The yield and chemical composition of the seed oil of Celastrus paniculatus were determined by solvent extraction and gas chromatography-mass spectrometry. The yield of crude fatty oil was found to be 45.5% in the mature seeds on dry weight basis. Fourteen compounds, constituting 97.57% of the oil, were identified. The oil was rich in unsaturated fatty acids (70.11%) followed by saturated fatty acids (25.2%). Oleic acid (54.42%), palmitic acid (20.0%), linoleic acid (15.51%), and stearic acid (4.18%) were the main fatty acids in the oil together with four other minor fatty acids. The current study identifies the novel compounds not previously reported in C. paniculatus oil, including 1,4-benzenediol (0.46%), cinnamic acid (0.15%), 2,6-di-tert-butyl-p-benzoquinone (0.03%), butylated hydroxytoluene (0.4%), and eudalene (0.16). 相似文献
Tuberose cut flowers, available as loose flowers, were treated with gamma (γ) irradiation and generally regarded as safe (GRAS) preservative solutions for extension of shelf life. The flowers were packaged in low-density polyethylene bags, heat sealed and stored at 23 ± 2°C, 80% relative humidity (RH) and 4 ± 1°C, 40% RH, respectively. The flowers stored at these two temperature regimes were subjected to sensory evaluation and biochemical analyses. From these assessments, the longest shelf life of tuberose flowers was found to be 8 days at 23 ± 2°C, 80% RH (compared to 4 days for control) and 24 days at 4 ± 1°C, 40% RH (compared to 8 days for control) using combination treatment of low dose γ-irradiation (0.02 kGy) and preservative solutions (4% sucrose and 0.02% CaCl2). Ethanolic extract of tuberose flowers of the most shelf stable set (stored at 4 ± 1°C), i.e. at the end of 24 days, showed antimicrobial potency against the common skin pathogen Staphylococcus aureus (ATCC 25923 and MDR strains), suggesting utility of the senesced tuberose flowers for therapeutic applications. Developmental biology has made great strides in recent years towards the quantification of cellular properties during development. This requires tissues to be imaged and segmented to generate computerised versions that can be easily analysed. In this context, one of the principal technical challenges remains the faithful detection of cellular contours, principally due to variations in image intensity throughout the tissue. Watershed segmentation methods are especially vulnerable to these variations, generating multiple errors due notably to the incorrect detection of the outer surface of the tissue.
Results
We use the level set method (LSM) to improve the accuracy of the watershed segmentation in different ways. First, we detect the outer surface of the tissue, reducing the impact of low and variable contrast at the surface during imaging. Second, we demonstrate a new edge function for a level set, based on second order derivatives of the image, to segment individual cells. Finally, we also show that the LSM can be used to segment nuclei within the tissue.
Conclusion
The watershed segmentation of the outer cell layer is demonstrably improved when coupled with the LSM-based surface detection step. The tool can also be used to improve watershed segmentation at cell-scale, as well as to segment nuclei within a tissue. The improved segmentation increases the quality of analysis, and the surface detected by our algorithm may be used to calculate local curvature or adapted for other uses, such as mathematical simulations.
Weeds caused serious problem on yield reduction of basmati rice worldwide. Losses caused by weeds varied from one country to another, depending on the presence of dominant weeds and the control methods practiced by farmers; therefore, suitable plant population and weed management practices should be adopted. Keeping these in mind, a field experiment was carried out during kharif seasons of 2009 and 2010 at crop Research Centre of SVPUA&T, Meerut, India comprising 4 planting geometries, viz. 20, 30, 40, and 50 hills m?2 as main plot factor, and 5 weed management practices (Butachlor @ 1.0 kg ha?1, Butachlor @ 1.0 kg ha?1 fb (followed by) one hand weeding, Butachlor @ 1.0 kg ha?1 fb Almix @ 4 g ha?1, two hand weedings and weedy check) in a split plot design with 3 replications. Experimental results revealed that plant population of 50 hills m?2 proved superior over that of 20 hills m?2 in respect of weed density, weed dry weight, number of tillers m?2, yield attributes, grain, straw, and biological yields. The maximum grain yield (29.00 and 31.00 q ha?1) and straw yield (51.30 and 52.50 q ha?1) were recorded in 50 hills m?2 followed by 40 hills m?2 during 2009 and 2010, respectively. In respect of nitrogen, phosphorus, and potassium removal, a reverse trend was observed: the highest in 20 hills m?2 followed by 30, 40, and 50 hills m?2. As far as the weed management practices are concerned, both chemical and mechanical methods of weed control were found superior over weedy check. The lowest weed density, dry weight, and highest weed control efficiency, maximum length of panicle?1, number of panicle (m2), and 1000-grain weight and grain yield of 30.40 and 32.60 q ha?1 were recorded with two hand weedings which was at par with Butachlor @ 1.0 kg ha?1 fb one hand weeding over rest of the weed management practices. 相似文献
