To obtain a more detailed understanding of the prevalence of classical scrapie infections in a heavily affected German sheep flock (composed of 603 sheep and 6 goats), we analysed 169 sheep and 6 goats that carried the genotypes susceptible to the disease and that were therefore culled following discovery of the index case. The initial tests were performed using the Biorad TeSeE ELISA and reactive results were verified by official confirmatory methods (OIE-immunoblot and/or immunohistochemistry (IHC)) to demonstrate the deposition of scrapie-associated PrP(Sc) in the brain stem (obex). This approach led to the discovery of 40 additional subclinically scrapie-infected sheep. Furthermore, peripheral lymphatic and nervous tissue samples of the 129 sheep and 6 goats with a negative CNS result were examined by IHC in order to identify any preclinical infections which had not already spread to the central nervous system (CNS). Using this approach we found 13 additional sheep with PrP(Sc) depositions in the gut-associated lymph nodes (GALT) as well as in the enteric nervous system. Moreover, in most of these cases PrP(Sc) was also deposited in the spleen and in the retropharyngeal and superficial cervical lymph nodes. Taken together, these results show a 30.3% infection prevalence in this scrapie-affected flock. Almost 7.4% of the infected animals harboured PrP(Sc) exclusively in the peripheral lymphatic and nervous tissue and were therefore missed by the currently used testing strategy. 相似文献

Am Beispiel der Mineralisierung organischer Bodensubstanz und der assoziativen biologischen Luftstickstoffbindung durch Bakterien wird der Einfluß der C‐ und N‐Verfügbarkeit in der Rhizosphäre auf die bakterielle Aktivität und die Expression des Nitrogenase Enzyms analysiert und diskutiert. Sowohl Düngung als auch die Bakterienpopulationsstruktur in der Rhizosphäre beeinflussen die Menge und Zusammensetzung der Wurzelexsudate und somit die Keimzahl und die Aktivität der Rhizosphärenmikroflora. Eigene und Literaturergebnisse weisen auf die Bedeutung der Ernährungs‐ und Wachstumsbedingungen der Pflanzen hin, wenn Bruttonährstoffumsetzungen in der Rhizosphäre gemessen werden sollen. 相似文献

Background
Different animal models are used as fracture models in orthopaedic research prior to implant use in humans, although biomechanical forces can differ to a great extend between species due to variable anatomic conditions, particularly with regard to the gait. The rabbit is an often used fracture model, but biomechanical data are very rare. The objective of the present study was to measure axial forces, bending moments, and bending axis directly in the rabbit tibia in vivo. The following hypothesis was tested: Axial forces and bending moments in the mid-diaphysis of rabbit tibia differ from other experimental animals or indirectly calculated data.Methods
A minifixateur system with 4 force sensors was developed and attached to rabbit tibia (n = 4), which were subsequently ostectomised. Axial forces, bending moments and bending angles were calculated telemetrically during weight bearing in motion between 6 and 42 days post operation.Results
Highest single values were 201% body weight [% bw] for axial forces and 409% bw cm for bending moments. Whereas there was a continous decrease in axial forces over time after day 10 (P = 0.03 on day 15), a decrease in bending moments was inconsistent (P = 0.03 on day 27). High values for bending moments were frequently, but not consistently, associated with high values for axial forces.Conclusion
Axial forces in rabbit tibia exceeded axial forces in sheep, and differed from indirectly calculated data. The rabbit is an appropriate fracture model because axial loads and bending moments in rabbit tibia were more closely to human conditions than in sheep tibia as an animal model. 相似文献164.

Bovine abortion of unknown infectious aetiology still remains a major economic problem. In this study, we focused on a new possible abortigenic agent called Parachlamydia acanthamoebae. Retrospective samples (n=235) taken from late-term abortions in cattle were investigated by real-time diagnostic PCR for Chlamydiaceae and Parachlamydia spp., respectively. Histological sections of cases positive by real-time PCR for any Chlamydia-related agent were further examined by immunohistochemistry using specific antibodies. Chlamydophila abortus was detected only in three cases (1.3%) by real-time PCR and ArrayTube Microarray playing a less important role in bovine abortion compared to the situation in small ruminants in Switzerland. By real-time PCR as many as 43 of 235 (18.3%) cases turned out to be positive for Parachlamydia. The presence of Parachlamydia within placental lesions was confirmed in 35 cases (81.4%) by immunohistochemistry. The main histopathological feature in parachlamydial abortion was purulent to necrotizing placentitis (25/43). Parachlamydia should be considered as a new abortigenic agent in Swiss cattle. Since Parachlamydia may be involved in lower respiratory tract infections in humans, bovine abortion material should be handled with care given the possible zoonotic risk. 相似文献

