Lipid A is a fundamental Gram-negative outer membrane component and the essential element of lipopolysaccharide (endotoxin), a potent immunostimulatory molecule. This work describes the metabolic adaptation of the lipid A acyl structure by Psychrobacter cryohalolentis at various temperatures in its facultative psychrophilic growth range, as characterized by MALDI-TOF MS and FAME GC-MS. It also presents the first elucidation of lipid A structure from the Colwellia genus, describing lipid A from strains of Colwellia hornerae and Colwellia piezophila, which were isolated as primary cultures from Arctic fast sea ice and identified by 16S rDNA sequencing. The Colwellia strains are obligate psychrophiles, with a growth range restricted to 15 °C or less. As such, these organisms have less need for fluidity adaptation in the acyl moiety of the outer membrane, and they do not display alterations in lipid A based on growth temperature. Both Psychrobacter and Colwellia make use of extensive single-methylene variation in the size of their lipid A molecules. Such single-carbon variations in acyl size were thought to be restricted to psychrotolerant (facultative) species, but its presence in these Colwellia species shows that odd-chain acyl units and a single-carbon variation in lipid A structure are present in obligate psychrophiles, as well.  相似文献

This study evaluated onset and incidence of palmar process fractures in foals and investigated corresponding changes in hoof conformation. Radiographs of 17 warmblood foals were taken at 1 week and at 2, 4, 6, 8, and 12 months of age and examined for evidence of palmar process fractures. Hoof parameters were measured from the radiographs, and comparisons were made between fractured and nonfractured feet. All foals suffered at least one fracture during the study period, with an average age of 3.68 ± 0.20 months at fracture occurrence. Hoof conformation and fracture occurrence were not significantly correlated. Foals consistently demonstrated higher lateral heights in the right foot and higher medial heights in the left foot. Hoof angle increased until 3 months of age and then gradually dropped. Hoof-pastern axis exhibited a broken forward conformation, but approached a more correct conformation by 12 months of age. There was a significant reduction in variation of variables for hoof and pastern angle in both feet, and hoof-pastern axis in the right foot. Hoof conformation does not appear to be a causative factor of palmar process fractures. It is possible that these fractures are a normal part of bone remodeling.  相似文献

The amount and fatty acid composition of intramuscular fat (IMF) are major factors in determining the nutritional value of beef. Understanding the mechanisms regulating IMF formation is important for designing strategies for manipulation of IMF quantity and quality. The aim of this study was to investigate the contribution of key lipogenic enzymes towards intramuscular fat formation in Aberdeen Angus and Limousin crossbred steers. This study reports the presence of 150 kDa acetyl-CoA-carboxylase α (ACCα), 150 kDa fatty acid synthase (FAS), 37 kDa stearoyl-CoA desaturase (SCD), 50 kDa Δ6-desaturase (Δ6d), and 50 kDa Δ5-desaturase (Δ5d) immunoreactive bands in cattle muscle (as detected by Western blot analyses). A significant positive relationship was established between ACCα protein content and IMF content; FAS protein content and IMF, as well as between expression of these enzymes and the products of their reactions, saturated fatty acid. IMF level was also positively related to the expression of Δ6d and Δ5d proteins. However, no relationship was found between IMF and the amount of SCD protein. It is suggested that the rate of saturated fatty acids and PUFA biosynthesis might have a significant impact on IMF deposition in cattle.  相似文献

Rangelands occupy 50% of Earth’s land surface and thus are important in the terrestrial carbon (C) cycle. For rangelands and other terrestrial ecosystems, the balance between photosynthetic uptake of carbon dioxide (CO2) and CO2 loss to respiration varies among years in response to interannual variation in the environment. Variability in CO2 exchange results from interannual differences in 1) environmental variables at a given point in the annual cycle (direct effects of the environment) and in 2) the response of fluxes to a given change in the environment because of interannual changes in biological factors that regulate photosynthesis and respiration (functional change). Functional change is calculated as the contribution of among-year differences in slopes of flux-environment relationships to the total variance in fluxes explained by the environment. Functional change complicates environmental-based predictions of CO2 exchange, yet its causes and contribution to flux variability remain poorly defined. We determine contributions of functional change and direct effects of the environment to interannual variation in net ecosystem exchange of CO2 (NEE) of eight rangeland ecosystems in the western United States (58 site-years of data). We predicted that 1) functional change is correlated with interannual change in precipitation on each rangeland and 2) the contribution of functional change to variance in NEE increases among rangelands as mean precipitation increases. Functional change explained 10–40% of the variance in NEE and accounted for more than twice the variance in fluxes of direct effects of environmental variability for six of the eight ecosystems. Functional change was associated with interannual variation in precipitation on most rangelands but, contrary to prediction, contributed proportionally more to variance in NEE on arid than more mesic ecosystems. Results indicate that we must account for the influence of precipitation on flux-environment relationships if we are to distinguish environmental from management effects on rangeland C balance.  相似文献

Rangeland ecosystems cover approximately one-third of the land area in the United States and half of the land area of California. This large land area, coupled with the propensity of grasses to allocate a considerable proportion of their photosynthate belowground, leads to high soil carbon (C) sequestration potential. Annual grasslands typical of the Mediterranean climates of the western United States differ in their life history strategies from the well-studied perennial grasslands of other regions and thus may also differ in their soil C pools and fluxes. In this study we use the literature to explore patterns in soil C storage in annual grass-dominated rangelands in California. We show that soil C is highly predictable with depth. Cumulative soil C content increased to 2–3-m depth in rangelands with a woody component and to at least 1-m depth in open rangelands. Soil C within a given depth varied widely, with C content in the top 1-m depth spanning almost 200 Mg C · ha?1 across sites. Soil C pools were not correlated with temperature or precipitation at a regional scale. The presence of woody plants increased C by an average of 40 Mg · ha?1 in the top meter of soil. Grazed annual grasslands had similar soil C content as ungrazed grassland at all depths examined, although few details on grazing management were available. Soil C pools were weakly positively correlated with clay content and peaked at intermediated levels of aboveground net primary production. Our results suggest that annual grasslands have similar soil C storage capacity as temperate perennial grasslands and offer an important resource for mitigation of greenhouse gas emissions and climate change.  相似文献

