Lung and serum samples from pigs that died or were emergency-slaughtered in a pooled, conventional fattening herd were examined to survey Actinobacillus pleuro-pneumoniae infection and to compare the sensitivity of different testing methods. A total of 110 lungs were used for cultural isolation of the agent and direct immunofluorescence (IF) of impression smears. Boiled lung suspensions were tested by coagglutination (Co-A) and agar gel precipitation (AGP). Eighty-seven sera were tested along with lung samples from the same pigs. The lungs yielded a varied bacterial flora most often containing Pasteurella multocida and less frequently Actinomyces (Corynebacterium) pyogenes, E. coli and Salmonella. A. pleuropneumoniae was isolated from 30 lungs: from 22 lungs it grew out in pure culture, from 7 as mixed culture with P. multocida and from 1 as mixed culture with A. pyogenes. The number of positive samples obtained by the different methods was as follows: coagglutination test (with boiled lung suspensions): 63 (57.3%); immunofluorescence: 43 (39.2%); AGP test (with serum): 31 (35.6%); AFP test (with boiled lung suspension): 25 (22.7%). A total of 23 samples (20.7%) were negative by all serological tests and by cultural isolation. Most samples gave positive results by two or more tests while 26 samples only by one test (most often, on 13 occasions, by the Co-A test). The Co-A test detected antigenic components of serotypes that have not been isolated in Hungary so far. This indicates that it is not enough to test one strain from a given lung sample: several colonies must be cultured and serotyped.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A disorder of central nervous white matter in Norwegian-bred silver foxes is described from the case histories of 21 clinically affected foxes. The main presenting sign of this disorder was caudal limb ataxia, which appeared between 2 1/2 and 4 months of age and progressed over the next 4-8 weeks. Only four affected foxes were allowed to live beyond this period, but they showed moderate to marked improvement. Light microscopic examination of specimens from 16 affected foxes necropsied between 3 1/2 and 6 1/2 months of age revealed lesions that were restricted to the white matter of brain and spinal cord. The lesions were characterized by a symmetrical spongy change with vacuoles of varying sizes and included significant myelin deficiency. There was a relative preservation of axons and nerve cells and no significant inflammation or vascular reaction. An astrocytic hypertrophy was usually associated with the spongy change. Ultrastructural examination of central nervous tissue from two, perfusion-fixed, 6-month-old foxes showed intramyelin vacuoles resulting from splitting of the myelin lamellae at the intraperiod line and was interpreted as indicating myelin edema. Expanded extracellular spaces and watery astrocytic processes also contributed to the vacuolar appearance. Astrocytic processes in affected areas were hypertrophic and contained abundant filaments. Although the 16 silver foxes had severe clinical signs, their lesions had features in common with the juvenile form of Canavan's disease in children and a spongy degeneration reported in Labrador Retrievers; however, the clinical course in the foxes was not uniformly progressive.  相似文献

Responses to a single or repeated infection with 7000 infective larvae of Haemonchus contortus were studied in an experiment using a total of 106 3-month-old lambs with AA, AB or BB haemoglobin (Hb) genotypes. Results were assessed by faecal egg counts, adult worm counts, haematocrit values, haemoglobin concentrations, total serum protein and serum antibody IgG1 and IgA ELISA titres. None of these parameters showed a strong relationship to the Hb type. The prevalence of low responder (greater than 500 worms) and of high responder (less than 50 worms) animals in groups AA, AB and BB Hb types was 3.8 and 34.6, 20.6 and 35.2, 28.1 and 43.7%, respectively, suggesting that the responsiveness to nematode infection is under the control of gene(s) not closely linked with those determining the Hb genotype. Worm counts of a primary infection are more subject to variation than those of a secondary infection. There is a strong relationship between adult worm counts and faecal egg counts taken close to the time of slaughter. In living animals low and high responder discrimination can be based on individual faecal egg counts around 50 days after a secondary infection. Haematocrit values proved to be of little value in the low and high responder selection. In this regard neither Hb concentration nor total serum protein values are of practical significance. In 3-month-old lambs primary infection induced partial immunity which could prevent the establishment of a part of the secondary infection, irrespective of the presence or absence of the primary worm population. The development of immunity was not associated with an increase of serum IgG1 and IgA antibody levels. Specific antibody production was not influenced by Hb types. Mean antibody levels of low responder lambs showed no difference from those of high responders. Thus, serum IgG1 and IgA levels are of no predictive value in identifying lambs which are genetically resistant to Haemonchus infection.  相似文献

