Protein microarrays provide a powerful tool for the study of protein function. However, they are not widely used, in part because of the challenges in producing proteins to spot on the arrays. We generated protein microarrays by printing complementary DNAs onto glass slides and then translating target proteins with mammalian reticulocyte lysate. Epitope tags fused to the proteins allowed them to be immobilized in situ. This obviated the need to purify proteins, avoided protein stability problems during storage, and captured sufficient protein for functional studies. We used the technology to map pairwise interactions among 29 human DNA replication initiation proteins, recapitulate the regulation of Cdt1 binding to select replication proteins, and map its geminin-binding domain. 相似文献

We report the detection of a planet whose orbit surrounds a pair of low-mass stars. Data from the Kepler spacecraft reveal transits of the planet across both stars, in addition to the mutual eclipses of the stars, giving precise constraints on the absolute dimensions of all three bodies. The planet is comparable to Saturn in mass and size and is on a nearly circular 229-day orbit around its two parent stars. The eclipsing stars are 20 and 69% as massive as the Sun and have an eccentric 41-day orbit. The motions of all three bodies are confined to within 0.5° of a single plane, suggesting that the planet formed within a circumbinary disk. 相似文献

We present a statistical representation of the aggregate effects of deep convection on the chemistry and dynamics of the upper troposphere (UT) based on direct aircraft observations of the chemical composition of the UT over the eastern United States and Canada during summer. These measurements provide unique observational constraints on the chemistry occurring downwind of convection and the rate at which air in the UT is recycled. The Kepler spacecraft has been monitoring the light from 150,000 stars in its primary quest to detect transiting exoplanets. Here, we report on the detection of an eclipsing stellar hierarchical triple, identified in the Kepler photometry. KOI-126 [A, (B, C)], is composed of a low-mass binary [masses M(B) = 0.2413 ± 0.0030 solar mass (M(⊙)), M(C) = 0.2127 ± 0.0026 M(⊙); radii R(B) = 0.2543 ± 0.0014 solar radius (R(⊙)), R(C) = 0.2318 ± 0.0013 R(⊙); orbital period P(1) = 1.76713 ± 0.00019 days] on an eccentric orbit about a third star (mass M(A) = 1.347 ± 0.032 M(⊙); radius R(A) = 2.0254 ± 0.0098 R(⊙); period of orbit around the low-mass binary P(2) = 33.9214 ± 0.0013 days; eccentricity of that orbit e(2) = 0.3043 ± 0.0024). Thirty Santa Ines adult, non-lactating, non-pregnant ewes, ten with a brown coat, ten black coated and ten white coated, as well as ten Bergamasca and ten of mixed breed were used to evaluate the effect of climate on physiological and blood parameters in sheep. Two sample collections were taken (6 am and 2 pm) on six days. Sweating rate (SR), heart (HR) and breathing rates (BR), complete hemogram, rectal (RT) and skin temperatures (ST) were measured. Variance analyses were carried out using SAS®. In general, there were significant differences between animals due to skin type, time and day of collection. White coated animals showed lower HR, BR and RT, with afternoon parameters higher than morning. Correlations between HR, BR, RT, SR and ST were medium and positive. Correlations between physiological and blood traits were in general negative and medium. Packed Cell Volume (PCV), total plasma proteins, red blood cell count and hemoglobin concentration had high positive correlations between each other. The first two autovectors explained 49% of variation between traits. White coated Santa Ines animals were shown to be better adapted to climatic conditions in Central Brazil and wool sheep more affected by heat. 相似文献

Aroma compound properties in food matrices, such as volatility and diffusivity, have to be determined to understand the effect of composition and structure on aroma release and perception. This work illustrates the use of mass transfer modeling to identify diffusion and partition properties of ethyl hexanoate in water and in carrageenan matrices with various degrees of structure. The comparison of results obtained with a diffusive model to those obtained with a convective model highlights the importance of considering the appropriate transfer mechanism. Modeling of the preliminary experimental steps ensures correct estimation of the conditions for the main aroma release step. The obtained values of partition and diffusion coefficients are in agreement with those found in the literature (either experimentally determined or predicted by theoretical equations) and demonstrate that the structure level of carrageenan matrices has little influence on diffusion properties of ethyl hexanoate (less than 20%). 相似文献

