The biochemical and gel properties of Pacific sardine and Pacific mackerel were characterized as affected by preparation and cooking methods. Four to eight times more salt soluble proteins were extracted from water-washed paste than fish protein isolate (FPI) paste. Higher total sulfhydryl content was measured in FPI, indicating the exposure of sulfhydryl groups during alkaline extraction. Comparing gel properties based on two cooking methods (slow and fast), the two pelagic fish proteins performed quite differently. Heating rate did not differentiate between surimi and FPI gels from sardine. However, mackerel exhibited higher texture values when using the fast cooking method, indicating the presence of high levels of proteolytic enzymes. Water-washed surimi gels were whiter than FPI for both species. Water retention ability appeared to be higher with mackerel than sardine, regardless of isolation and cooking method. Sodium dodecyl sulfate polyacrylamide gel eletrophoresis (SDS-PAGE) also supported a difference in processing chemistry and thermal behavior between two methods for protein isolation and cooking, respectively.  相似文献

Chaetognatha were among the most dominant macrozooplankton taxa collected in 6 years of springtime collections, both throughout the water column and near-bottom on the continental shelf in the northern Gulf of Alaska. Three species ( Sagitta elegans, S. scrippsae, and Eukrohnia hamata ) were collected in 248 collections, although S. elegans was the numerically dominant species overall during most cruises. Collections taken in epibenthic sleds generally contained about two orders of magnitude more chaetognaths than those that sampled throughout the water column. Two size modes were apparent in the S. elegans size-frequency distributions which were believed to belong to different cohorts. Growth rate was on the order of 2–3 mm per month. The two cohorts showed asynchronous diel vertical distribution patterns, with the smaller individuals found near the surface during the day whereas the larger individuals were near the surface at night. About 7.5% of the S. elegans examined contained food. Copepods made up the majority (∼89% by number) of the diet of both large and small individuals. Euphausiid juveniles and cirripede larvae were also observed, as well as several incidences of cannibalism by large chaetognaths on smaller individuals. Based on the results of two diel series, this species was observed to feed mainly during the night-time.  相似文献

Abstract. Eleven freshwater salmonid hatcheries in southern Australia were surveyed for bacterial pathogens and diseases between 1981 and 1985, Twenty-five populations of fish were examined in the study, representing a total of 2755 fish, from which kidney, liver, spleen, and in some cases peritoneum, blood and faeces were cultured. Bacteria of pathogenic significance isolated included Aeromonas hvdrophila, Streptococcus sp., Lactobacillus piscicola, Yersinia ruckeri, Mycobacterium fortuitum, M. chelonei and a filamentous acid-fast organism of uncertain taxonomic position. Lacto-bacillus piscicola and Streptococcus sp. were associated with clinical and subclinical peritonitis. Mycobacterium chelonei was isolated from visceral granulomas in an externally normal fish. Aeromonas salmonicida, Renibacterium salmoninarum and Edwardsiella tarda were not isolated, indicating that the diseases furunculosis, bacterial kidney disease and edwardsiellosis are exotic to Australian salmonids. Similarly, while Y. ruckeri was isolated, enteric redmouth disease had not been recorded and is considered an exotic disease.  相似文献

We tested the effect of extended drying of half the root system on fruit yield and fruit Ca concentration, an indirect measure of fruit quality, in avocado (Persea americana Mill. cv Hass). In a field experiment on a sandy soil, withholding irrigation and plastic sheeting was used to dry the root-zone beneath the whole canopy (DD) or half the canopy (WD), compared with well-watered trees (WW). The irrigation water contained added nutrients and was slightly saline. Yield, shoot growth, leaf conductance, leaf and fruit water status and mineral concentrations of leaves and fruit were studied. The responses of treated trees were assessed in the following season during which normal irrigation practices were restored. With respect to yield, the WD treatment behaved the same as the DD treatment. It reduced yield by more than half and proportionately more than the reduction in water supply thus reducing irrigation efficiency. Re-watering did not restore yield of WD or DD-trees in the next season. The WD and DD treatments had no effect on the concentration of Ca in the fruit mesocarp and so are unlikely to affect fruit quality. The main impact of reduced water supply on the trees was fruit abscission and this was linked to dry soil around the roots rather than the water status of the leaves or fruits. We conclude that extended drying of half of the root-zone in one season reduced irrigation efficiency for two seasons by promoting the abscission of developing fruit to the same extent as occurred when the whole root system was exposed to extended drying.  相似文献

The β-glucuronidase (GUS) gene reporter system is one of the most effective and employed techniques in the study of gene regulation in plant molecular biology. A new cleanup procedure is described for chlorinated hydrocarbon residues in butterfat. The method is based on the dropwise addition of H2SO4 to a fat solution column and continuous removal of the lipids and the acid. The cleanup of 0.25-2.0 g fat requires only 10-40 ml sulfuric acid and 12-17 ml petroleum ether. There is no need for any further cleanup step, solvent evaporation, or centrifugation. The method is easy to standardize and is suitable for automation. At least 30 fat samples can be cleaned up manually by one analyst in one day. Recoveries were complete (greater than 90%) for polychlorinated biphenyl compounds and for 13 chlorinated pesticides of 16 examined. The method was tested on chlorinated hydrocarbon residues in commercial butter and the results were compared with those obtained with the acetonitrile method. The versatility and limitations of the method were investigated by varying the sulfuric acid strength, initial fat solution concentration, and column dimensions.  相似文献
