The main source of Leishmania infantum infection in humans is a naturally infected dog. This study reports on the infectivity to phlebotomine sandflies (Lutzomyia longipalpis) of serologically positive mongrel dogs that differed in clinical status, haematology and humoral responses to immunoglobulin (Ig) G(T) (total anti-Leishmania IgG), IgG(1) and IgG(2) subclasses of antibody to crude antigen of L. infantum. Forty-five female L. longipalpis were allowed to feed directly on the ears of dogs classified as asymptomatic, oligosymptomatic or symptomatic before being dissected five days later. Promastigotes were detected in 88% of the dissected sandflies. The highest rate of infectivity to sandflies was found in symptomatic dogs, followed by oligosymptomatic and asymptomatic animals. The results suggest that dogs naturally infected with L. infantum with higher total IgG and IgG(2) concentrations and lower haematocrit levels were able to infect the highest proportion of L. longipalpis. No correlation was observed between anaemia and the intensity of clinical signs. Symptomatic dogs presented the highest infection rate and intensity of infection.  相似文献

Teak is a very important tropical timber in Ecuador. In 2017, teak samples displaying stem canker symptoms were collected in two provinces of Ecuador. From 11 symptomatic trees, 11 isolates resembling a species of Lasiodiplodia were obtained. All isolates obtained induced stem canker on teak plants after artificial inoculation, confirming them as the cause of the observed canker symptoms. Bayesian inference with concatenated sequences of complete ITS and partial TEF-α and β-TUB gene sequences from two representative isolates clustered teak isolates with other sequences of Lasiodiplodia theobromae available in Genbank. This is the first report of L. theobromae causing stem canker on teak plants in Ecuador.  相似文献

The effects of edible coatings and mild heat shocks on quality aspects of refrigerated broccoli were studied. Minimally processed broccoli was coated with either chitosan or carboxymethyl-cellulose with or without a previous application of a mild heat shock of 1.5 min at 50 °C. Product was packaged in multilayered polyolefin bags and stored at 5 °C for 18 d. Quality parameters such as weight loss, texture, colour, ascorbic acid content, total chlorophyll content, oxygen concentration inside the bags, browning potential, mesophilic aerobic counts, and sensory quality, were evaluated during storage. Edible coatings exhibited a beneficial impact on broccoli quality. The weight loss in uncoated broccoli was found to be between 2 and 5 times higher compared to coated samples. During storage, coated florets from both thermally and non-thermally treated samples, presented higher retention of the (?a*/b*) ratio indicating better green colour retention and a reduced rate of floret yellowing. Chitosan coating always presented the lower ascorbic acid degradation rates (twofold lower compared with control samples). Broccoli texture for uncoated samples increased significantly during storage. However, for carboxymethyl-cellulose coated broccoli a slight increase in texture was observed while for chitosan coated broccoli no significant changes in texture were observed throughout the storage period. After the edible coating application the microbial broccoli load dropped by around 1.5 and 0.9 logarithmic units in chitosan and carboxymethyl-cellulose films, respectively. During storage, the application of chitosan coating significantly reduced total microbial counts in the thermally and non-thermally treated uncoated samples. Among the assayed edible coatings, chitosan effectively maintained quality attributes and extended shelf life of minimally processed broccoli. The single application of a mild heat shock had a measurable influence in reducing weight loss, enzymatic browning in broccoli stems, and in delaying yellowing of broccoli florets. Moreover, chitosan coating combined with a mild heat shock showed the best performance for long-term refrigerated storage of minimally processed broccoli.  相似文献

Purpose

The relative sensitivity of two freshwater invertebrate organisms to the organophosphorus insecticide fenitrothion was assessed by measuring cholinesterase (ChE) activity, a well-known biomarker of both exposure and effect to organophosphorus pesticides. The influence of different concentrations of humic acids (HAs) and particulate matter on fenitrothion bioavailability was assessed in the more sensitive species.

Materials and methods

The selected invertebrates were the dwelling feeding oligochaete Lumbriculus variegatus and the pulmonate gastropod Biomphalaria glabrata. Acute 48-h bioassays were performed exposing organisms to different fenitrothion concentrations. The concentrations that induced 50 % inhibition of enzyme activity (EC50) were calculated. Fenitrothion bioavailability was investigated using different concentrations of commercial HA or particulate matter. Sand and a diverse selection of chromatographic resins that have been proposed as analogues of natural sediments were selected. For these experiments, animals were exposed to a fenitrothion value similar to the EC50.

