The cation-exchange capacity (CEC) of the soils in the Colina district is in interrelation neither with the texture nor the organic matter content. This apparent inconsistency is largely attributable to the CEC of the coarse fractions of the soils, which contain appreciable amounts of montmorilionite and zeolite with some vermiculite. The coarse particles containing exchange materials are widely distributed in the soils of central and northern Chile regardless of the soil type, and appear to be rather stable under semi-arid climates.  相似文献

The tiger pufferfish (fugu) is one of the most important food fishes in East Asia. Since its testes are regarded as a delicacy, sex determination is economically relevant. Previous studies have identified a missense single-nucleotide polymorphism (SNP) in the Amhr2 (anti-Müllerian hormone receptor type II) gene as a strong candidate for a master sex-determining polymorphism. To distinguish genotypic sex efficiently, we developed a high-resolution melting (HRM) assay for this SNP site. By screening 396 fish from two independent crosses reared under controlled conditions, we observed perfect concordance between the SNP genotype and phenotypic sex. Thus, this method holds great potential for use in high-throughput sexing. When analyzing 293 progeny from a third cross reared under unknown conditions, we unexpectedly found that 25 % of phenotypic males exhibited female genotype. These results suggest that environmental factors such as rearing conditions could influence the sex-determination pathway in pufferfish. Alternatively, genetic modifiers might override the signals from Amhr2. This finding raises a concern regarding enhanced stock management of this species, because sex-reversed fish could compromise the sex ratio in subsequent generations. The HRM assay will also be useful for monitoring the degree of sex reversal before release.  相似文献

An experiment was conducted to study the association between cold shock treatment (CST) (0 °C, 1 h) and heat shock treatment (HST) (55 °C, 30 s) on antioxidant enzymes of broccoli at different storage temperatures (20, 10, or 5 °C). There was no difference in antioxidant enzyme activities between CST and HST under storage at 20 °C; superoxide dismutase (SOD) and catalase (CAT) activities were increased in comparison with the controls. However, activities of antioxidant enzymes, except for ascorbate peroxidase (APX), with CST were higher than those with HST during storage at 10 °C, while there were opposite results except for CAT activity at 5 °C. These results suggest that CST and HST have the same effect on the antioxidant enzymes of broccoli during 20 °C storage, but have different effects at below 10 °C, which become more pronounced as the temperature is lowered.  相似文献

Laboratory experiments were conducted to investigate (1) the effects of the addition of rice (Oryza sativa. L.) bran to paddy soil on the germination of Monochoria vaginalis (Burm. f.) Kunth, and (2) the relationship between the electric conductivity (EC) of the soil solution and germination. Soil samples were collected at 4 sites in Japan. After flooded soils with rice bran had been incubated for 7?d at 30°C, the soil solution was collected using a porous cup and the EC of the soil solution was measured. The amounts of rice bran added to the soil were 0%, 0.3%, 0.6% and 0.9% (weight(w)/w). In the soil solution obtained, seeds of M. vaginalis were incubated for 3?d at 30°C, and the germination percentage was then analyzed. The addition of rice bran suppressed germination, and the degree of suppression increased with increasing content of rice bran. Although the same amount of rice bran was applied to each soil, the degree of growth suppression by rice bran as well as the EC of the soil solution differed among the soils. In each soil, there was a positive correlation between the amount of rice bran and EC, and the degree of growth suppression significantly increased with an increase in EC. When EC was higher than 150?mS?m?1, seeds of M. vaginalis hardly germinated. There was no significant correlation between the oxidation-reduction potential (Eh) of soil and seed germination, suggesting that EC is a more reliable and convenient indicator than Eh for evaluating the relationship between the addition of organic material and seed germination. In conclusion, the addition of rice bran to soil increases the EC of the soil solution, and EC is one of the factors that suppress the germination of M. vaginalis. The suppressive effect of rice bran on germination is different among soils. This fact is attributed to the difference in EC due to the addition of rice bran. Thus, it is expected that EC can be used as an indicator for determining how much rice bran to add.  相似文献

Complete nucleotide sequences of eight Japanese isolates of Tomato yellow leaf curl virus (TYLCV) were determined and compared with four TYLCV isolates already reported. These isolates separated into three groups – Shizuoka (Sz), Aichi (Ai), Nagasaki (Ng) – and had 99% identities within the groups. Full-length molecules of DNA-A of group Sz consist of 2791nt and those of group Ai contain 2787nt. Both were closely related to TYLCV-Is.M, although those of group Ng had 2793nt and were more closely related to TYLCV-Is. Comparison of common sequences of isolates belonging to groups Sz and Ai had substitutions of 4nt in the intergenic region and nonsynonymous substitutions at open reading frames between the groups. None of the isolates tested had DNA molecules. Agroinfection of four plant species with a DNA-A dimeric infectious clone of TYLCV-SzY, a member of group Sz, resulted in systemic infection. The present study demonstrates the postnatal developmental changes in immunohistochemical localization of alpha-smooth muscle actin (SMA) and vimentin in the bovine testis. In the peritubular myoid cells of seminiferous tubules and the sub-epithelial and stromal cells of straight tubules and the rete testis, alpha-SMA starts appearing at around 4 months of age. Peritubular alpha-SMA attains the continuous mature pattern at around 5 months of age whereas sub-epithelial and stromal alpha-SMA increases with advancing age. Vimentin is localized in the perinuclear zone of Sertoli cells, peritubular and vascular wall cells, a few interstitial cells, and in the basal part of the epithelia of straight and rete tubules. Developmental changes are only evident in the Sertoli cell vimentin, which is basal and weak at birth and increases moderately until 4 months of age. From around 5 to 8 months of age when the Sertoli cells are under morphological transformation, vimentin intensity is considerably increased and the characteristic vimentin extensions connect the Sertoli nuclei to the basal membrane. These extensions get shorter at around 9 month of age as the Sertoli nuclei are positioned basally. The mature Sertoli cell perinuclear vimentin is strong and stable without infranuclear extension. In conclusion, the age of appearance of alpha-SMA coincides with the onset of postnatal division of spermatogonia, and vimentin may play a key role in stabilizing Sertoli cell nuclei during their transformation in bovine.  相似文献

