Unidentified olive plants naturally grow in the Golestan province of Iran, on different soils and under climates spanning from sub-temperate to desert conditions, represented by single trees or groups of few trees. We collected samples from representative sites and analyzed them by simple sequence repeat markers in order to determine their identity and their relationships to prominent Iranian and Mediterranean reference cultivars. Population structure analysis separated these ecotypes from Mediterranean and, surprisingly, from all Iranian cultivars, the parentage test excluded their direct contribution as candidate parents or offspring of cultivars, and they also showed a high level of admixture. Their differentiation from cultivated olives may be attributed to different factors: they could represent wild plants or could derive from natural dissemination of ancestral cultivated trees. Their survival up to now may be due to the fact that most of them are grown on sacred sites such as necropolis. Anyhow, the adaptation to strong environmental stresses, and their fruit size and oil content make the olive Golestan ecotypes a valuable source of genetic variation previously uncharacterized and currently threatened with extinction.  相似文献

Purpose

Sustainable management of riparian zone soils is required to ensure the health of natural ecosystems and maintenance of soil nitrogen (N) pools and soil N cycling. However, the effect of revegetation type and age on soil N pools remains poorly understood.

Materials and methods

This study compiled data from published articles to understand the effects of revegetation types and age on soil total N (TN) and soil inorganic N (NH4+-N, and NO3?-N) using a meta-analysis. We extracted 645 observations from 52 published scientific articles.

Results and discussion

The revegetation of riparian zones led to a significant increase of soil TN (mean effect size: 11.5%; 95% CI: 3.1% and 20.6%). Woodland increased soil TN significantly by 14.0%, which was associated with the presence of N fixing species and high litter inputs. Soil NH4+-N concentration significantly increased (mean effect size: 20.1%; 95% CI: 15.1% and 25.4%), whereas a significant decrease in soil NO3?-N (mean effect size: ? 21.5%; 95% CI: ? 15.0% and ? 27.5%) was observed. Of the revegetation types considered in this paper, NO3?-N concentration in soil followed the order: grassland < shrubland < woodland, suggesting that woodland might be more efficient in soil NO3?-N retention than grassland. The high plant N uptake and accelerated NO3?-N leaching in grassland could be related to the decreased soil NO3?-N in grassland compared with other revegetation types. Revegetation significantly decreased soil moisture by (mean effect size: ? 7.9%; 95% CI: ? 3.3% and ? 12.2%) compared with the control, which might be associated with the selection of exotic species as dominant vegetation in the riparian zone. Soil TN increased in revegetation ages between 10 and 40 years following revegetation and was related to increased soil organic carbon inputs within those ages following the establishment.

Conclusions

This study provides insight into influence of different vegetation types and age on soil N pools and soil moisture. This study also highlights the importance of revegetation in riparian zones to increase soil TN.

Background: One of the most widely used anticancer agents is microbial L-ASNase. Herein, we assessed the biochemical and biological properties of an isolated L-ASNase from a Gram-negative bacteria strain, Escherichia coli MF-107. Methods:Using garden asparagus, we obtained several bacterial isolates. These strains were further screened for L-ASNase activity. A promising bacterial isolate was selected for L-ASNase production and subsequent purification. The molecular weight of purified L-ASNase was determined. The MTT assay was applied to assess the cytotoxic effect of the purified enzyme. Also, for caspase activity determination and the apoptotic effect of purified enzyme on in cells, we conducted a real-time PCR method. Results:The molecular weight of the enzyme was approximately 37 kDa. In the pH range of 7.5 to 8, the enzyme had considerable stability. At 35 °C, the purified L-ASNase optimum activity was recorded. The cytotoxic effect of the enzyme on treated cells was dose-dependent with an IC50 value of 5.7 IU/ml. The Bax gene expression considerably raised by 5.75-fold (p < 0.001) upon L-ASNase treatment. On the other hand, the anti-apoptotic Bcl-2 gene expression showed a 2.63-fold increase compared to the control (p < 0.05). It was detected that the mRNA levels of caspase-3 and p53 were considerably upregulated (5.93 and 1.85-fold, respectively). We did not find any alternation in the caspase-8 activity of the treated cells compared to untreated cells. Conclusion:In this research, the proliferation of the breast cancer cells remarkably inhibited via the cytotoxic effect of isolated L-ASNase from microbial sources.Key Words: Apoptosis, Breast cancer, Escherichia coli, MCF7 cell line  相似文献

Studies were conducted into effects of Rift Valley fever virus on oestrous and vaginal cytology in non-pregnant ewes. 6 pregnant and 8 non-pregnant local Barki ewes were subcutaneously inoculated with different doses of virus isolated from Sharkia Province. Observed were irregularities in the oestrous cycle (20-28 days inoculation) and anoestrum. The anoestrum smears included large non-cornified epithelial cells with large well stains centrally located nuclei together with a large number of bacteria. Abortion and retention of placenta occurred in pregnant ewes with prolonged puerperal heat up to 70 days. Inoculation of virus was followed by thermal response. Re-isolation of virus in BHK cells was possible from aborted foetuses and placenta.  相似文献

