, the cats were euthanized and various sites in the genitourinary tract and the internal iliac lymphatic glands subjected to bacteriological and pathological examination for evidence of CEM infection with negative results. The findings of this study, although establishing the transmissibility of the CEM organism for the cat, demonstrate the limited value of this species as an experimental model system for the disease in the horse.  相似文献

Measurements (n = 126) of end-tidal halothane concentrations were taken from 21 horses anesthetized for routine and emergency surgery. One hundred five paired values allowed comparison of gas samples taken near the oral end of the endotracheal tube (Y1) to samples obtained at the cuffed end of the endotracheal tube (Y2). Twenty-one paired readings were assessed to compare samples taken 25 cm beyond the cuffed end of the tube (Y3) to samples from Y1. Measurements were made at all locations at 15-minute intervals starting 30 minutes after beginning halothane. All measurements were made in triplicate at end-expiration, and both sites were sampled within 1 minute of each other. Halothane concentration was measured by rapid infrared analysis with a gas sampling rate of 150 ml/min and displayed as a digital reading. Calibration of the machine was checked regularly. The difference between readings (Y1-Y2 or Y1-Y3 = Sdif) was tested using general linear models and a significance level of p less than 0.05 was used. The variable Sdif was analyzed with respect to time, mode of ventilation, and type of recumbency; no effects of these variables were detected. The mean values (+/- SD) of 105 readings for Y1 and Y2 were 2.41 vol% (+/- 0.49) and 2.39 vol% (+/- 0.49) respectively, and the Pearson's correlation coefficient (Y1 vs Y2) was 0.96. The mean values (+/- SD) of 21 measurements for Y1 and Y3 were 2.31 vol% (+/- 0.27) and 2.32 vol% (+/- 0.28) respectively and the Pearson's correlation coefficient (Y1 vs Y3) was 0.98.  相似文献

Spiral shaped bacteria present in blood smears and bone marrow of a sick rhinoceros iguana were examined by light and electron microscopy. The organisms averaged 10 micron in length and had at least three spiral turns. The cell contained nuclear areas, vacuoles and ribosomes, except at the poles where there was a virtual absence of organelles. The bacterium was found by a cell membrane, cell wall and enveloping sheath. "Blebs" were present with regularity on the cell surface. About 14 flagella were present at each pole, and at these sites there was a specialized thickening of the cell membrane. Organisms were present within a phagocytic vacuole of macrophages in the blood and bone marrow, and often these engulfed organisms were degenerated. The taxonomic position of the bacterium is unknown.  相似文献