A field study on assessment of crop establishment methods on yield, economics and water productivity of rice cultivars under upland and lowland production ecologies was conducted during wet seasons (June–November) of 2012 and 2013 in Eastern Indo-Gangetic Plains of India. The experiment was laid-out in a split-plot design (SPD) and replicated four times. The main-plot treatments included three crop establishment methods, viz. dry direct-seeded rice (DSR), system of rice intensification (SRI) and puddled transplanted rice (PTR). In sub-plots, five rice cultivars of different groups like aromatic (Improved Pusa Basmati 1 and Pusa Sugandh 5), inbreds (PNR 381 and Pusa 834) and hybrid (Arize 6444) were taken for their evaluations. These two sets of treatments were laid-out simultaneously in two production ecologies, upland and lowland during both years. In general, lowland ecology was found favourable for rice growth and yield and resulted in 13.2% higher grain yield as compared to upland ecology. Rice grown with SRI method produced 19.4 and 7.0% higher grain yield in 2012 and 20.6 and 7.1% higher in 2013, over DSR and PTR. However, PTR yielded 13.1 and 14.5% higher grain over DSR during 2012 and 2013, respectively. On an average, Arize 6444 produced 26.4, 26.9, 28.9 and 54.7% higher grain yield as compared to PS 5, P 834, PNR 381 and IPB1, respectively. Further, the interaction of production ecologies × crop establishment methods revealed that, in upland ecology, SRI recorded significantly higher grain yield as compared to PTR and DSR, but in lowland, grain yield resulting from SRI was similar to the yield obtained with PTR and significantly higher than DSR. The latter two methods (PTR and DSR) yielded alike in lowland ecology in both study years. The production ecologies × crop establishment methods × cultivars interaction on grain yield showed that the growing of Arize 6444 cultivar using SRI method in upland ecology resulted in the higher grain yield (8.87 t/ha). But the cost of production was also highest in SRI followed by PTR and DSR across production ecologies and cultivars. Cultivation of hybrid (Arize 6444) involved higher cost of production than all other cultivars. Irrespective of crop establishment methods and cultivars, gross returns, net returns and B:C ratio were significantly higher in lowland compared to upland ecology. Owing to higher grain yield, SRI method fetched significantly higher gross returns and net returns over PTR and DSR. Average increase in net return with Arize 6444 was 68.8, 41.0, 37.7 and 33.1% over IPB 1, PNR 381, P 834 and PS 5, respectively. There was a saving of 30.7% water in SRI and 19.9% in DSR over PTR under upland ecology. Similarly in lowland ecology, water saving of 30.2% was observed in SRI and 21.2% in DSR over PTR. Due to higher yield and saving on water, SRI returned significantly higher total water productivity (TWP) (5.9 kg/ha-mm) as compared to DSR (3.5 kg/ha-mm) and PTR (3.6 kg/ha-mm) under upland ecology. In lowland ecology, also SRI (6.2 kg/ha-mm) resulted in higher TWP as compared to other two methods. However, DSR gave significantly higher TWP as compared to PTR. Among cultivars, hybrid Arize 6444 recorded the highest TWP in both upland and lowland production ecologies across crop establishment methods. Hence, growing of hybrid Arize 6444 with SRI method can enhance rice productivity and water-use efficiency in lowland and upland production ecologies of Eastern Indo-Gangetic Plains and in other similar regions. 相似文献
In vegetable brassicas, cytoplasmic male sterility (CMS) system is the most preferred mechanism for hybrid seed production and Ogura cybrid cytoplasm is the only source used widely. Effects of the alien cybrid cytoplasm on important quality traits need to be understood for their effective use in breeding programme. In analysing 38 Ogura cybrid cytoplasm‐based cauliflower CMS lines, it was revealed that introgression of Ogura cytoplasm resulted in significant alteration in important quality traits. Cupric reducing antioxidant activity and ferric reducing ability of plasma values were increased up to 5–10 times in different genotypes. Among 38 analysed CMS lines, concentration of plant pigments such as anthocyanin, total chlorophylls and ascorbic acid was reduced in 21, 17 and 32 genotypes, respectively. However, the concentration of total carotenoids and β‐carotene was elevated in most of the CMS lines and total carotenoid was increased up to 20 times in the CMS line, Ogu13‐85‐2A. The results indicate the role of nuclear–cytoplasmic interaction and mitochondrial genome in determining concentration of different quality traits. 相似文献