Pulmonary alveolar macrophages are considered to be the main phagocytic cell of the pulmonary defense mechanism. However recent studies indicate that neutrophils may also participate in the defense against inhaled bacteria. The aim of this investigation was to study in mice the correlation between numbers of phagocytic cells in the bronchoalveolar space and the pulmonary clearance of bacteria. White mice were exposed to aerosols of Pasteurella haemolytica (n = 129) or Staphylococcus aureus (n = 129) in three different experimental replicates. Another group of mice (n = 22) was sham exposed to an aerosol of sterile phosphate buffered solution in a single replicate. Animals were sacrificed at various times postaerosolization. The numbers of neutrophils and alveolar macrophages in lung lavages and the pulmonary bacterial clearance rates were determined and statistically analysed. No significant differences (p greater than 0.05) were observed in the rates of pulmonary clearance between the two genera of bacteria, but P. haemolytica had a significant (p less than 0.05) replicate effect. The number of alveolar macrophages was not significantly affected by either bacteria or phosphate buffered solution. Exposure to P. haemolytica resulted in dramatic, significant (p less than 0.01) but transient increases in neutrophils in the bronchoalveolar space as well as a significant (p less than 0.01) increase in the weights of lung. The correlation between neutrophils and clearance was positive for P. haemolytica but negative for S. aureus. These results indicate that both species of bacteria are rapidly eliminated from the lung despite a rather different cellular response.  相似文献

Pharmacokinetics and cardiopulmonary effects of guaifenesin in donkeys   总被引:2,自引:0,他引:2  
Five donkeys and three horses were given guaifenesin, intravenously, by gravity administration, until recumbency was produced. The time and dose required to produce recumbency, recovery time to sternal and standing were recorded. Blood samples were collected for guaifenesin assay at 10, 20, 30, 40, 50, 60 min, and 2, 3, 4 and 6 h after guaifenesin administration. Serum was analysed for guaifenesin using HPLC and pharmacokinetic values were calculated using a computer software package (RSTRIP). In donkeys, heart and respiratory rates and blood pressures were recorded before and at 5-min intervals during recumbency. Arterial blood samples were collected before and at 5 and 15 min intervals during recumbency for analysis of pH, CO2, and O2. anova was used to evaluate dynamic data, while t -tests were used for kinetic values.
Respiratory rate was decreased significantly during recumbency, but no other significant changes from baseline occurred. The mean (±SD) recumbency dose of guaifenesin was 131 mg/kg (27) for donkeys and 211 mg/kg (8) for horses. Recovery time to sternal (min) was 15 (SD, 11) for donkeys and 34 (SD, 1.4) for horses. Time to standing was 32 min for donkeys and 36 min for horses. Calculation of AUC (area under the concentration–time curve) (μg.h/mL) (dose-dependent variable) was 231 (SD, 33) for donkeys and 688 (SD, 110) for horses. The clearance ( CL ) (mL/h.kg) was 546 (SD, 73) for donkeys, which was significantly different from 313 (SD, 62) for horses. Mean residence time ( MRT ) (h) was 1.2 (SD, 0.1) for donkeys and 2.6 (SD, 0.5) for horses. Volume of distribution V d(area) (mL/kg) was 678 (SD, 92) for donkeys and 794 (SD, 25) for horses. At the rate of administration used in this study, donkeys required less guaifenesin than horses to produce recumbency, but cleared it more rapidly.  相似文献

