Twelve killer whale (Orcinus orca) were hemmed in by ice floes, and nine died on the Aidomari coast in the Nemuro Strait in Rausu, Shiretoko, Hokkaido, Japan on 8 February 2005. Tissue samples collected from 8 whales were tested for Neospora caninum, Toxoplasma gondii, and Brucella species DNA by polymerase chain reaction (PCR) assay. Gamma-globulin isolated from blood samples by ammonium sulfate precipitation was tested for antibodies to these pathogens by means of agglutination tests and immunoblotting. None of the 8 tissue samples had antibodies to the pathogens, when subjected to agglutination tests. In immunoblotting, one sample (sample No.5) showed antibody binding to N. caninum antigens. In the PCR assay, none of the samples was positive. Further study is necessary to examine the prevalence of the pathogens in marine mammals inhabiting this area.  相似文献

Human basophilic KU812 cells express the high-affinity IgE receptor Fc epsilon RI, which plays a central role in the IgE-mediated allergic response. The effect of several major tea catechins, (+)-catechin, (-)-epicatechin, (-)-epigallocatechin, (-)-epicatechin gallate, and (-)-epigallocatechin gallate (EGCg), on the cell surface expression of Fc epsilon RI in KU812 cells was studied. Flow cytometric analysis showed that only EGCg was able to decrease the cell surface expression of Fc epsilon RI after a 24 h treatment in a dose-dependent manner. Moreover, immunoblot analysis revealed that the total cellular expression of the Fc epsilon RI alpha chain decreased upon treatment with EGCg. Fc epsilon RI is a tetrameric structure comprising one alpha chain, one beta chain, and two gamma chains. The level of mRNA production of each subunit in KU812 cells was investigated. KU812 cells treated with EGCg expressed lower levels of Fc epsilon RI alpha and gamma mRNA than nontreated cells. These results suggest that EGCg has an ability to down-regulate Fc epsilon RI expression, and this suppressive effect may be due to the down-regulation of Fc epsilon RI alpha and gamma mRNA levels.  相似文献

We examined the effect of flavones on the expression of the high-affinity IgE receptor FcepsilonRI, which plays a central role in the IgE-mediated allergic response. Flow cytometric analysis showed that the flavones chrysin and apigenin were able to reduce the cell surface expression of FcepsilonRI in human basophilic KU812 cells. Immunoblot analysis revealed that the total cellular expression of the FcepsilonRI alpha and gamma chains decreased upon treatment with chrysin and apigenin. Moreover, the level of mRNA expression of the FcepsilonRI alpha and gamma chains also decreased when the cells were cultured with the two flavones. Previously, we demonstrated that the reduction of extracellular signal-regulated kinase1/2 (ERK1/2) phosphorylation was involved in the downregulation of FcepsilonRI expression. The two flavones were shown to reduce the level of ERK1/2 phosphorylation. These results suggested that chrysin and apigenin have the ability to downregulate FcepsilonRI expression and this suppressive effect may be due to the reduction of ERK1/2 phosphorylation.  相似文献

Antifungal activities of seven compounds, taxinine (1), paclitaxel (2), phenylisoserine methyl ester (3), sciadopitysin (4), ginkgetin (5), isorhamnetin (6), and quercetin (7), isolated from the leaves of kyaraboku, Taxus cuspidata var. nana, against five plant pathogenic fungi, Gibberella fujikuroi, Cladosporium cucumeninum, Fusarium oxysporum, Colletotrichum fragariae, and Corynespora cassiicola, were investigated for utilization of extractives from trees of the genus Taxus. Also, the amounts of compounds 2 and 3 on the leaf surface was measured in relation to the antifungal activities of compounds. Taxinine (1) showed antifungal activity against G. fujikuroi, C. cucumeninum, F. oxysporum, and C. cassiicola. The minimum inhibitory concentration of taxinine for the four fungi was 0.4mol. In addition, from the results of antifungal tests, it may be concluded that paclitaxel on the leaves and stem of T. cuspidata var. nana does not play an important role as an antifungicide in the resistance of trees to plant pathogenic fungal attack.  相似文献

Seen worldwide, many restrictive policies on timber harvests or log exports have been enforced in the major timber-exporting countries in the last two and half decades. This study aims to assess the impacts of the log export restrictions in the Southeast Asian countries on the Japanese plywood market, whose supply mainly comes from Southeast Asia and the domestic plywood industries. Both plywood industries process tropical raw logs. This econometric analysis reveals the followings; the log export restriction of Indonesia, along with its policy to promote the plywood industry, increased plywood exports to Japan, leaving the amount of log supply rather unchanged. Sabah’s log export restriction and Sarawak’s log export cutback policies decreased the amount of log exports from Southeast Asia to Japan significantly. Furthermore, the producer surplus of Southeast Asian plywood industries increased notably after the log export restriction of Indonesia. The consumer surplus in the Japanese plywood market increased accordingly. On the other hand, the producer surplus of the Japanese plywood industry decreased significantly as a result of Sabah’s log export restriction and Sarawak’s log cutback policies. Consequently, these log export restrictions led to large profits for the plywood industries of Southeast Asia, and that, enforced costs were levied on Japanese plywood industry. To understand effects of Bisphenol-A (BPA) exposure on the reproductive organ across generations, we analyzed morphology of the uterus and ovary, and the methylation pattern of HOXA10 gene of the 2(nd) generation. Pregnant mice (F0) were treated with sc injection of BPA in sesame oil at various doses of 0-1,000 mg/kg Bwt on days 12-16 of gestation. Their offspring (F1) were bred by foster mice, and the offspring (F2) from F1 mice were prepared. That is, F1 mice experienced in utero BPA exposure during the developmental period of reproductive organs, while F2 mice did not at all. Using these F2 mice, the present study was carried out. Comparing to the control, the body weights in BPA exposure groups were significantly increased. Correlating with the increase of body weight, the relative weights of the ovary and uterus in each group were decreased. The histological analysis revealed expansion or emphraxis of the uterine lumen and partial loss of the uterine epithelium. Unmethylation of HOXA10 gene in the uterus was observed in the intron region. The present study suggested that BPA exposure to F0 mice could affect reproductive organ of F2 mice who were not exposed to BPA.  相似文献