To assess the suitability of water reuse technology for raising Pacific salmon Oncorhynchus spp. for stocking purposes, fish health and welfare were compared between two groups of juvenile Chinook salmon O. tshawytscha from the same spawn: one group was reared in a pilot partial water reuse system (circular tanks), and the other group was reared in a flow-through raceway. This observational study was carried out over a 21-week period in Washington State. Reuse and raceway fish were sampled repeatedly for pathogen screening and histopathology; fin erosion and whole-blood characteristics were also evaluated. By the study's end, no listed pathogens were isolated from either cohort, and survival was 99.3% and 99.0% in the reuse and raceway groups, respectively. Condition factor was 1.28 in raceway fish and 1.14 in reuse fish; this difference may have been attributable to occasional differences in feeding rates between the cohorts. Fin indices (i.e., length of the longest dorsal or caudal fin ray, standardized by fork length) were lower in reuse fish than in raceway fish, but fin erosion was not grossly apparent in either cohort. The most consistent histological lesion was gill epithelial hypertrophy in reuse fish; however, blood analyses did not suggest any corresponding physiological imbalances. Overall, results suggest that water reuse technology can be employed in rearing juvenile anadromous salmonids for stocking purposes. 相似文献

Grassland regions of the southern Great Plains are fragmented by agricultural activity and many habitat remnants have experienced
encroachment by juniper (Juniperus virginiana L.). Recently, many cropland areas have been converted to monoculture grassland (pastures) and enrolled into the Conservation
Reserve Program (CRP). Our objectives were to develop spatial and temporal Markov models to characterize land cover dynamics
relative to juniper expansion and CRP using aerial photography from 1965, 1981, and 1995. We used landscapes surrounding three
Breeding Bird Survey routes with varying levels of juniper encroachment in Oklahoma as study areas. As expected, land cover
changes from 1965 to 1995 included increases in juniper woodland, mixed juniper-deciduous woodland, and pastures from CRP
activity. Markov models revealed that juniper had a low likelihood of self-replacement in early stages of encroachment. In
all areas, relatively little native grassland was lost to juniper encroachment, but other native habitat types such as deciduous
woodland were heavily impacted. Transition probabilities for land cover dynamics varied significantly both spatially and temporally.
Projections of these raw transition matrices produced widely varying models of future land cover conditions. By modifying
the matrices to account for recent and potential socio-political and ecological changes occurring in this region, a number
of more plausible land cover scenarios were produced than those resulting from simple projections of raw transition matrices. 相似文献

- Among the several threats to the conservation of mangrove ecosystems in most South Asian countries, shrimp farming is predominant. Since the introduction of shrimp farming in Sri Lanka in the 1980s, mangroves on the island’s north-western coast have been continually cleared to create new shrimp farms, leading to a decline in the social-ecological services provided by the mangrove ecosystems.
- Using aerial (1973) and satellite (1996–2020) images, this study assessed areal changes in mangroves and shrimp farms in the Pambala-Chilaw lagoon complex and Ihala Mahawewa, as well as the ecological footprint of shrimp farming in the study area.
- Mangroves around the Chilaw lagoon had decreased in areal extent by 45% from 1973 to 2020 of which 92% of this change was attributed to shrimp farming. There was, however, a decrease in the areal extent of shrimp farms from 2001 to 2020, and a corresponding increase in mangroves from 2006 to 2020.
- The ecological footprint of shrimp farming was assessed by comparing the expected surface ratios with those recorded for shrimp farms with mangroves and surface water bodies in the study area from 1973 to 2020. The results showed that the current shrimp farming was unsustainable (i.e. high ecological footprint).
- While the results support the current view that there is cause for cautious optimism with mangrove conservation (as evidenced by an increase in mangrove areal extent), it also reveals that semi-intensive shrimp farming in Sri Lanka and probably other similar tropical countries is unsustainable.
- If immediate actions such as effective regulation of shrimp farming activities and mangrove restoration are not taken, the mangrove ecosystem will continue to decline.
