The purpose of this study was to explore the mechanism of by which docosahexaenoic acid (DHA) inhibit the accumulation of adipose tissue lipid in grass carp (Ctenopharyngodon idella). We therefore designed two semi-purified diets, namely DHA-free (control) and DHA-supplemented, and fed them to grass carp (22.19 ± 1.76 g) for 3 and 6 weeks. DHA supplementation led to a significantly lower intraperitoneal fat index (IPFI) than that in the control group by reducing the number of adipocytes but significantly higher adipocyte size (P < 0.05). In the intraperitoneal adipose tissue, the DHA-fed group showed significantly higher peroxisome proliferator-activated receptor (PPAR)γ, CCAAT enhancer-binding protein (C/EBP)α, and sterol regulatory element-binding protein (SREBP)1c mRNA expression levels at both 3 and 6 weeks (P < 0.05). However, the ratio of the expression levels of B cell leukemia 2 (Bcl-2) and Bcl-2-associated X protein (Bax) was significantly lower in the DHA-fed group than in the control group (P < 0.05), and the protein expression levels of the apoptosis-related proteins caspase 3, caspase 8, and caspase 9 were also significantly higher (P < 0.05). Overall, although DHA promotes lipid synthesis, it is more likely that DHA could suppress the lipid accumulation in adipocytes of grass carp by inducing adipocyte apoptosis. 相似文献

A disease caused by Alternaria alternata occurred on the leaves of European pear cultivar Le Lectier in Niigata Prefecture, Japan, and was named black spot of European pear. In conidial inoculation tests, the causal pathogen induced not only small black lesions on the leaves of European pear cultivar Le Lectier, but severe lesions on the leaves of apple cultivar Red Gold, which is susceptible to the A. alternata apple pathotype (previously called A. mali) causing Alternaria blotch of apple. Interestingly, the apple pathotype isolate showed the same pathogenicity as the European pear pathogen. HPLC analysis of the culture filtrates revealed that A. alternata causing black spot of European pear produced AM-toxin I, known as a host-specific toxin of the A. alternata apple pathotype. AM-toxin I induced veinal necrosis on leaves of Le Lectier and General Leclerc cultivars, both susceptible to the European pear pathogen, at 5?×?10?7 M and 10?6 M respectively, but did not affect leaves of resistant cultivars at 10?4 M. PCR analysis with primers that specifically amplify the AM-toxin synthetase gene detected the product of expected size in the pathogen. These results indicate that A. alternata causing black spot of European pear is identical to that causing Alternaria blotch of apple. This is the first report of European pear disease caused by the A. alternata apple pathotype. This study provides a multiplex PCR protocol, which could serve as a useful tool, for the epidemiological survey of these two diseases in European pear and apple orchards. 相似文献

Adult Mesenchymal Stem Cells (MSC) are cells that can be defined as multipotent cells able to differentiate into diverse lineages, under appropriate conditions. These cells have been widely used in regenerative medicine, both in preclinical and clinical settings. Initially discovered in bone marrow, MSC can now be isolated from a wide spectrum of adult and foetal tissues. Studies to evaluate the therapeutic potential of these cells are based on their ability to arrive to damaged tissues. In this paper we have done a comparative study analyzing proliferation, surface markers and OCT4, SOX9, RUNX2, PPARG genes expression in MSC cells from Bone marrow (BMMSC) and Adipose tissue (ASC). We also analyzed the role of Stem Cell Factor (SCF) on MSC proliferation and on ASCs metalloproteinases MMP-2, MMP-9 secretion. Healthy dogs were used as BMMSC donors, and ASC were collected from omentum during elective ovariohysterectomy surgery. Both cell types were cultured in IMDM medium with or without SCF, 10% Dog Serum (DS), and incubated at 38 °C with 5% CO2. Growth of BMMSCs and ASCs was exponential until 25–30 days. Flow citometry of MSCs revealed positive results for CD90 and negative for CD34, CD45 and MCH-II. Genes were evaluated by RT-PCR and metalloproteinases by zymografy. Our findings indicate morphological and immunological similarities as well as expression of genes from both origins on analyzed cells. Furthermore, SCF did not affect proliferation of MSCs, however it up-regulated MMP-2 and MMP-9 secretion in ASCs. Standing and downed deadwood at different stages of decay provides a crucial habitat for a wide range of organisms. It is particularly abundant in unmanaged forests, such as strictly protected areas of national parks and nature reserves. The present work used the available data for such sites in Poland, analyzing a total of 113 studies concerning 79 sites to determine the causes contributing to variation in deadwood volume based on the duration of conservation, changes in deadwood volume over time (for those sites which were examined multiple times), elevation above sea level, forest type, stage of forest development, input of dead trees from the years preceding deadwood measurements, live tree volume, and the proportion of downed to standing deadwood). Depending on species composition and site altitude, most tree stands fell into one of four categories: subalpine spruce forests, montane beech-fir forests, low altitude beech-fir forests, or oak-hornbeam and riparian forests. The mean deadwood volume for all forest types amounted to 172.0 m3/ha. The mean volume of deadwood in montane beech-fir forests (223.9 m3/ha) was statistically significantly greater than in the other three forest types, for which it ranged from 103.5 to 142.5 m3/ha. A direct effect of the duration of conservation on deadwood volume was not identified. Nevertheless, analysis of repeated measurements on the same sample plots at 10-year intervals showed a consistent rise in mean deadwood volume. A linear regression model for all the analyzed factors reported from montane beech-fir forests and subalpine spruce forests showed that in addition to site altitude, another statistically significant variable was the input of dead trees (R2?=?63.54%). 相似文献

