Phlorotannins are polyphenolic compounds in marine alga, especially the brown algae. Among numerous phlorotannins, dieckol and phlorofucofuroeckol-A (PFF-A) are the major ones and despite a wider biological activity profile, knowledge of the G protein-coupled receptor (GPCR) targets of these phlorotannins is lacking. This study explores prime GPCR targets of the two phlorotannins. In silico proteocheminformatics modeling predicted twenty major protein targets and in vitro functional assays showed a good agonist effect at the α2C adrenergic receptor (α2CAR) and an antagonist effect at the adenosine 2A receptor (A2AR), δ-opioid receptor (δ-OPR), glucagon-like peptide-1 receptor (GLP-1R), and 5-hydroxytryptamine 1A receptor (5-TH1AR) of both phlorotannins. Besides, dieckol showed an antagonist effect at the vasopressin 1A receptor (V1AR) and PFF-A showed a promising agonist effect at the cannabinoid 1 receptor and an antagonist effect at V1AR. In silico molecular docking simulation enabled us to investigate and identify distinct binding features of these phlorotannins to the target proteins. The docking results suggested that dieckol and PFF-A bind to the crystal structures of the proteins with good affinity involving key interacting amino acid residues comparable to reference ligands. Overall, the present study suggests α2CAR, A2AR, δ-OPR, GLP-1R, 5-TH1AR, CB1R, and V1AR as prime receptor targets of dieckol and PFF-A.  相似文献

Despite high expectations of nongovernmental organizations (NGOs) as project partners in bilateral-aid community forestry (CF) projects in Nepal, it remains unclear whether the NGO involvement favored by donors is efficient. This study examined the influence of NGOs on local people's perceptions of CF management by using a quantitative analysis based on a questionnaire survey. Three different forest user groups (FUGs) supported by the Nepal Swiss Community Forestry Project were selected from the Dolakha and Ramecchap districts: an FUG supported by another FUG acting as an NGO, an FUG supported by an NGO specializing in CF, and an FUG without NGO support. A structured survey examined the perception of CF management based on satisfaction, involvement, and self-reliance in 244 selected households belonging to the three FUGs. Knowledge of CF and personal information was also surveyed and analyzed. The results showed that NGO involvement influences factors such as self-reliance and knowledge of CF. FUG members who were supported by an NGO or an FUG had a significantly higher perception with respect to self-reliance and knowledge than did members without such support. This indicates that experienced FUGs can serve as adequate service providers for the CF formation procedure, in a manner similar to that of NGOs. On the other hand, the limited support from NGOs or FUGs during the formation process seemed insufficient to fulfill the expectations of aid agencies with respect to the enhancement of satisfaction and active participation on the part of FUG members. If aid agencies expect NGOs and FUGs to enhance the satisfaction and participation of members, these intermediary organizations should be assigned to support more CF activities than just the formation procedure. Aid agencies should take responsibility for strengthening the capabilities of NGOs and FUGs, as needed.  相似文献

Vast areas of dry subtropical evergreen forests of Olea cuspidata in the northwestern mountains of Nepal have been abandoned in recent decades. The status of this species was found to be very poor in almost all the areas of our study sites, located in Bajura district. However, the lack of past information on this species did not allow us to determine the exact number of hectares that have declined. Overgrazing, illegal cuttings, and lack of environmental awareness were found to be the major causes of Olea spp. forest degradation. Direct survey and Geographic Information System (GIS) methods were used to create a current distribution map of wild olive. Potential sites based on the sparse population and dense patches of wild olive were still found in Bajura. A large amount of wild olive fruit was found unused in these area. If properly managed, subsistence farming of this fruit can be an income supplement for the poorest and vulnerable local communities. The scope of our study was to map the vulnerable status of one of the International Union for Conservation of Nature (IUCN)-listed wild olive species from the remote far-western district of Bajura. The necessity of an immediate national policy is discussed to develop and consolidate control techniques and ecological restoration strategies based on conservation and sustainable approaches in order to prevent future decline of native plant diversity.  相似文献

Soil enzyme activities and water stable aggregates have been identified as sensitive soil quality indicators, but few studies exist comparing those parameters within buffers, grazed pastures and row-crop systems. Our objective was to examine the effects of these land uses on the activities of selected enzymes (β-glucosidase, β-glucosaminidase, fluorescein diacetate (FDA) hydrolase, and dehydrogenase), proportion of water stable aggregates (WSA), soil organic carbon and total nitrogen content. Four management treatments [grazed pasture (GP), agroforestry buffer (AgB), grass buffer (GB) and row crop (RC)] were sampled in 2009 and 2010 at two depths (0 to 10- and 10 to 20-cm) and analyzed. Most of the soil quality indicators were significantly greater under perennial vegetation when compared to row crop treatments. Although there were numerical variations, soil quality response trends were consistent between years. The β-glucosaminidase activity increased slightly from 156 to 177 μg PNP g−1 dry soil while β-glucosidase activity slightly decreased from 248 to 237 μg PNP g−1 dry soil in GB treatment during 2 years. The surface (0–10 cm depth) had greater enzyme activities and WSA than sub-surface (10–20 cm) samples. WSA increased from 178 to 314 g kg−1 in row crop areas while all other treatments had similar values during the 2 year study. The treatment by depth interaction was significant (P < 0.05) for β-glucosidase and β-glucosaminidase enzymes in 2009 and for dehydrogenase and β-glucosaminidase in 2010. Soil enzyme activities were significantly correlated with soil organic carbon content (r ≥ 0.94, P < 0.0001). This is important because soil enzyme activities and microbial biomass can be enhanced by perennial vegetation and thus improve several other soil quality parameters. These results also support the hypothesis that positive interactions among management practices, soil biota and subsequent environmental quality effects are of great agricultural and ecological importance.  相似文献

Smallholder farmers dominate agriculture in Nepal. These farmers have poor knowledge about agriculture and lack of support for soil management and integrated plant-nutrient systems. Focusing on the importance and need for soil-fertility management, a soil-testing mobile van program has recently been introduced in Nepal by Soil Management Directorate, Hariharbhawan. With the introduction of the mobile lab, famers can get their soil tested for nutrient deficiencies and fertilizer requirements at their doorsteps. Using mobile lab, spatial distributions of chemical properties, including pH, organic matter (OM), total nitrogen (N), available phosphorus (as P2O5), and available potassium (as K2O) were examined in soil samples taken from the 0 to 15 cm depth from selected agricultural fields in eight different districts in the mid-hills and Terai regions of Nepal. Tests conducted on 1,479 soil samples in the soil-testing mobile van revealed the following: the mean soil OM ranged from 0.01 to 1.77%; total N content ranged from 0.01 to 0.08%; mean available P2O5 ranged from 16.47 to 197.82 kg ha?1; and mean available K2O ranged from 84.3 to 422.57 kg ha?1. For each crop to be grown, farmers were provided with individual soil health reports and fertilizer recommendations (rate, amount, and type). This program not only allows scientists and farmers to work closely and share information but also serves as a model for the nation to successfully transfer technology for improving soil health and sustainability.  相似文献
