Chlorophyll (Chl) and nitrogen (N) status of leaves provide valuable information about the physiological condition of plants. The conventional methods for measuring Chl and N contents in leaves are destructive, costly, time-consuming, and do not allow repetitive measurement of the same sample. The Damask Rose (Rosa damascena Mill) is an important aromatic crop in the western Himalaya region in India. Generally, flower yield and oil yield of the Damask rose are correlated with nitrogen, phosphorus, and potassium (NPK) levels in the leaf at the bud development stage. The dynamics of N within the rose plant have not been reported clearly. Thus, there is a pressing need for non-destructive techniques to estimate Chl and N content in the leaf of the Damask rose. Our objective was to establish an appropriate mathematical relationship between the Chl content index (CCI) value and the total Chl/N contents for non-destructive estimation of total Chl and N in the leaf of the Damask rose. The regression models were developed with destructively measured parameters (total Chl and N) as the dependent variable and a parameter derived from CCM-200 as the independent variable (CCI). We found that polynomial regression models are suitable for non-destructive estimation of total Chl, and the model predicted values were very close to traditionally measured values with a root mean square prediction error (RMSEp) less than 0.20?mg?g?1 of Chl. In the case of N estimation, a power regression model was appropriate with lowest Akaike's information criteria (AIC) and root mean square validated error (RMSEv) value. Significant correlations (P?≤?0.001) were observed between traditionally measured values and our model predicted values in both cases.  相似文献

Background: Brucella abortus, the major causative agent of abortion in cattle and a zoonotic pathogen, needs to be diagnosed at an early stage. Loop-mediated isothermal amplification (LAMP) test is easy to perform and also promising to be adapted at field level.

Objective: To develop a LAMP assay for specific and rapid detection of B. abortus from clinical samples of cattle.

Methods: LAMP primers were designed targeting BruAb2_0168 region using specific software tool and LAMP was optimized. The developed LAMP was tested for its specificity with 3 Brucella spp. and 11 other non-Brucella spp. Sensitivity of the developed LAMP was also carried out with known quantity of DNA. Cattle whole blood samples and aborted fetal stomach contents were collected and used for testing with developed LAMP assay and results were compared with polymerase chain reaction (PCR).

Results: The developed LAMP assay works at 61 °C for 60 min and the detection limit was observed to be 100-fold more than the conventional PCR that is commonly used for diagnosis of B. abortus. Clinical sensitivity and specificity of the developed LAMP assay was 100% when compared with Rose Bengal plate test and standard tube agglutination test. SYB® green dye I was used to visualize the result with naked eye.

Conclusion: The novelty of the developed LAMP assay for specifically detecting B. abortus infection in cattle along with its inherent rapidness and high sensitivity can be employed for detecting this economically important pathogen of cattle at field level as well be exploited for screening of human infections.  相似文献

The objectives of the present investigations are to see the effects of biomass power plant effluent on the carbohydrates, aminoacids, nitrite and nitrite enzyme activitiesand proline of Arachis hypogaea L. var TCGS 320 under controlled pot culture methods. Plants were cultivated with 25, 50, 75 and 100% of the effluent and a control without the effluent. The treatment of the crop with 25% of the effluent has shown stimulatory effect on all the biochemical parameters studied. Carbohydrates, starch, aminoacids, protein, nitrate and nitrite reductase enzymatic activities have increased in 10, 15, 20 DAS (days after sowing). In 25 and 30 DAS all biochemical parameters have decreased, due to environmental factors (rainfall, temperature, humidity, etc.). The present investigation clearly indicated that the biomass power plant effluent has stimulatory effect on all the biochemical contents at lower concentration, and at higher concentration they have deleterious effects.  相似文献

