Objective
The fixed-dose combination of butorphanol, azaperone and medetomidine (BAM; 30, 12 and 12 mg mL?1, respectively) with subsequent antagonism by naltrexone–atipamezole was evaluated for reversible immobilization of captive blesbok (Damaliscus pygargus phillipsi).Study design
Prospective, clinical trial.Animals
Sixteen blesbok (four males and twelve females), weighing 52.5?71.0 kg, were immobilized in South Africa.Methods
The total dose of BAM ranged from 0.5 to 0.7 mL for females and 0.7 to 0.9 mL for males. In seven animals chosen randomly, 8000 units of hyaluronidase was added to the dart. Physiologic variables were recorded every 5 minutes beginning at 10?20 minutes after darting. Arterial blood samples were collected three times at 20, 30 and 40 minutes after darting for analysis of blood acid-base status.Results
The mean administered doses of BAM were as follows: butorphanol (0.34 ± 0.08 mg kg?1), azaperone (0.14 ± 0.03 mg kg?1) and medetomidine (0.14 ± 0.03 mg kg?1). The inductions were calm and smooth. The mean induction time was 9.6 ± 3.2 minutes with just BAM and 5.1 ± 0.8 minutes with BAM and hyaluronidase combination. Heart rate (45 ± 6 beats minute?1) and respiratory frequency (38 ± 4 breaths minute?1) were stable throughout immobilization. The mean arterial blood pressure for all animals was stable but elevated (137 ± 7 mmHg). Rectal temperature slightly increased over time but remained within an acceptable range. The recovery time after administering naltrexone and atipamezole was 4.8 ± 0.7 minutes.Conclusion and clinical relevance

Strains and isolates of Coxiella burnetii, an obligate intracellular bacterium, carry a single plasmid or a plasmid-homologous sequence integrated into the chromosome. The plasmids QpH1, QpRS, QpDV and the chromosome-integrated plasmid-homologous region have been completely sequenced, whereas no sequence data are available for the QpDG plasmid. In this study, we used total genomic DNA from reference strain C. burnetii Dugway 5J108-111 to demonstrate and characterize the QpDG plasmid by pulsed-field gel electrophoresis (PFGE) and Southern hybridization. Primers derived from regions shared among C. burnetii plasmids were used to construct a physical map of the QpDG plasmid by extra long (XL) PCR. Both approaches, Southern hybridization and XL PCR indicated that QpDG and QpH1 represent a closely related and likely identical plasmid. 相似文献

The process of recolonization after disturbance is crucial for the persistence and dynamics of patch-tracking metapopulations.
We developed a model to compare the spatial distribution and spatial genetic structure of the epiphytic lichen Lobaria pulmonaria within the perimeter of two reconstructed 19th century disturbances with a nearby reference area without stand-level disturbance.
Population genetic data suggested that after stand-replacing disturbance, each plot was colonized by one or a few genotypes
only, which subsequently spread clonally within a local neighborhood. The model (cellular automaton) aimed at testing the
validity of this interpretation and at assessing the relative importance of local dispersal of clonal propagules vs. long-distance
dispersal of clonal and/or sexual diaspores. A reasonable model fit was reached for the empirical data on host tree distribution,
lichen distribution, and tree- and plot-level genotype diversity of the lichen in the reference area. Although model calibration
suggested a predominance of local dispersal of clonal propagules, a substantial contribution of immigration of non-local genotypes
by long-distance dispersal was needed to reach the observed levels of genotype diversity. The model could not fully explain
the high degree of clonality after stand-replacing disturbance, suggesting that the dispersal process itself may not be stationary

Twenty lions, 11 males and nine females, weighing 38–284 kg were immobilized in South Africa.Methods
The BAM volume dose rate administered was 0.005–0.008 mL kg?1 (0.6 mL 100 kg?1). Physiologic variables were recorded every 5 minutes. Four arterial blood samples were collected from all animals at 20, 30, 40 and 50 minutes after immobilization for analysis of blood-gases and acid-base status.Results
The actual doses administered were as follows: butorphanol, 0.18 ± 0.03 mg kg?1; azaperone, 0.07 ± 0.01 mg kg?1; and medetomidine, 0.07 ± 0.01 mg kg?1. The inductions were calm and smooth, and induction time ranged from 4 to 10 minutes (7 ± 2 minutes). The amount of time needed to work with each lion was 70 minutes, and no additional drug doses were needed. Heart rate (40 ± 8 beats minute?1) and respiratory frequency (15 ± 4 breaths minute?1) were stable throughout immobilization. The mean arterial blood pressure of all animals was stable but elevated (142 ± 16 mmHg). The rectal temperature slightly increased over time but remained within acceptable range. The recovery time was significantly shorter when using naltrexone and atipamezole (9 ± 1 minutes) compared to using naltrexone and yohimbine (22 ± 7 minutes).Conclusion and clinical relevance
The BAM combination proved to be reliable for general veterinary anaesthesia in lions. During anaesthesia, minor veterinary procedures such a blood collection, intubation, vaccination and collaring could safely be performed with no additional dosing required. 相似文献145.