Protein synthesis requires several guanosine triphosphatase (GTPase) factors, including elongation factor Tu (EF-Tu), which delivers aminoacyl-transfer RNAs (tRNAs) to the ribosome. To understand how the ribosome triggers GTP hydrolysis in translational GTPases, we have determined the crystal structure of EF-Tu and aminoacyl-tRNA bound to the ribosome with a GTP analog, to 3.2 angstrom resolution. EF-Tu is in its active conformation, the switch I loop is ordered, and the catalytic histidine is coordinating the nucleophilic water in position for inline attack on the γ-phosphate of GTP. This activated conformation is due to a critical and conserved interaction of the histidine with A2662 of the sarcin-ricin loop of the 23S ribosomal RNA. The structure suggests a universal mechanism for GTPase activation and hydrolysis in translational GTPases on the ribosome.  相似文献

Controlled chemical modifications of single-walled carbon nanotubes (SWCNTs) that tune their useful properties have been sought for multiple applications. We found that beneficial optical changes in SWCNTs resulted from introducing low concentrations of oxygen atoms. Stable covalently oxygen-doped nanotubes were prepared by exposure to ozone and then light. Treated samples showed distinct, structure-specific near-infrared fluorescence at wavelengths 10 to 15% longer than displayed by pristine semiconducting SWCNTs. Dopant sites harvest light energy absorbed in undoped nanotube regions by trapping mobile excitons. The oxygen-doped SWCNTs are much easier to detect and image than pristine SWCNTs because they give stronger near-infrared emission and do not absorb at the shifted emission wavelength.  相似文献

Regional conservation planning frequently relies on general assumptions about historical disturbance regimes to inform decisions about landscape restoration, reserve allocations, and landscape management. Spatially explicit simulations of landscape dynamics provide quantitative estimates of landscape structure and allow for the testing of alternative scenarios. We used a landscape fire succession model to estimate the historical range of variability of vegetation and fire in a dry forest landscape (size ca. 7900 km2) where the present-day risk of high severity fire threatens the persistence of older closed canopy forest which may serve as Northern Spotted Owl (Strix occidentalis caurina) habitat. Our results indicated that historically, older forest may have comprised the largest percentage of the landscape (∼35%), followed by early successional forest (∼25%), with about 9% of the landscape in a closed canopy older forest condition. The amount and condition of older forest varied by potential vegetation type and land use allocation type. Vegetation successional stages had fine-grained spatial heterogeneity in patch characteristics, with older forest tending to have the largest patch sizes among the successional stages. Increasing fire severities posed a greater risk to Northern Spotted Owl habitat than increasing fire sizes or frequencies under historical fire regimes. Improved understanding of historical landscape-specific fire and vegetation conditions and their variability can assist forest managers to promote landscape resilience and increases of older forest, in dry forests with restricted amounts of habitat for sensitive species.  相似文献

While food abundance is known to limit bird populations, few studies have simultaneously examined both temporal and habitat-related variation in food abundance for ground-foraging birds. We surveyed ground-dwelling arthropods throughout the breeding season and in three forest harvest types (clearcut, selection, and shelterwood stands managed for spruce, fir and hemlock) at the Penobscot Experimental Forest (PEF), Bradley, Maine, USA. Pitfall trapping during 2003 and 2004 was used to measure seasonal and habitat-related variation in abundance of ground-dwelling arthropod food items for hermit thrushes. Arthropods were sorted into relevant taxonomic and size class categories based on the known diet of hermit thrushes. Habitat variables included forest management type, stand-level structure and composition of tree species, and microhabitat measures in a 1 m2 area around the pitfall traps. We used redundancy analysis to find stand-level and microhabitat measures that best explained variation in arthropod abundance during 2004. The majority of arthropod captures comprised three groups: ants (Formicidae, 44%), ground beetles (Carabidae, 13%), and spiders (Araneae, 11%). There was significant temporal variation in captures for many taxa. Notably, there were as many as six times more Araneae in early compared to late season samples suggesting the importance of Araneae to ground-foraging birds. Carabidae were more abundant in selection versus either shelterwood or clearcut stands on one sample date. Conversely, Formicidae and Curculionidae were more abundant in shelterwood plots on one and three sampling dates, respectively. Overall, our data provided little evidence that forest management type impacts arthropod food abundance, in stands 5–20 years post-harvest. Abundance of Carabidae was positively correlated with the number of softwoods less than 5 cm DBH and the number of hardwoods greater than 10 cm DBH, while abundance of Formicidae and Homopterans was negatively associated with these same measures. Several taxa were negatively associated with percent of the 1 m2 around the pitfall that was shaded, including Araneae, Carabidae, Formicidae, and Staphylinidae. These results suggest that less-shaded forest microhabitats are important areas of increased food abundance for ground-foraging birds, regardless of forest management type or tree composition. Consequently, forest managers seeking to maximize food abundance for ground-foraging birds in managed spruce-fir forests should consider retaining some large diameter hardwood trees and use management techniques that reduce forest floor shading.  相似文献