An individual-based distance-dependent modeling approach was used to model the growth of dbh and to evaluate neighborhood effects for four species groups in Mexican mixed-species stands.Methods
Twenty-two species were classified into four groups: Pinus (seven species), other conifers (three species), other broadleaves (four species), and Quercus (eight species). Four methods were used to select neighboring trees and 12 competition indices (CIs) were calculated. Comparisons of the neighboring trees selection methods and CIs and tests of assumptions about neighborhood effects were conducted.Results
Intra-species-group competition significantly reduced diameter growth for all species groups, except for the Quercus group. The Pinus, other conifers, and Quercus groups had significant and negative neighborhood effects on the other broadleaves species group, and not vice versa. The Quercus group also had negative neighborhood effect on the Pinus and other conifers species groups, and not vice versa. The Pinus and other conifers species groups had negative neighborhood effects on each other. All fitted age-independent dbh growth models showed a good of fit to the data (adjusted coefficient of determination larger than 0.977).Conclusion
The growth models can be used to predict dbh growth for species groups and competition in mixed-species stand from Durango, Mexico.958.
In this work, dopamine hydrochloride, an environmental friendly compound, was applied on polyester fabric through conventional simple impregnation method in alkaline solution (pH=8.5) at room temperature. In situ spontaneous oxidative polymerization of dopamine form polydopamine (PDA) along with aminolysis of polyester fabric surface. Also, a range of colored polyester fabric were successfully achieved by formation of polydopamine adhesive coating layer at different concentration of dopamine hydrochloride (0.001-4 g/l). Fourier transform infrared spectroscopy and field emission scanning electron microscopy showed deposition of polydopmaine on the polyester fabric surface. The modified colored polyester fabric showed reasonable durability against washing, rubbing and light. The treated polyester fabric with 2 g/l dopamine hydrochloride as optimum concentration indicated not only lower spreading time for water droplet and electrical resistance with higher tensile strength but also very good bactericidal activity against Staphylococcus aureus and Escherichia coli. 相似文献

ABSTRACT: Hermatypic (reef-building) corals harbor dinoflagellate endo-symbionts Symbiodinium spp. In studying gene expression in such symbiotic corals, problems arise regarding how to distinguish the coral and symbiont mRNA, and how to estimate their fractions in the mRNA population of the holobiont (symbiotic complex of the coral and Symbiodinium cells). In this study, these issues were addressed using juveniles of hermatypic coral Acropora tenuis in symbiosis with Symbiodinium cells of strain PL-TS-1. First, the guanine-cytosine (GC) contents were determined in expressed sequence tags (EST) from PL-TS-1 cells cultured in vitro and symbiont-free larvae of A. tenuis , and their average GC contents were found to be significantly different. The average GC content of the EST from the holobiont was much closer to that of A. tenuis larvae, suggesting that the majority (>90%) of mRNA isolated from the holobiont originated in the host. In protein-coding sequences, little overlap was observed between the GC-content distributions of PL-TS-1 cells and A. tenuis larvae. All of the coding sequences ( n = 59) found in the A. tenuis EST had GC contents below 0.5, whereas the GC content exceeded 0.5 in the majority (43/44) of coding sequences from the nuclear genome of PL-TS-1 cells. 相似文献

Host immune responses conducted against antigens of Eimeria bovis are key factors for the development of protective immunity against this protozoan disease. In this study we investigated
the expression of E. bovis-derived antigens on the host cell surface membrane during E. bovis first merogony in vitro. Host cells carrying E. bovis-meront I stages expressed E. bovis host cell surface antigens (EbHCSAg) on their surface membrane which were recognised by hyperimmune sera of calves and by
sera from rats immunized with E. bovis merozoites I, when tested by indirect immune fluorescent antibody test (IIFAT), laser scanning confocal microscopy (LSCM)
and immune electron microscopy. Expression of EbHCSAg on permissive host cells was earliest detected 7 days p. i., thus coinciding
with the onset of the parasite replication. Membrane-associated EbHCSAg were removed from infected host cells by proteinase
K, partially by Triton X-100, Triton X-114 and Triton X-405, but not by 1 M NaCl, CHAPS or phospholipase C treatment. Antibodies,
affinity-purified on paraformaldehyde/glutardialdehyde (PAGA)-fixed E. bovis meront I-infected bovine host cells bound to the surface meront I-carrying cells and to merozoites I (IIFAT, LSCM) but, in
contrast to untreated sera, not to sporozoites. When tested on methanol-fixed merozoites I and sporozoites by IIFAT, affinity-purified
antibodies bound to structures in the apical complex area of merozoites I, but not to sporozoites, whilst untreated sera caused
diffuse labelling of internal structures of both parasite stages. Immune electron microscopy demonstrated binding of affinity-purified
antibodies to micronemes and dense granules of merozoites I. Although the function of EbHCSAg is still unknown, results of
