Tissue culture micropropagation of Douglas-fir 总被引:1,自引:0,他引:1
A procedure was developed for plantlet production from embryos of Douglas-fir [Pseudotsuga menziesii (Mirb.) Franco]. The Medium for Conifer Morphogenesis, used at full strength, and supplemented with 10 M benzyladenine for 17 days produced an average of 6.8 shoots on more than 90% of the embryos. The percentage of shoot-forming embryos as well as the average number of shoots per embryo varied significantly among eight seedlots. For secondary multiplication, 89% of the adventitious shoots produced axillary buds on MCM with 5 M benzyladenine. However, 0.5 M BA was more suitable for the elongation of axillaries. Rooting ranged from 0–87% depending upon the treatment. The highest percentage was obtained with a 7-week incubation in peat:perlite containing 1/5-strength medium, 1% sucrose and 2.7 M naphthaleneacetic acid, followed by 5 weeks on peat:perlite with 1/5-strength major and minor salts, full iron and organics, 1% sucrose and 0.1% charcoal. 相似文献

A study was conducted to investigate the effect of dough formulation and hydration level on the rheological properties of pasta dough during pasta extrusion. Semolina 100%, whole wheat 100%, and the following mixtures semolina-whole wheat (49:51), semolina-flaxseed flour (90:10), whole wheat-flaxseed flour (90:10), and semolina-whole wheat-flaxseed flour (39:51:10) were the formulations used for the experiments. Dough was hydrated at 30, 32, and 34% moisture content. Pasta was extruded with a capillary and a semi-commercial pasta extruder to determine the apparent viscosity of the dough during extrusion conditions and its relationship to the behavior of the dough during pasta processing. Results showed that non-traditional pasta dough behaved like a shear thinning fluid that can be described by the Power Law model. Increased hydration levels and/or presence of flaxseed flour on the dough formulation decreased the apparent viscosity of the dough, which correlated with extrusion pressure, mechanical energy, and specific mechanical energy that were required to extrude the dough in the pasta extruder. The strong correlations found between the apparent viscosity of the dough and the pasta extrusion parameters indicates the possibility of using a capillary rheometer to determine the appropriate hydration level of ingredient formulations before extruding with a pasta press. 相似文献

A 2-year field study was conducted to evaluate the effect of two organics, farmyard manure and vermicompost, each at three rates (0, 5, 10 t ha?1 and 0, 1, 2 t ha?1, respectively), along with two levels of mineral fertilizer (75% and 100% of recommended dose), on crops yields and soil properties under a wheat–fodder maize cropping sequence. Individual addition of organics at a higher level increased yields of wheat and subsequent maize. Soil microbial biomass carbon was enhanced as both a direct and residual effect with the addition of farmyard manure followed by vermicompost and fertilizer treatments, and also by combined addition of manure with either vermicompost or mineral fertilizer. Farmyard manure increased the availability of soil macro- and micronutrients, whereas vermicompost influenced only the availability of micronutrients at wheat harvest. A residual effect of farmyard manure and mineral fertilizers was found for available N. Meanwhile, the residual status of micronutrients in the soil was either maintained or significantly improved due to organic amendments (Mn and Zn with farmyard manure; Fe and Zn with vermicompost). Interaction of farmyard manure and vermicompost at a higher level benefited the next crop by increasing the yield of fodder maize and improving the availability of P and metals in soil. 相似文献

Summary Studies on the inheritance pattern of bacterial leaf spot (BLS), yellow mosaic (YM) and Cercospora leaf spot (CLS) reactions in crosses of BLS and YM resistant/tolerant but CLS susceptible × CLS resistant but BLS and YM susceptible parents indicated that resistances to BLS and CLS were governed by single dominant genes, whereas YM tolerance was a monogenic recessive character. The studies also indicated that these three genes were inherited independently. Whey protein concentrate solutions (12% w/v, pH 6.65 +/- 0.05) were pressure treated at 800 MPa for 20-120 min and then examined using size exclusion chromatography (SEC), small deformation rheology, transmission electron microscopy, and various types of one-dimensional (1D) and two-dimensional (2D) polyacrylamide gel electrophoresis (PAGE). The pressure-treated samples showed a time-dependent loss of native whey proteins by SEC and 1D PAGE and a corresponding increase in non-native proteins and protein aggregates of different sizes. These aggregates altered the viscosity and opacity of the samples and were shown to be cross-linked by intermolecular disulfide bonds and by noncovalent interactions using 1D PAGE [alkaline (or native), sodium dodecyl sulfate (SDS), and SDS of reduced samples (SDS(R))] and 2D PAGE (native:SDS and SDS:SDS(R)). The sensitivity of the major whey proteins to pressure was in the order beta-lactoglobulin B (beta-LG B) > beta-LG A > bovine serum albumin (BSA) > alpha-lactalbumin (alpha-LA), and the large internal hydrophobic cavity of beta-LG may have been partially responsible for its sensitivity to high-pressure treatments. It seemed likely that, at 800 MPa, the formation of a beta-LG disulfide-bonded network preceded the formation of disulfide bonds between alpha-LA or BSA and beta-LG to form multiprotein aggregates, possibly because the disulfide bonds of alpha-LA and BSA are less exposed than those of beta-LG either during or after pressure treatment. It may be possible that intermolecular disulfide bond formation occurred at high pressure and that hydrophobic association became important after the high-pressure treatment. 相似文献

BACKGROUND: The insulin-like growth factor (IGF) system is implicated in the pathogenesis of diabetes and cardiovascular disease. OBJECTIVE: We report the effects of total energy intake on the IGF system in two populations with markedly different dietary macronutrient intake and cardiovascular event rate. DESIGN, SUBJECTS AND SETTING: Dietary macronutrient intake was measured in a specific Gujarati migrant community in Sandwell, UK (n=205) compared with people still resident in the same villages of origin in India (n=246). Fasting IGF-I, IGF-binding protein (IGFBP)-1 and IGFBP-3, insulin and glucose (0 and 2-hour) were measured. RESULTS: Total energy and total fat intake were higher in UK migrants, as were IGFBP-3 and IGF-I (mean (95% confidence interval): 145.9 (138.1-153.6) vs. 100.9 (94.6-107.3) ng ml(-1); F=76.6, P<0.001). IGFBP-1 was lower in UK migrants (29.5 (25.9-33.0) vs. 56.5 (50.6-62.5) microg l(-1); F=48.4, P<0.001). At both sites, IGF-I correlated positively with total energy (Spearman's rho=0.45, P<0.001) and total fat (rho=0.44, P<0.001) as did IGFBP-3 with total energy (rho=0.21, P<0.05) and fat (rho=0.26, P<0.001). Conversely, in Indian Gujaratis, IGFBP-1 fell with increasing total energy (rho=-0.27, P<0.001) and fat intake (rho=-0.26, P<0.01) but not in UK Gujaratis. Multiple linear regression modelling showed that increasing quartiles of fat intake were associated with higher IGF-I (beta=0.42, P=0.007) independent of age, body mass index, plasma insulin, fatty acids and 2-hour glucose. CONCLUSION: In these genetically similar groups, migration to the UK and adoption of a different diet is associated with marked changes in the IGF system, suggesting that environmental factors profoundly modulate serum concentrations and actions of IGFs. 相似文献