In nine Bolivian purple corn ( Zea mays L.) varieties the content of phenolic compounds as well as the anthocyanin composition has been determined. The phenotypes under investigation included four red and five blue varieties (Kulli, Ayzuma, Paru, Tuimuru, Oke, Huaca Songo, Colorado, Huillcaparu, and Checchi). In purple corn, phenolic compounds were highly concentrated in cell walls. Thus, simultaneous determination of soluble and bound-form phenolics is essential for analysis, extraction, and quantification. The present study reports the determination of soluble and insoluble-bound fraction of phenolic compounds by HPLC-DAD and HPLC-ESI-MS(n) in Bolivian purple corn varieties. Enzymatic, thermal, and alkaline hydrolyses were used to obtain the cell wall-linked phenolic compounds. Ferulic acid values ranged from 132.9 to 298.4 mg/100 g, and p-coumaric acid contents varied between 251.8 and 607.5 mg/100 g dry weight (DW), respectively, and were identified as the main nonanthocyanin phenolics. The total content of phenolic compounds ranged from 311.0 to 817.6 mg gallic acid equivalents (GAE)/100 g DW, and the percentage contribution of bound to total phenolics varied from 62.1 to 86.6%. The total monomeric anthocyanin content ranged from 1.9 to 71.7 mg cyanidin-3-glucoside equivalents/100 g DW. Anthocyanin profiles are almost the same among the different samples. Differences are observed only in the relative percentage of each anthocyanin. Cyanidin-3-glucoside and its malonated derivative were detected as major anthocyanins. Several dimalonylated monoglucosides of cyanidin, peonidin, and pelargonidin were present as minor constituents. 相似文献

The organic matter supply can promote the dispersal and activity of applied plant growth–promoting rhizobacteria (PGPR), but the complementary effect of organic fertilization and PGPR application on the turnover of P is scarcely known. The effects of the application of two PGPR strains (Pseudomonas fluorescens strain DR54 and Enterobacter radicincitans sp. nov. strain DSM 16656) alone and in combination with organic fertilization (cattle manure and biowaste compost) on growth and P uptake of maize (Zea mays L.) and oilseed rape (Brassica napus L.) were investigated under semi–field conditions. Furthermore, P pools and phosphatase activities in soil and the arbuscular mycorrhizal colonization of maize were examined. The organic‐fertilizer amendments increased the growth and P uptake of both plant species and the soil P pools. The application of the E. radicincitans strain increased P uptake of oilseed rape when no organic fertilizer was added. Furthermore, the application of both bacterial strains increased the activities of phosphatases under both plant species. Here, the effect of the PGPR application even exceeded the effect of organic fertilization. The magnitude of this effect varied between the different fertilizing treatments and between the two bacterial strains. Phosphatase activities were increased to the greatest extent after application of P. fluorescens in the unfertilized soil. Under rape increases of 52% for acid phosphatase activities (ACP), 103% for alkaline phosphatase activities (ALP), and 133% for phosphodiesterase (PDE) were observed therewith. In the unfertilized soil, the application of P. fluorescens also resulted in a strong increase of the arbuscular mycorrhizal colonization of maize. We conclude that application of PGPR can promote the P mobilization and supply of crops in P‐deficient soils, however, in combination with organic fertilization these effects might be masked by a general improved P supply of the crops. Interactive effects of applied bacterial strains and organic fertilization depend on the sort of organic fertilizer and crop species used. 相似文献