BACKGROUND: Diamondback moth, Plutella xylostella (L.), is a major insect pest of crucifers in the biodiversity‐rich north‐western Indian Himalayan hills. The present investigation was aimed at determining the susceptibility pattern of P. xylostella populations collected from different locations of this region to autochthonous and standard Bacillus thuringiensis strains. RESULTS: Among the reference as well as indigenous B. thuringiensis strains tested, sub spp. kurstaki HD‐1, kurstaki HD‐73, galleriae HD‐8, local galleriae/colmeri strain BtOa1 and some of their Cry1 class toxins were found to be highly toxic. Surprisingly, the sub sp. tolworthi HD‐125, local tolworthi strain BtHa1 and Cry9 class toxins were found to be non‐toxic. Midgut homogenate from fourth‐instar larvae was found to activate 130 kDa protoxin from the local tolworthi strain BtHa1 into 68 kDa toxin, but failed to exert any larval mortality, probably owing to lack of receptor binding. CONCLUSION: The present study provides valuable baseline susceptibility data for the deployment of B. thuringiensis‐based control methods, as well as for future monitoring of development of resistance in P. xylostella to B. thuringiensis in this ecologically sensitive region. A study was undertaken to evaluate the nutritional response of organically grown diets in buffaloes on nutrients utilization and nutrient efficiency for milk production. For this, ten milch Murrah buffaloes of average body weight (490.72?±?6.65 kg), milk yield (8.13?±?0.33 kg), and lactation stage (85?±?5.28 days) were distributed in inorganic (InDg) and organic dietary groups (OrDg) with five animals in each. Buffaloes of InDg and OgDg were fed sorghum hay–berseem fodder-concentrate mixture-based diets grown inorganically and organically, respectively. After 60 days of feeding, a digestion cum metabolism trial was conducted to assess feed intake, nutrient utilization, and N balance, while milk yield and composition were recorded fortnightly. DM, CP, digestible crude protein (DCP), and metabolizable energy (ME) intake (g/Kg w0.75) were similar in animals of InDg and OrDg. NDF, ADF, and hemicellulose digestibility were (P?<?0.05) higher in animals on OrDg (59.20, 51.55, and 62.67) than InDg diet (54.57, 43.72, and 56.61 %), respectively. Urinary n loss (g/day) was (P?<?0.05) lower in OrDg (67.23) than in InDg (83.55); however, milk N was comparable in animals of both dietary groups (47.36 vs 45.82 g/day). Nitrogen balance was higher in animals of OrDg (39.72) than in InDg (28.08). DCP, TDN, and ME values of both diets were similar. No effect of diet was observed on milk yield and its composition; however, increased lactation length decreased milk yield. Buffaloes on both diets had similar efficiency and conversion ratio of nutrients for milk production. Results revealed that diets (organically vs. inorganically grown) have no effect on milk yield and its composition; however, buffaloes on organic diet have higher fiber digestibility and low urinary N loss which did not affect the dietary nutrients efficiency and conversion ratio for milk production.  相似文献

Simulation studies have demonstrated that limited maximum transpiration rate (TR) at high air vapor pressure deficit (VPD) in water-limited environments could result in significant increases in sorghum yield. However, such a restriction on TR at high VPD has not been documented in sorghum. The objective of this study was to search within sorghum germplasm for the possibility of restricted TR at high VPD. Twenty six genotypes were selected for measurement of VPD response based on field observations including yield, leaf temperature, and the stay-green phenotype. These genotypes were grown in a greenhouse for about 24-d growth, and then placed into individual chambers in which VPD was varied and TR measured. The results of this study showed marked variation among sorghum genotypes in TR response to VPD. Seventeen genotypes were identified as exhibiting a breakpoint in their VPD response in the range from 1.6 to 2.7 kPa, above which there was little or no further increase in TR. Therefore, these genotypes with a breakpoint have the possibility of soil water conservation when VPD during the midday cycle exceeds the breakpoint VPD. This trait would be desirable in less humid environments for increasing yields in water-deficit seasons. The observed range in the value of the BP among genotypes offers the possibility of developing genotypes with BP appropriate for specific environments.  相似文献