Results and discussion

The 48-h EC50 values were 12?±?2 and 23?±?3 μg?l?1 for L. variegatus and B. glabrata, respectively. Depending on HA concentration and the characteristics of particles, ChE activity was similar or higher than the value recorded for animals exposed only to the pesticide in aqueous solution.

Conclusion

The results indicated that L. variegatus was the more sensitive species of the two. In this species, fenitrothion bioavailability did not increase due to the presence of either different HA concentrations or particulate matter. The experimental approach may constitute a useful tool to predict the influence of dissolved organic matter and sediment particles on fenitrothion bioavailability and toxicity to non-target aquatic invertebrates.  相似文献

In montane cloud forests (MCF), the main soil organic carbon (SOC) pool is believed to be constituted by organic debris accumulated on soil surface and, to a lesser extent, by the organic fraction associated with the mineral matrix. The vertical distribution of SOC within soil has strong implications on the composition, stabilization and turnover of the soil organic matter (SOM). In ecosystems like MCF, where the climatic and edaphic conditions varied with altitude, the SOM accumulation and stabilization mechanisms possibly respond to these changes. For that reason, we studied the vertical distribution, accumulation and chemical composition of SOM in five montane cloud forest communities located between 1,500 and 2,500?m a.s.l. Two main SOC accumulation patterns were found: one at 1,500, 1,950 and 2,400?m a.s.l., with SOC content gradually decreasing with depth (cumulative); and another at 2,050 and 2,500?m a.s.l. where SOC had a strong maximum in the surface horizon and a less pronounced increase the spodic horizon (eluviation–illuviation pattern). The total SOC pool in soil decreased in inverse relation to altitude from 227?C?ha?1 at 1,500?m a.s.l. down to 143?mg?C?ha?1 at 2,500?m a.s.l. About 40–60?% of total SOC content corresponded to the surficial organic horizon. The chemical fractionation of the SOM denoted in general predominance of the fulvic acid fraction, and high content of humin and humic acid fractions. We considered that the main SOC vertical distribution processes were related to the raw humus accumulation, decomposition in situ, podzolization in the eluviation–illuviation pattern soils mainly.  相似文献

The complete nucleotide sequence of a seedling-yellows-inducing isolate NUagA of Citrus tristeza virus (CTV) was determined. It consisted of 19302 nucleotides and contained 12 open reading frames (ORF) organized identically to those of previously sequenced isolates. This genome is the largest among the CTV genome sequenced so far ; it is 6 nucleotides (nt), 76 nt, 43 nt, and 53 nt longer than that of T36 (quick decline, Florida), VT (seedling yellows, Israel), T385 (mild, Spain), and SY568 (stem pitting, California), respectively. Sequence comparison of NUagA and the other isolates revealed approximately 90% identities throughout the 3′ half of the genome. The 5′ half of the genome was only about 70% identical to that of T36 but still high at about 90% to those of VT, SY568, and T385. Comparison of amino acid sequences on ORF1a encoding polyproteins, the most variable region, reflects the CTV isolate relationship ; NUagA is closely related to VT, SY568, and T385, but distantly related to T36. Three commercially available assays, designed to specifically detect the presence of Mycobacterium avium subsp. paratuberculosis (MAP) in fecal samples by IS900-PCR, were compared with a conventional culture method. Fecal samples from 100 dairy cows were tested. Fifty-four (67.5%) of 80 culture-positive samples were positive for an assay that detects MAP DNA by dot spot hybridization of polymerase chain reaction products (kit A), 48 (60%) were positive by an assay using ethidium bromide staining for agar gel visualization of amplification products (kit B), and 49 (61.3%) were positive by an assay in which amplified products are detected by a colorimetric detection system (kit C). Relative sensitivity of all tests increased in proportion to the presence of MAP in fecal samples. Specificity was 100% based on results from 20 culture-negative samples from an MAP-free herd.  相似文献

In an effort to expand the spectrum of larvicides effective againstLiriomyza huidobrensis (Blanchard), the effect of spinosad was studied on the mortality of the leafminer under laboratory conditions. Bean plants infested with various leafminer stages (egg through third instar) were treated by dipping leaves in a liter of water containing 24, 48 or 96 mg a.i. spinosad or by drenching the soil of plants with 200 ml of water containing 12 mg, 24 mg or 48 mg a.i. spinosad. In general, leaf dipping was more efficacious and adverse effects were observed sooner than with soil drench. All concentrations of spinosad significantly reduced the number of adults that emerged except leaf dip of third instar; only the highest concentration caused significant reduction of pupae and adults.
