We improved the dry ash procedure for detecting titanium dioxide (TiO2) in cattle feces containing chromium dioxide (Cr2O3). First, the effect of amount of sodium sulfate (Na2SO4) on the recovery of TiO2 from cattle feces that contained Cr2O3 was evaluated. Average recovery of TiO2 at the 2.5 g Na2SO4 level was significantly higher (P < 0.0001) than that at 0.75 g Na2SO4. Second, the effect of Cr2O3 concentration on the recovery of TiO2 of cattle feces by using two levels of Na2SO4 addition was examined. The recovery of TiO2 decreased with the increase in the amount of Cr2O3 at the 0.75 g Na2SO4 level but was consistently high at 2.5 g Na2SO4. Third, the recovery of Cr2O3 from cattle feces was checked. The recoveries of TiO2 and Cr2O3 were high enough at the 2.5 g Na2SO4 level. Fourth, the improved dry ash procedure (5 mL of concentrated H2SO4 and 2.5 g of Na2SO4 were used for sample digestion) was compared to the wet ash procedure. Average recovery of TiO2 by the improved dry ash procedure was significantly higher (P = 0.0077) than that by the wet ash procedure. Thus, the improved dry ash procedure can be used for TiO2 analysis in cattle feces containing Cr2O3.  相似文献

Strips cut from cold storage (5 C), potatoes were immersed in difluorodichloromethane (R-12) from 5 to 13 sec. This rapid freezing was found to selectively damage only the frozen cells and to increase their permeability with respect to intracellular soluble materials. Subsequent immersion in water (17 to 52 C) leached the soluble materials from the frozen cells. This method of leaching in 20 min removed less than 15% of the starch released by potato strips leached at 65 C for 15 min (simulated commercial conditions). The depth of freeze determined by physical methods and by analysis of the leached surface layers showed reducing sugar removal 0.1 to 0.2 mm below the calculated average depth of freeze. In experimental lots the weight of leached materials increased with increased freezing time. Manipulating freezing depth and leaching time made it possible to achieve the required sugar removal from different lots of tubers.  相似文献

Plasma somatolactin (SL) concentrations were examined in chum salmon in relation to gonadal maturation; immature salmon in the Bering Sea at various stages of maturation, and mature salmon during upstream migration caught at the ocean, bay and river. Plasma SL concentrations as well as plasma prolactin (PRL) and growth hormone (GH) levels in the immature fish caught in the Bering Sea were maintained essentially at similar levels. Plasma SL in mature salmon increased significantly from the fish in the ocean to the fish in the river in both sexes. Although all the fish had fully developed gonads, females completed ovulation while still in the bay, whereas final spermeation in males was achieved after entry into the river. Thus, no clear correlation was seen between plasma SL levels and final gonadal maturation. On the other hand, plasma PRL concentrations in both male and female fish were higher in the fish in the river than those in the ocean and bay, and plasma GH levels were higher in both sexes in the fish in the bay and river than those in the ocean. Plasma levels of triglycerides, glucose, free fatty acids and ionized sodium and calcium were also examined. Significant-negative correlations were seen between plasma SL and plasma ionized calcium in mature male salmon, and between plasma SL and plasma triglycerides in mature female salmon. Although our findings do not rule out the possibility of the involvement of SL in final maturation, the results indicate that SL seems to be involved at least in energy and/or calcium metabolism during the spawning migration.  相似文献

ABSTRACT:   The clonal triploid ginbuna crucian carp Carassius auratus langsdorfii , a naturally occurring gynogenetic fish, is a useful model for studying T-cell-mediated immunity. CD4, a T-cell receptor (TCR) coreceptor, is a membrane-bound glycoprotein found on helper T-cells, and assists in the binding of major histocompatibility complexes. In the present study, full-length cDNAs encoding the CD4 molecule from the S3n strain of ginbuna crucian carp were cloned and characterized. 5'-rapid amplification of cDNA ends (RACE) and 3'-RACE yielded two distinct cDNA clones of CD4 homolog from the ginbuna, and these sequences share 95% identity at the amino acid level. These ginbuna CD4 molecules consisted of a signal peptide, immunoglobulin superfamily (IgSf) like domains, a transmembrane domain, and a cytoplasmic domain similar to other known CD4. A tyrosine protein kinase p56lck binding motif is conserved in the cytoplasmic tail of ginbuna CD4. Phylogenetic tree analysis indicated that ginbuna CD4 sequences are closely related to CD4L-1 from other fish species. Expression of ginbuna CD4 mRNA was detected in the gill, thymus, head kidney, trunk kidney and peripheral blood leukocytes, indicating that its expression pattern is similar to that of ginbuna TCRβ mRNA. The results suggest that ginbuna CD4 sequences are useful as molecular probes for helper T-cells.  相似文献