ABSTRACT:   The effect of storage temperature (0, 5, 10, and 15°C) and killing procedure (instant, struggled, temperature shocked, and spinal cord destruction killing) on post-mortem changes in the muscle of the horse mackerel caught near Nagasaki, Japan, was investigated. Temporal changes in adenosine triphosphate (ATP), inosine monophosphate (IMP), and lactic acid concentrations were slowest at 10°C storage temperature. The increase in K -values was slower at 10°C and below than at 15°C storage temperature. In addition, 10°C storage temperature was most suitable for maintaining a constant breaking strength value of the muscle. Regarding the effect of killing procedure, temporal changes in ATP, IMP, and lactic acid concentrations were slowest when killed by spinal cord destruction. The increase in K -value and the decrease in breaking strength were also slowest in the spinal cord destruction group. From these results, it was considered that storage at 10°C temperature and spinal cord destruction killing procedure were most effective in delaying post-mortem change in the horse mackerel.  相似文献

A female Japanese Black cow showed hemorrhage under the mucous membrane of the eye conjunctiva and the vagina, and thrombocytopenia in peripheral blood. The clinical manifestations repeated spontaneously during long-term observation. The clinical findings were stable except for severe periodic bleeding. Prothrombin time, activated partial thromboplastin time and fibrinogen levels were normal. The anti-platelet antibody in the serum was analyzed by indirect platelet coagulation tests. Because the platelets in the micro-plate wells of the subject series showed coagulation up to 1:128, it was judged that anti-platelet antibody was present in the serum. A number of megakaryocytes were recognized in smear samples obtained from bone marrow of the scapula immediately after euthanasia. Therefore, this case was diagnosed as idiopathic thrombocytopenia.  相似文献

The spread of morning glory (Ipomoea spp.) in soybean fields in Japan has severely decreased soybean yield. Yet, current control measures do not control the proliferation of Ipomoea spp. As little is known about the flowering period and seed production among the different invading Ipomoea spp., it is challenging to create targeted control measures based on ecological characteristics. This study aimed to reveal the characteristics of the flowering phenology and seed production of four morning glory species, namely, Ipomoea coccinea L. (red morning glory), Ipomoea lacunosa L. (pitted morning glory), Ipomoea hederacea L. Jacq. var. integriuscula A. Gray (entireleaf morning glory), and Ipomoea triloba L. (three-lobe morning glory). Between 2017 and 2019, the four selected study species were grown under similar conditions of soil quality, irrigation, and environmental influences and their flowering phenology and seed data were recorded. The flowering period ranged from 36 to 40 days, and the initial flowering of I. triloba was approximately 2 weeks later than the others. I. coccinea had the highest flowering number and seed production, followed by I. lacunosa, I. triloba, and I. hederacea var. integriuscula. The fruit setting rate of I. triloba decreased later in the reproductive stage but tended to increase as the daily mean temperature increased on each flowering day. Thus, we report that the flowering phenology and seed production differed greatly among the Ipomoea spp. These findings can provide crucial insights into designing targeted species-specific control measures against the spread of Ipomoea spp. in Japan.  相似文献

1. The purpose of the present study was to determine if central interleukin-1β (IL1β), interleukin-6 (IL6) and interleukin-8 (IL8) affect feeding behaviour in chicks (Gallus gallus) and examine if central interleukins are related to the lipopolysaccharide (LPS)-induced anorexia.

2. Intra-abdominal (IA) injection of LPS significantly suppressed feeding behaviour and significantly increased mRNA expression of IL1β and IL8 in the diencephalon when compared to the control group, while IL6 tended to be increased.

3. Intracerebroventricular (ICV) injection of 200 ng IL1β significantly decreased food intake at 60 min after the injection while IL6 and IL8 had no effect.

4. IA injection of these ILs (200 ng) had no effect on food intake in chicks.

5. ICV injection of 200 ng IL1β did not affect water intake and plasma corticosterone concentration, suggesting that central IL1β might not be related to the regulation of drinking behaviour and the hypothalamus–pituitary–adrenal axis.

6. The present study demonstrated that central IL1β but not IL6 and IL8 might be related to the inhibition of feeding in chicks.  相似文献