Abstract Measurements of gross N transfer in soils have as yet not distinguished between biological or physico-chemical processes. Here, we present a new approach that allows microbially-mediated gross N transfer rates to be estimated in undisturbed soils without adding 15N. It is based on the assumption that in undisturbed soil, the soil microbial growth rate is equal to its death rate. To assess the contribution of biological versus physico-chemical N transfer processes, we combined the new approach with the 15N-pool dilution technique. The relationship between both processes varied with soil C and fine particle contents. Nearly equal rates were observed within the carbon-poor soil (0.35% Corg, low fine particle content), whereas up to 2.5 times higher physico-chemical than biological N transfer rates were measured within the carbon-enriched soil (0.86% Corg, higher fine particle content). Furthermore, microbially-mediated gross N transfer rates increased three-fold after N fertilization compared to the unfertilized control. 相似文献

One of the challenges in organic farming systems is to match nitrogen (N) mineralization from organic fertilizers and crop
demand for N. The mineralization rate of organic N is mainly determined by the chemical composition of the organic matter
being decomposed and the activity of the soil microflora. It has been shown that long-term organic fertilization can affect
soil microbial biomass (MB), the microbial community structure, and the activity of enzymes involved in the decomposition
of organic matter, but whether this has an impact on short-term N mineralization from recently applied organic substances
is not yet clear. Here, we sampled soils from a long-term field experiment, which had either not been fertilized, or fertilized
with 30 or 60 t ha−1 year−1 of farmyard manure (FYM) since 1989. These soil samples were used in a 10-week pot experiment with or without addition of
FYM before starting (recent fertilization). At the start and end of this experiment, soil MB, microbial basal respiration,
total plant N, and mineral soil N content were measured, and a simplified N balance was calculated. Although the different
treatments used in the long-term experiment induced significant differences in soil MB, as well as total soil C and N contents,
the total N mineralization from FYM was not significantly affected by soil fertilization history. The amount of N released
from FYM and not immobilized by soil microflora was about twice as high in the soil that had been fertilized with 60 t ha−1 year−1 of FYM as compared with the non-fertilized soil (p < 0.05). 相似文献

Experiments to assess the rate of absorption and translocation of foliar‐applied, isotopically labeled boric acid (BA) were carried out with lychee (Litchi chinensis Sonn.) and soybean (Glycine max [L.] Merr.) plants. Boron (B) absorption and translocation within the plant, one week after treatment, was investigated after adding to the boric acid (BA solutions 0.5 mM CaCl2 and/or 50 or 500 mM sorbitol). The contribution of stomata to the absorption process was assessed by applying the solutions either to the adaxial or to the abaxial leaf side. Both plant species differed greatly in total absorption rates. The adaxial leaf surface (lacking stomata) of lychee leaves was nearly impermeable, while the stomatous abaxial surface was permeable to BA solutions. In this species, no translocation of 10B to other leaf parts and no effect of adjuvants in increasing 10B absorption were recorded. In contrast, 10B was absorbed both by adaxial and abaxial leaf surfaces of soybean leaves. Boron concentrations measured in treated soybean leaves were sixfold higher after application to the abaxial as compared to the adaxial leaf surface. The addition of adjuvants significantly enhanced the rate of 10B absorption, but not its translocation within the plant. Treatments containing 500 mM sorbitol led to increased 10B absorption and enhanced acropetal 10B movement, whereas adding only 50 mM sorbitol had no significant effect. Application of 0.5 mM CaCl2 in combination with 500 mM sorbitol decreased the rate of 10B absorption, compared to the performance of 500 mM sorbitol alone. Basipetal 10B translocation was very limited. A distinct effect of B‐sorbitol complexes on B translocation apart from the pure adjuvant effect could not be discerned in this investigation. 相似文献

The Global Oscillation Network Group (GONG) project estimates the frequencies, amplitudes, and linewidths of more than 250,000 acoustic resonances of the sun from data sets lasting 36 days. The frequency resolution of a single data set is 0.321 microhertz. For frequencies averaged over the azimuthal order m, the median formal error is 0.044 microhertz, and the associated median fractional error is 1.6 x 10(-5). For a 3-year data set, the fractional error is expected to be 3 x 10(-6). The GONG m-averaged frequency measurements differ from other helioseismic data sets by 0.03 to 0.08 microhertz. The differences arise from a combination of systematic errors, random errors, and possible changes in solar structure. 相似文献
