BACKGROUND: Wild radish, a problem weed worldwide, is a severe dicotyledonous weed in crops. In Australia, sustained reliance on ALS‐inhibiting herbicides to control this species has led to the evolution of many resistant populations endowed by any of several ALS mutations. The molecular basis of ALS‐inhibiting herbicide resistance in a novel resistant population was studied. RESULTS: ALS gene sequencing revealed a previously unreported substitution of Tyr for Ala at amino acid position 122 in resistant individuals of a wild radish population (WARR30). A purified subpopulation individually homozygous for the Ala‐122‐Tyr mutation was generated and characterised in terms of its response to the different chemical classes of ALS‐inhibiting herbicides. Whole‐plant dose‐response studies showed that the purified subpopulation was highly resistant to chlorsulfuron, metosulam and imazamox, with LD50 or GR50 R/S ratio of > 1024, > 512 and > 137 respectively. The resistance to imazypyr was found to be relatively moderate (but still substantial), with LD50 and GR50 R/S ratios of > 16 and > 7.8 respectively. In vitro ALS activity assays showed that Ala‐122‐Tyr ALS was highly resistant to all tested ALS‐inhibiting herbicides. CONCLUSION: The molecular basis of ALS‐inhibiting herbicide resistance in wild radish population WARR30 was identified to be due to an Ala‐122‐Tyr mutation in the ALS gene. This is the first report of an amino acid substitution at Ala‐122 in the plant ALS that confers high‐level and broad‐spectrum resistance to ALS‐inhibiting herbicides, a remarkable contrast to the known mutation Ala‐122‐Thr endowing resistance to imidazolinone herbicide. The Channel Catfish Ictalurus punctatus is an important ecological, recreational and aquaculture species. In both natural and pond culture systems, Channel Catfish are periodically exposed to acute increase in temperature, especially in the summer, which may induce a stress response. With the effects of global climate change, the impact of rising temperatures on aquatic systems is expected to be exacerbated in the future. Hence, there is the need to understand the physiological mechanisms controlling the thermal stress response in fish. Acid–base, haematological, osmotic and metabolic regulations were investigated following acute temperature increase from 28°C to either 32°C or 36°C at a rate of 2°C/hr to assess the physiological response of Channel Catfish to temperature increase. Blood was sampled 1 hr after treatment temperatures were reached. When temperature increased from 28 to 32°C, blood bicarbonate () decreased while cortisol, partial pressure of carbon dioxide (pCO2) and glucose increased. When temperature increased from 28 to 36°C, blood pH, , chloride, total proteins and mean corpuscular haemoglobin concentration decreased while cortisol, pCO2, haematocrit, haemoglobin, glucose, lactate, mean corpuscular volume and mean corpuscular haemoglobin all increased. The changes observed are characteristic of acute stress responses in fish and suggest that acute temperature increases to both 32 and 36°C are physiologically stressful to Channel Catfish.  相似文献

Objective: To review the current understanding of mechanisms involved in normal hemostasis and to describe the changes associated with pro‐inflammatory disease processes such as sepsis. Data sources: Original research articles and scientific reviews. Human data synthesis: Organ damage caused by sepsis is created in part by the interdependent relationship between hemostasis and inflammation. Markers of coagulation have been found to have prognostic value in human patients with sepsis and there are both experimental and clinical investigations of the therapeutic potential of modulating the hemostatic system in sepsis. Improvement of 28‐day all‐cause mortality in severe sepsis by treatment with recombinant human activated Protein C strongly supports the interdependence of hemostasis and inflammation in the pathophysiology of sepsis. Veterinary data synthesis: Publications reporting clinical evaluation of the hemostatic changes occurring in septic dogs or cats are minimal. Experimental animal models of sepsis reveal significant similarity between human and animal sepsis and may provide relevance to clinical veterinary medicine until prospective clinical evaluations are published. Conclusions: It is now apparent that inflammation and the coagulation system are intimately connected. Understanding this relationship provides some insight into the pathogenesis of the hemostatic changes associated with sepsis. This new updated view of hemostasis may lead to the development of novel therapeutic approaches to sepsis and disseminated intravascular coagulation in veterinary medicine.  相似文献

Profitability of a beef operation is determined by the proportion of cows attaining pregnancy early in the breeding season and those that are pregnant at the end of breeding season. Many factors, including temperament, contribute to those reproductive parameters. The objective of this study was to evaluate effects of temperament on reproductive performance of beef cows. In Experiment 1, Angus and Angus‐cross beef cows (n = 1546) from eight locations were assigned a body condition score (BCS; 1 = emaciated; 9 = obese) and chute exit and gait score (1 = slow exit, walk; calm temperament; 2 = jump, trot or run; excitable temperament). Cows were grouped with bulls (1 : 25 to 1 : 30; with satisfactory breeding potential and free of venereal disease) for an 85‐day breeding season. Pregnancy status and stage of gestation were determined (transrectal palpation) 35 days after the end of the breeding season. Controlling for BCS (p < 0.01) and handling facility (p < 0.0001) and handling facility by temperament score interaction (p < 0.001), breeding season pregnancy rate was lower in excited versus calm cows [88.6% (798/901) vs 94.1% (607/645); p < 0.001]. Cows with an excitable temperament took 24 more days to become pregnant compared to calm cows (median days to pregnancy, 35 vs 59 days; p < 0.0001). In Experiment 2, Angus and Angus‐cross beef cows (n = 1407) from 8 locations were assigned scores for body condition and chute exit and gait (as described in Experiment 1) and assigned to bulls (breeding sound and free of venereal disease; 1 : 25 to 1 : 30) for 85 days. Pregnancy status was determined by transrectal palpation at 2 and 6 months after the onset of the breeding season. Controlling for BCS (p < 0.05), pregnancy loss was higher in excited versus calm cows [5.5% (36/651) vs 3.2% (20/623), p < 0.0001]. In conclusion, beef cows with an excitable temperament had significantly lower reproductive performance than calmer cows. The modified two‐point chute exit–gait scoring method was repeatable and identified cattle with an excitable temperament.  相似文献

Prevalence and pathogenicity of Aeromonas hydrophila   总被引:2,自引:0,他引:2  
A field survey to determine the prevalence of Aeromonas hydrophila revealed a recovery rate of 8% in 141 specimens derived from a range of live, companion, and exotic avian species. The prevalence rate was similar in 240 sequential postmortem submissions during 1984. Studies on the pathogenicity of A. hydrophila showed that 2-to-4-day-old chicks and turkey poults were highly susceptible to exposure via the subcutaneous, yolk-sac, and intracerebral routes, and mortality of 80-100% occurred within 48 hours of inoculation. Ducklings aged 4 days were generally refractory to exposure.  相似文献