Pyrolysis of cellulose was studied for the purpose of practical production of 1,6-anhydro-β-D-glucopyranoside (levoglucosan, LG). To minimize secondary degradation of levoglucosan, two methods were examined: (1) conductive heating by glass bottle, and (2) radiation heating from the surface by CO2 laser beam, both under vacuum and in a nitrogen atmosphere. Glass-bottle pyrolysis under vacuum gave levoglucosan yield of 50%–55% in the optimum temperature range of 350°–410°C, where placing the cold trap in the vicinity of heated area was effective in improving the yield. In contrast, glass-bottle pyrolysis under nitrogen gave low yields of 17%–20%, probably due to slower diffusion of pyrolysis product from hot region. The CO2 laser pyrolysis under vacuum gave the product as aerosol (white smoke), causing difficulty in recovery of the product, and the maximum yield was 5%–17%. In this case the treatment under nitrogen flow was effective for recovery of aerosol, and the maximum yield reached approximately 25%.  相似文献

Wheat can cause severe IgE-mediated systematic reactions, but knowledge on relevant wheat allergens at the molecular level is scanty. The aim of the present study was to achieve a more detailed and comprehensive characterization of the wheat allergens involved in food allergy to wheat using proteomic strategies, referred to as "allergenomics". Whole flour proteins were separated by two-dimensional gel electrophoresis with isoelectric focusing and lithium dodecyl sulfate-polyacrylamide gel electrophoresis. Then, IgE-binding proteins were detected by immunoblotting with sera of patients with a food allergy to wheat. After tryptic digestion, the peptides of IgE-binding proteins were analyzed by matrix-assisted laser desorption ionization tandem time-of-flight mass spectrometry. In this study, we identified four previously reported wheat allergens or their sequentially homologous proteins [serpin, alpha-amylase inhibitor, gamma-gliadin, and low molecular weight (LMW) glutenin] by a database search. As a result of the high resolution of two-dimensional gel electrophoresis, nine subunits of LMW glutenins were identified as the most predominant IgE-binding antigens. The two-dimensional allergen map can be beneficial in many ways. It could be used, for example, for precise diagnosis of wheat-allergic patients and assessment of wheat allergens in food. Additionally, we compared allergenomics to conventional biochemical methods and evaluated the usefulness of a proteomic strategy for identifying putative allergens to wheat allergy.  相似文献

Propolis is a resinous substance collected by honeybees from various plant sources. The composition of propolis depends upon the vegetation at the site of collection. We previously isolated four prenylated flavonoids from propolis collected in Okinawa, Japan. In this study, further fractionation of the extracts of Okinawan propolis resulted in the isolation of a new prenylated flavonoid, prokinawan, and four known compounds. The structure of prokinawan was determined by MS and NMR spectroscopic methods. Furthermore, the antioxidant activity using 1,1-diphenyl-2-picryl-hydrazyl radical scavenging and beta-carotene bleaching systems was investigated. The present study proved that the position of the geranyl or prenyl groups on the flavonoid skeleton plays an important role in exhibiting antioxidant activity.  相似文献

The effects of microhabitat diversity and geographical isolation on the structure of oribatid communities were studied in mangrove forests on the Ryukyu Islands of Japan. The study took place at three sites on two islands 470 km apart. Oribatid mites (Oribatida) were extracted from leaves, branches, bark of trunks (0-50, 50-100, and 100-150 cm high) and of knee roots, and from forest-floor soil and littoral algae, each defined as a microhabitat of oribatid mites. At the 0-50 cm height, the species composition of the oribatid communities on the knee-root bark and the bark of trunks of Bruguiera gymnorrhiza differed significantly from that on the other microhabitats. This difference was attributed to tidal flooding of the mangrove forests. Cluster analysis showed that oribatid communities in the same microhabitat at different sites tended to be more similar than those on different microhabitats at the same site. This implies that the species composition of oribatid communities in mangrove forests is more likely to be affected by factors responsible for microhabitat diversity (characterized specifically by the flooded trunks) than by geographical distance between the islands.  相似文献