Phytoremediation is a promising approach for reclamation of salt-affected soil. Phytoextraction is the most commonly used process, which exploits plants to absorb, immobilize, and accumulate salt in their shoots. In this study, halotolerant plant growth-promoting rhizobacteria (PGPR) were isolated from the rhizosphere of wild grasses growing naturally in salt-affected areas of Lucknow, Uttar Pradesh (India) and were tested for their efficacies of salt-tolerance and plant growth-promoting (PGP) abilities. Based on 16S rRNA sequences, the most efficient halotolerant isolates possessing PGP traits were identified as Pseudomonas plecoglossicida (KM233646), Acinetobacter calcoaceticus (KM233647), Bacillus flexus (KM233648), and Bacillus safensis (KM233652). Application of these isolates as bio-inoculants significantly (P < 0.05) increased the growth and bacoside A yield of a medicinal plant, Bacopa monnieri (L.) Nash, grown on natural salt-affected soil. The phytoremediation of salt-affected soil was evident by the substantial increase in shoot Na+:K+ ratio of bio-inoculant-treated plants. When compared to un-inoculated control plants, the soil physico-chemical properties of bio-inoculant-treated plants were improved. The shoot and root biomass (fresh and dry weights), soil enzymes, and soil nutrient parameters showed significant positive correlations with the shoot Na+:K+ ratio. Consequently, the halotolerant PGPR screened in this study could be useful for the reclamation of saline soils concomitant with improved plant growth and bacoside A yield. 相似文献
Biochars produced from different feedstocks (such as wood, pig manure) possess varying physical and chemical properties, which have influence on crack and evaporation rate of biochar-amended soil (BAS). Furthermore, influence of compaction state and drying-wetting cycles on evaporation rate and cracking of BAS has not been investigated comprehensively. The objective of this study was to investigate the effects of biochar types, compaction state of BAS, and drying-wetting cycles on crack propagation and retained water (or evaporation rate).
Material and methods
An animal and plant feedstock-based biochars were produced in-house from pig manure (PM) and wood (W), respectively. In addition, nano structured chalk and wheat biochar (CWB) were also produced. Soil amended with individual biochars was compacted in petri-glass discs at two densities. Disc specimens were subjected to multiple drying-wetting cycles, and evaporation rate of specimens and crack area were monitored throughout the experimental period (70 days). Images were captured after every 24 h and processed using image processing technique to obtain the crack intensity factor (CIF).
Results and discussion
The results show that plant-based W BAS showed the high water retention, i.e., low evaporation rate and low CIF. Furthermore, the crack potential of CW BAS was seen to be higher. In dense compacted soil, maximum CIF% can be reduced from 3.9 to 0.4% for W BAS, from 3.9 to 1.7% for PM BAS, and from 3.9 to 1.6% for CW BAS.
Conclusion
WB was able to resist cracking more efficiently than other types of biochar. Evaporation was found to be minimal for plant-based W BAS at 10% biochar percentage. Higher biochar content in soil was seen to increase the water retention of BAS significantly. Dense state of BAS at high biochar content (i.e., 10%) was effective in reducing evaporation rate and crack progression.
It has been widely hypothesized that a warmer climate in Greenland would increase the volume of lubricating surface meltwater reaching the ice-bedrock interface, accelerating ice flow and increasing mass loss. We have assembled a data set that provides a synoptic-scale view, spanning ice-sheet to outlet-glacier flow, with which to evaluate this hypothesis. On the ice sheet, these data reveal summer speedups (50 to 100%) consistent with, but somewhat larger than, earlier observations. The relative speedup of outlet glaciers, however, is far smaller (<15%). Furthermore, the dominant seasonal influence on Jakobshavn Isbrae's flow is the calving front's annual advance and retreat. With other effects producing outlet-glacier speedups an order of magnitude larger, seasonal melt's influence on ice flow is likely confined to those regions dominated by ice-sheet flow. 相似文献
Soil ecosystem is experiencing stresses due to climate change, and soil inhabitants try to demonstrate their inherent resistance and resilience against those stresses. Application of nanomaterials as agricultural inputs could bring shifts in resistance and resilience patterns of soil microbes and associated enzymes, especially under short-term heat stress. With this background, the impacts of multi-walled carbon nanotube (MWCNT) on the resistance and resilience of soil biological indicators were evaluated. An incubation experiment was conducted with varied MWCNT concentrations (0, 50, 100, 250, and 500 mg kg-1 soil) for 90 d after 24-h heat stress at 48 ±2 ℃ to assess the impacts of MWCNT on soil enzyme activities and microbial populations vis-à-vis their resistance and resilience indices under short-term exposure to heat stress. Enzyme activities were reduced after exposure to heat stress. Resistance indices of enzyme activities were enhanced by MWCNT application on day 1 after heat stress, whereas there was no recovery of enzyme activities after 90-d incubation. Like soil enzyme activities, resistance index values of soil microbial populations followed the similar trend and were improved by MWCNT application. Multi-walled carbon nanotube has the potential to improve resistance indices of soil enzyme activities and microbial populations under heat stress, although they could not recover to their original state during periodical incubation after heat stress.