Nine strains of paramyxovirus isolated from racing pigeons in southern Bohemia, Moravia and western Slovakia in 1983 were identified by the haemagglutination-inhibition test with antisera to seven types of paramyxovirus and three types of influenza A virus as PMV-1, Newcastle disease virus, in all cases. The haemagglutination activity and pathogenicity of the isolates for chicken embryos, chicken fibroblast cultures, and chickens of different age were determined. The mean death time of chicken embryos (MDT/MLD) was 52.8 to 95.4 h, the average being 75.7 h. The intracerebral pathogenicity index (ICPI8) was on an average 1.42 +/- 0.10 (P = 0.95). Experimental infection of chickens at the age of one, two, three and eight weeks did not cause any clinical disease but increased the level of haemagglutination-inhibiting (HI) serum antibodies up to 1 : 256 within three weeks. The course of heat inactivation of pigeon viruses at the temperature of 56 degrees C was practically identical with the inactivation of the velogenic viscerotropic strain California/1082/71. On the basis of the results, the pigeon isolates may be considered the Newcastle disease virus of velogenic viscerotropic type whose pathogenicity for chickens has been reduced to the level of mesogenic strains by long-time passaging in pigeons.  相似文献

The effect of forward (F) and lateral (L) creep grazing, as two possible management alternatives of intensive production systems, on the gastro-intestinal nematode epidemiology of ewes and lambs was studied. Two groups of Romanov x Rasa Aragonesa ewes rearing twins and maintained on an autumn-contaminated pasture at a mean stocking density of 35 ewes ha-1, were used. Measurements were made of the population of infective larvae on the pasture, level of serum pepsinogen, worm eggs in faeces of ewes and lambs, and lambs' growth rate. In addition, post-mortem worm counts from 'indicator' lambs were used to establish the level of infection at each rotational grazing cycle. Two different waves of nematode infection were identified. In both treatments, the over-wintering larvae were responsible for the first outbreak of parasitism which was particularly important for lambs on Treatment F. The second wave of infection apparently came up with several overlapped L3 generations and had different effects on the animals of each group. While early pasture contamination was suffered by the lambs of Treatment F, lambs on Treatment L were not seriously affected until the end of the third grazing cycle (end of May). The different grazing behaviour of lambs in both treatments appeared to be related to the outbreak of parasitism in lambs. The general pattern of liveweight gains was similar for both groups of animals. However, during the first 90 days on pasture lamb growth rate under Treatment L (193 g day-1) was significantly higher than that under Treatment F (164 g day-1). The serum pepsinogen values, worm burdens and liveweight gains indicate that under intensive systems where lateral creep grazing is allowed for lambs, the level of parasite infection is maintained within acceptable limits for the first 90 days on pasture with lambs' growth rate close to their potential. However, the parasitic consequences of grazing under a forward creeping system indicate that anthelmintic drenchings should be used at lambing and at 3-week intervals thereafter during the first 42 days on pasture, after which the risk of contamination from the over-wintering population is over.  相似文献

Edible coatings based on high molecular weight chitosan, pure or combined with methylcellulose or oleic acid, were applied to fresh-cut carrots cv. Nantesa by simple immersion and by applying a vacuum pulse (5 kPa for 4 min). Water vapour resistance, colour, mechanical properties and respiration rates of non-coated and coated samples were determined. Coatings improved sample appearance, since they diminished the occurrence of the white blush during storage. When applied by simple immersion, they neither conferred significant barrier properties nor the preservation of the mechanical properties of fresh-cut carrot samples. In contrast, coating application with a vacuum pulse enhanced all the positive effects, since the resistance of water vapour transmission of the samples was significantly improved, and better preservation of the sample colour and mechanical response during cold storage was obtained. Differences in film composition did not significantly affect the coating behaviour, probably due to the variability induced by different factors when coatings were applied to the carrot surface.  相似文献
