Seedcoats of 16 almond varieties were screened for flavonol glycosides by using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Flavonol glycosides were extracted by a simple methanolic extraction followed by a quick cleanup procedure with a Sep-Pak C(18) cartridge. Each of the 16 seedcoat samples exhibited a unique composition. Four flavonol glycosides, isorhamnetin rutinoside, isorhamnetin glucoside, kaempferol rutinoside, and kaempferol glucoside, were detected and quantified with use of rutin as an internal standard. Individual peak ratios were very consistent across triplicate analyses of all samples; the average standard deviation was 9%. In all almond varieties, isorhamnetin rutinoside was the most abundant flavonol glycoside, and the total content ranged from 75 to 250 microg/g.  相似文献

Tea cream is the precipitate formed as tea cools. Its formation has been studied by X-ray scattering, and it is shown that a higher tea concentration leads to earlier onset of creaming and larger particles and that addition of theaflavin and calcium promotes creaming. Association constants between the major components of black tea have been obtained using NMR and show that calcium and glucose enhance the self-association of caffeine, polyphenols, and theaflavin but have little effect on hetero-association. Glycosylation of a polyphenol reduced self-association and reduced binding to caffeine. We conclude that theaflavin is important for the initiation of creaming, forming nanoclusters of typically 3 nm diameter, whereas caffeine acts more to fill in the gaps within the clusters and thus adds to the bulk of tea cream without being necessary for its initiation. Tea creaming may be reduced by increasing the solubility of the polyphenols (i.e., by glycosylation) or by removing calcium. A LC-ES-MS/MS method for the identification of dilignols formed by the oxidative cross-coupling of coniferyl alcohol and apocynol has been developed. The identification is based on the generation of ammonium adduct ions [M + NH(4)](+) by electrospray ionization and thereafter the following fragmentation patterns for the selected precursor ions. Fragmentation of several arylglycerol-beta-aryl ether (beta-O-4) and phenyl coumarane (beta-5) model compounds were studied as a reference.  相似文献

Methods to refine the assessment of exposure of wild birds and mammals to pesticides required measurement of pesticide residues in very small samples of their diets. Sample sizes were in the 1-100 mg range, and the target residue for measurement was 0.01 mg/kg. Gas chromatography-mass spectrometry (GC-MS) with large volume injection was compared with the use of an accelerator mass spectrometer (AMS) to measure residues of pesticide labeled at near-background levels with carbon-14. The GC-MS method was able to detect residues down to 0.1 ng per item of diet, and the AMS detected the radiolabel down to 1 mBq (0.06 disintegration per minute, 1 ng of pesticide at the specific activity used) per sample. The target residue level was achieved by the GC-MS method for samples down to 10 mg. The GC method appeared to be best suited to monitoring residues in field studies, and the AMS shows great potential for use in laboratory experiments concerning pesticide degradation.  相似文献

We studied the reactions of humus layer (F/H) microbial respiratory activity, microbial biomass C, and the fungal biomass, measured as the soil ergosterol content, to the application of three levels of wood ash (1000, 2500, and 5000 kg ha-1) and to fire treatment in a Scots pine (Pinus sylvestris L.) stand. Physicochemical measurements (pH, organic matter content, extractable and total C content, NH 4 + and total N content, cation-exchange capacity, base saturation) showed similarity between the fire-treated plots and those treated with the lowest dose of wood ash (1000 kg ha-1). The ash application did not change the level of microbial biomass C or fungal ergosterol when compared to the control, being around 7500 and 350 g g-1 organic matter for the biomass C and ergosterol, respectively. The fire treatment lowered the values of both biomass measurements to about half that of the control values. The fire treatment caused a sevenfold fall in the respiration rate of fieldmoist soil to 1.8 l h-1 g-1 organic matter compared to the values of the control or ash treatments. However, in the same soils adjusted to a water-holding capacity of 60%, the differences between the fire treatment and the control were diminished, and the ash-fertilized plots were characterized by a higher respiration rate compared to the control plots. The glucose-induced respiration reacted in the same way as the water-adjusted soil respiration. The metabolic quotient, qCO2, gradually increased from the control level with increasing applications of ash, reaching a maximum in the fire treatment. Nitrification was not observed in the treatment plots.  相似文献

Sri Lanka supports a diverse fauna of freshwater fishes, 24% of which are endemic. A survey of the island's streams indicated that a number of the endemic fishes are threatened with extinction and others are depleted due to the interaction of deforestation, urbanization, gem mining, pesticides, exploitation, water diversions, and introductions of exotic species. It is clear that these declines should be halted for ethical, economic, and ecological reasons. Methods proposed to reverse the declines include captive breeding, better watershed management, translocation, and regulation of fisheries, especially that for aquarium fishes. The depauperate Second Peneplain is proposed as a site for translocation of endangered species.  相似文献

Sverdrupet al. (1992) proposed a model intended to evaluate effects of soil solution (Ca2++Mg2+/Al3+ ratios on the uptake of Ca2+ and Mg2+ (here called base cations=BC) by tree roots. They considered the adsorption of BC on exchange sites on root surfaces to be a major step, and the competition between Al3+ and BC for exchange sites on roots to be crucial. Although this may certainly affect the rates of uptake, it will not exclude base cations from entering the root cells through ion channels or by carriers. Above all, the model overlooks important gradients in the rhizosphere, e.g. pH-gradients and root exudates, which have profound influences on the speciation of Al. One should instead consider modifying existing models accounting for processes in a buffered soil medium and handling gradients in the rhizosphere (e.g. Nye and Tinker, 1977). This would also require considerations on mycorrhizas and possible changes in the capacity for uptake of certain ions, which could be induced by deficiencies.  相似文献

The observation that the bitterest peptides from casein hydrolysates contain several proline residues led us to hypothesize that a proline-specific protease would be instrumental in debittering such peptides. To identify the desired proline-specific activity, a microbiological screening was carried out in which the chromogenic peptide benzyloxycarbonyl-glycine-proline-p-nitroanilide (Z-Gly-Pro-pNA) was used as the substrate. An Aspergillus niger (A. niger) strain was identified that produces an extracellular proline-specific protease with an acidic pH optimum. On the basis of sequence similarities, we conclude that the A. niger-derived enzyme probably belongs to the S28 family of clan SC of serine proteases rather than the S9 family to which prolyl oligopeptidases belong. Incubating the overexpressed and purified enzyme with bitter casein hydrolysates showed a major debittering effect. Reversed phase HPLC analysis revealed that this debittering effect is accompanied by a significant reduction of the number of hydrophobic peptides present.  相似文献

Beer contains a very complex mixture of nutrients, which in this work are identified to some extent by high-field high-resolution nuclear magnetic resonance (NMR) one- and two-dimensional methods. The (1)H NMR spectrum of beer shows a predominance of strongly overlapped peaks arising from several carbohydrates. Minor components are clearly observed both in the aliphatic and in the aromatic regions of the spectrum. With the aid of two-dimensional methods, spectral assignment was carried out, enabling the identification of approximately 30 compounds and identifying about the same number of spin systems for further assignment. The variability of the spectral profile of beers differing in type and label was studied by principal component analysis (PCA), and it was found that, although some distinction is achieved on the basis of the aliphatic and sugar compositions, clearer separation between ales and lagers is obtained by PCA of the aromatic profiles alone. The potential of this technique as a rapid and informative quality control tool is discussed.  相似文献