BackgroundTo evaluate the efficacy of a single intramuscular adminsitration of long‐acting omeprazole (LA‐OMEP) in increasing gastric pH in dogs.HypothesisWe hypothesized that LA‐OMEP would meet in healthy dogs the clinical goals defined for human patients for treatment of gastroduodenal ulceration.AnimalsNine healthy research dogs.MethodsProspective experimental study. Dogs were given a 4 mg/kg intramuscular injection of LA‐OMEP. Intragastric pH was continuously recorded on treatment days 0 to 7. Daily mean pH and mean percentage time (MPT) intragastric pH was ≥3 or ≥4 were determined.ResultsThe mean onset of action for the LA‐OMEP was 98.11 min (SD 46.39). The mean number of days the dogs'' pH met established goals for MPT pH ≥3 was 5.5 days (range, 3‐7) and 5.25 days for MPT pH ≥4 (range, 3‐7). Long‐acting omeprazole met the human clinical goals pH ≥3 for 72 hours in 8/8 of the dogs and MPT pH ≥4 for 96 hours in 7/8 of dogs.Conclusions and Clinical ImportanceThe LA‐OMEP formulation produced gastric acid suppression in healthy dogs for an average of 5 days and up to 7 days, after a single intramuscular injection. A 6-month-old male bilaterally cryptorchid Boxer was examined for acute abdominal pain. The results of physical examination, laboratory testing, and ultrasonographic examination are presented. Intra-abdominal testicular torsion was suspected and confirmed at surgery. No evidence of neoplasia was found at histopathologic examination of the twisted testicle. Thoracolumbar myelopathy encompasses a number of disease processes such as intervertebral disc disease, discospondylitis, trauma, congenital malformations, neoplasia, and intramedullary spinal cord disease. Compressive disc herniations are most common in dogs and require imaging procedures such as myelography, computed tomography (CT), and/or magnetic resonance imaging (MRI) to determine the need and location for decompressive surgery. The purposes of this retrospective, cross‐sectional study were to evaluate all dogs undergoing thoracolumbar CT imaging as the initial diagnostic step between 2010 and 2015 and determine whether any of the imaging characteristics could be used to predict the need for additional imaging in the form of myelography, CT myelography, and/or MRI. A total of 555 dogs were identified in this time frame which underwent CT imaging for myelopathy of the thoracolumbar region. Various parameters including age, gender, sexual status, breed, chronicity, site of lesion, time of study, and contrast administration were evaluated. Findings indicated that 7.6% of dogs needed additional imaging after CT. Dachshunds were less likely to need additional imaging (P = 0.0111) as were patients scanned during normal business hours (P = 0.0075). Increasing age of the patient increased the likelihood of additional imaging (P = 0.0107). Dogs which did not have additional imaging performed were 21.89 times more likely to require surgery than those which did have additional imaging (P < 0.0001). Findings supported the use of CT as a first‐line imaging modality for dogs presenting with thoracolumbar myelopathy. 相似文献

Oxygen (O2) supply and the related redox potential (EH) are important parameters for interactions between roots and microorganisms in the rhizosphere. Rhizosphere extension in terms of the spatial distribution of O2 concentration and EH is poorly documented under aerobic soil conditions. We investigated how far O2 consumption of roots and microorganisms in the rhizosphere is replenished by O2 diffusion as a function of water/air‐filled porosity. Oxygen concentration and EH in the rhizosphere were monitored at a mm‐scale by means of electroreductive Clark‐type sensors and miniaturized EH electrodes under various matric potential ranges. Respiratory activity of roots and microorganisms was calculated from O2 profiles and diffusion coefficients. pH profiles were determined in thin soil layers sliced near the root surface. Gradients of O2 concentration and the extent of anoxic zones depended on the respiratory activity near the root surface. Matric potential, reflecting air‐filled porosity, was found to be the most important factor affecting O2 transport in the rhizosphere. Under water‐saturated conditions and near field capacity up to –200 hPa, O2 transport was limited, causing a decline in oxygen partial pressures (pO2) to values between 0 and 3 kPa at the root surface. Aerobic respiration increased by a factor of 100 when comparing the saturated with the driest status. At an air‐filled porosity of 9% to 12%, diffusion of O2 increased considerably. This was confirmed by EH around 300 mV under aerated conditions, while EH decreased to 100 mV on the root surface under near water‐saturated conditions. Gradients of pO2 and pH from the root surface indicated an extent of the rhizosphere effect of 10–20 mm. In contrast, EH gradients were observed from 0 to 2 mm from the root surface. We conclude that the rhizosphere extent differs for various parameters (pH, Eh, pO2) and is strongly dependent on soil moisture. 相似文献

A multivalent in ovo vaccine (MIV) was tested for safety and efficacy in a commercial broiler complex. The MIV comprised five replicating live viruses including serotypes 1, 2, and 3 of Marek's disease virus (MDV), an intermediate infectious bursal disease virus (IBDV) and a recombinant fowl poxvirus (FPV) vector vaccine containing HN and F genes of Newcastle disease virus (NDV). The performance of MIV-vaccinated broilers was compared with that of hatchmates that received turkey herpesvirus (HVT) alone (routinely used in ovo vaccine in the broiler complex). The chickens that hatched from the MIV-injected and HVT-injected eggs were raised under commercial conditions in six barns. Barn 1 housed 17,853 MIV-vaccinated chickens and each of the barns 2-6 housed 18,472-22,798 HVT-vaccinated chickens. The HVT-vaccinated chickens were given infectious bronchitis virus (IBV) and NDV vaccines at hatch and at 2 wk of age. The MIV-vaccinated chickens received IBV vaccine at hatch and IBV + NDV at 2 wk of age. The relative values of hatchability of eggs, livability and weight gain of chickens, and condemnation rates at processing were comparable between the MIV and the HVT groups (P > 0.05). Chickens from the MIV- and the HVT-vaccinated groups were challenged with virulent viruses under laboratory conditions. The resistance of vaccinated chickens against Marek's disease could not be assessed because of high natural resistance of unvaccinated commercial broilers to virulent MDV. The relative resistances of the MIV- and the HVT-vaccinated groups, respectively, against other virulent viruses were as follows: IBDV, 100% for both groups; NDV, 81% vs. 19%; FPV, 86% vs. 0%. The successful use of MIV under field conditions expands the usefulness of the in ovo technology for poultry. 相似文献