Chicory (Cichorium spp.) is a valuable vegetable crop worldwide for its edible leaves and for the production of coffee substitutes from roots. Agrobacterium rhizogenes-mediated genetic transformation of two species of chicory (C. intybus and C. endivia) was investigated using Agrobacterium strain K599 harbouring p35SGFPGUS+ plasmid and two types of explants: leave and leaf stalks. This Agrobacterium strain proved to be competent in the transformation with transformation rate about (23.1%) in leaf explants of C. intybus. However, the transformation rate with C. endivia was much lower (3.6%). Moreover, the hairy roots appeared from different infection sites of the leaf explants. Several hairy roots of the two species were acquired, out of them 11 clones (C. intybus) and two clones (C. endivia) were selected due to their fast-growing character. Growth of hairy roots was determined on the basis of total root biomass accumulation. It was found that the liquid MS-basal medium seems to be the most suitable for biomass production. PCR analysis revealed foreign DNA integration in the selected transgenic hairy root clones. Notable, the transgenic hairy roots exhibited substantially higher growth rates and accumulated higher amount of inulin than non transgenic roots (WT). Preservation of elite key lime (Citrus aurantifolia) genotypes using efficient micropropagation and regeneration protocols is very important for new breeding programmes. The objective of the present study was to develop an efficient protocol for micropropagation and maintenance of elite genotypes of key lime based on slow growth and starvation strategy. To do this, regenerated shoots were starved, and the growth was compromised via avoiding subcultures for 4, 8, 12 and 24 months. The numbers of responsive explants and adventitious shoots per each explant were evaluated in the presence of benzylaminopurine (BAP). The results showed that the 12- and 24-month starved explants treated with 1 mg/l BAP were able to produce four times more shoots than the control. In addition, the influences of horizontal or vertical layering explants over medium were evaluated. The results indicated that all horizontally layered explants significantly produced 25% adventitious shoots more than vertical explants, most likely owing to more contact with the surface medium. Finally, it could be concluded that application of slow growth, starvation strategy and also horizontally layering of the explants could efficiently enhance micropropagation and maintenance of threatened elite key lime genotypes with a low cost.  相似文献

ABSTRACT

1. The use of chitosan nanoparticles (CNPs) for the entrapment of active components has gained considerable interest in food and pharmaceutical industries due to its mucous adhesiveness, non-toxicity, biocompatibility and biodegradability.

2. This study aimed to investigate the usefulness of feeding encapsulated thyme essential oil (E-TEO) on performance, blood parameters, immunity and ileal microflora in broilers.

3. CNPs were prepared by using ionotropic gelation and structurally characterised by dynamic light scattering; scanning electron microscope and Fourier transform infrared spectroscopy. The encapsulation efficiency percentage of TEO in CNPs and its in vitro release were determined using ultraviolet visible spectroscopy and dialysis method, respectively.

4. An in vivo experiment was conducted with five treatments of five replicates with ten chicks per pen. Dietary treatments consisted of an unsupplemented control diet, or supplemented with chlortetracycline (80 and 50 mg/kg during the starter and grower phase, respectively); CNPs (60 mg/kg); TEO (40 mg/kg diet) or E-TEO (60 mg/kg).

5. Results showed uniform distribution of nanoparticles with roughly spherical morphology, and the size of particles ranged from 30 to 100 nm. It was observed that 68.3% of TEO was encapsulated into CNPs, from which more than 90% was released within the first 96 h. Birds treated with antibiotic and E-TEO showed significantly (P < 0.05) higher body weight gain than the control and the other treatments were intermediate. Compared to the control group, adding of E-TEO into diet significantly (P < 0.05) improved feed conversion ratio at 42 days of age. Feeding birds with E-TEO significantly (P < 0.05) decreased the number of coliform and total aerobic bacteria compared to other treatments. Birds supplemented with E-TEO showed lower heterophile to lymphocyte ratio compared to birds that received TEO.

6. The results suggested that the entrapment of TEO in CNPs and its sustained release promoted broiler performance via the improvement of physiological status and modulating intestinal microbiota and therefore could be used as substitute for antibiotic growth promoter in broiler diets.  相似文献

The role of dietary lemon peel (LM) and/or Bacillus licheniformis (BL) on the growth, immunity, and resistance against Aeromonas hydrophila in common carp, Cyprinus carpio was investigated in this study. LM and BL were included in diets at 0% (T0), 108 CFU/g BL (T1), 1.5% LM and 108 CFU/g BL (T2), and 3% LM and 108 CFU/g BL (T3). Fish fed with T1, T2, or T3 had higher weight gain, specific growth rate, white blood cells count, and blood total protein with lower feed conversion ratio than T0 group (p < .05). The albumin increased significantly (p < .05) in fish fed both BL and LM (T3). The serum superoxide dismutase (SOD), catalase, lysozyme, and bactericidal activities were significantly increased in fish fed both BL and LM (T2 and T3), while serum glutathione peroxidase increased in fish fed BL (T2) (p < .05). Fish fed T1, T2, and T3 diets displayed higher SOD and lower malondialdehyde than fish fed T0 (p < .05). After the A. hydrophila challenge, the mortality rate was significantly lower in T1, T2, and T3 groups than the T0 group (p < .05). The obtained results revealed that LM and BL could be used to increase resistance against A. hydrophila infection in carp. However, further field studies should be performed to confirm the obtained results.  相似文献
