Maize prolamin (zein), together with starch, hydroxypropyl methylcellulose, sugar, salt, yeast and water can form wheat-like cohesive, extensible, viscoelastic dough when mixed above room temperature (e.g. 40 °C). This dough is capable of holding gas. However, it is excessively extensible, and when used for hearth-type rolls, it tends to become flat. Bench-scale defatting of zein with chloroform at room temperature significantly (P < 0.05) improved specific volume (4.5 ml/g vs. 3.3 ml/g) and shape of the rolls (width-to-height 2.0 vs. 3.9). The total lipid content determined by accelerated solvent extraction (100 °C, 69 bar, chloroform), however, only decreased from 8.0 to 6.6% due to this bench-scale defatting. Staining experiments with Naphthol Blue Black suggested that bench-scale defatting removed surface lipids from the zein particles, and thus facilitated their aggregation. Aggregation experiments with zein and water at 40 °C, and laser scanning confocal microscopy with zein-starch dough confirmed that zein particles aggregated more easily when surface-defatted. Dynamic oscillatory temperature sweeps demonstrated that surface-defatting lowered the temperature at which protein cross-linking occurred by 2 °C. This research can help to produce superior gluten-free bread and could also possibly contribute to the better understanding of wheat dough.  相似文献

Nitrogen (N) fertilizer represents a significant cost for the grower and may also have environmental impacts through nitrate leaching and N2O (a greenhouse gas) emissions associated with denitrification. The objectives of this study were to analyze the genetic variability in N-use efficiency (grain dry matter (DM) yield per unit N available from soil and fertilizer; NUE) in winter wheat and identify traits for improved NUE for application in breeding. Fourteen UK and French cultivars and two French advanced breeding lines were tested in a 2 year/four site network comprising different locations in France and in the UK. Detailed growth analysis was conducted at anthesis and harvest in experiments including DM and N partitioning. Senescence of either the flag leaf or the whole leaf canopy was assessed from a visual score every 3-4 days from anthesis to complete canopy senescence. The senescence score was fitted against thermal time using a five parameters monomolecular-logistic equation allowing the estimation of the timing of the onset and the rate of post-anthesis senescence. In each experiment, grain yield was reduced under low N (LN), with an average reduction of 2.2 t ha−1 (29%). Significant N × genotype level interaction was observed for NUE. Crop N uptake at harvest on average was reduced from 227 kg N ha−1 under high N (HN) to 109 kg N ha−1 under LN conditions while N-utilization efficiency (grain DM yield per unit crop N uptake at harvest; NUtE) increased from 34.0 to 52.1 kg DM kg−1 N. Overall genetic variability in NUE under LN related mainly to differences in NUtE rather than N-uptake efficiency (crop N uptake at harvest per unit N available from soil and fertilizer; NUpE). However, at one site there was also a positive correlation between NUpE and NUE at LN in both years. Moreover, across the 2 year/four site network, the N × genotype effect for NUpE partly explained the N × genotype effect for grain yield and NUE. Averaging across the 16 genotypes, the timing of onset of senescence explained 86% of the variation in NUtE amongst site-season-N treatment combinations. The linear regression of onset of senescence on NutE amongst genoytpes was not significant under HN, but at three of the four sites was significant under LN explaining 32-70% of the phenotypic variation amongst genotypes in NutE. Onset of senescence amongst genotypes was negatively correlated with the efficiency with which above-ground N at anthesis was remobilized to the grain under LN. It is concluded that delaying the onset of post-anthesis senescence may be an important trait for increasing grain yield of wheat grown under low N supply.  相似文献

Golden retriever and Labrador retriever muscular dystrophy are inherited progressive degenerative myopathies that are used as models of Duchenne muscular dystrophy in man. Thoracic lesions were reported to be the most consistent radiographic finding in golden retriever dogs in a study where radiographs were performed at a single-time point. Muscular dystrophy worsens clinically over time and longitudinal studies in dogs are lacking. Thus our goal was to describe the thoracic abnormalities of golden retriever and Labrador retriever dogs, to determine the timing of first expression and their evolution with time. To this purpose, we retrospectively reviewed 390 monthly radiographic studies of 38 golden retrievers and six Labrador retrievers with muscular dystrophy. The same thoracic lesions were found in both golden and Labrador retrievers. They included, in decreasing frequency, flattened and/or scalloped diaphragmatic shape (43/44), pulmonary hyperinflation (34/44), hiatal hernia (34/44), cranial pectus excavatum (23/44), bronchopneumonia (22/44), and megaesophagus (14/44). The last three lesions were not reported in a previous radiographic study in golden retriever dogs. In all but two dogs the thoracic changes were detected between 4 and 10 months and were persistent or worsened over time. Clinically, muscular dystrophy should be included in the differential diagnosis of dogs with a combination of these thoracic radiographic findings.  相似文献

The effect of Fasciola hepatica excretory-secretory products (FhESPs) on mitogen-induced proliferation of sheep peripheral blood mononuclear cells (PBMCs) and PBMC subsets (CD2(+), CD4(+), CD8(+), gammadeltaTCR(+) or CD21(+) cells) were studied. PBMCs were incubated with Concanavalin A (ConA) or phytohemagglutinin (PHA) at optimal (1 microg per well) or suboptimal (0.25 microg per well) doses and with FhESPs at several doses (1.25-20 microg per well). PBMC subsets were incubated with ConA at a suboptimal dose and with FhESPs at 5 microg per well. These cells were incubated with or without monocytes (CD14(+) cell). FhESPs slightly increased the proliferation of PBMCs stimulated with optimal doses of PHA. FhESPs (10 and 20 microg per well) inhibited the PBMCs stimulated with optimal doses of ConA. FhESP dose-dependent inhibition was observed on PBMCs stimulated with suboptimal doses of ConA. CD21(+) lymphocytes (B lymphocytes), CD14(+) cells (monocytes) and gammadeltaTCR(+) cells were not stimulated by ConA. T lymphocyte subsets (CD2(+), CD4(+) or CD8(+) cells) proliferation was decreased by FhESPs at 5 microg per well. FhESPs inhibits the ConA-induced stimulation of sheep PBMCs and sheep T lymphocyte subsets. Further studies should be done to investigate the mechanism of this FhESP immunomodulatory effect.  相似文献

The resistance of several white poplar-clones against Pollaccia radiosa. Continuing the work of Weisgerber (1967,1968 and 1969) clone-typical forms of resistance against spring defoliation (shoot-dieback), caused by Pollaccia radiosa were investigated in several Leuce-poplarclones. The most obvious differences in attack occur under natural conditions. Respiratory diseases, including inflammatory airway disease (IAD), viral and bacterial infections, are common problems in exercising horses. The airway epithelium constitutes a major physical barrier against airborne infections and plays an essential role in the lung innate immune response mainly through toll-like receptor (TLR) activation. The aim of this study was to develop a model for the culture of equine bronchial epithelial cells (EBEC) in vitro and to explore EBEC innate immune responses in trained horses. Bronchial epithelial biopsies were taken from 6 adult horses during lower airway endoscopy. EBEC were grown in vitro by an explant method. The innate immune response of EBEC was evaluated in vitro by treatment with TLR ligands. TLR3 is the most strongly expressed TLR at the mRNA level in EBEC and stimulation of EBEC with Poly(I:C), an analog of viral dsRNA, triggers a strong secretion of IFN-β, TNF-α, IL-6 and CXCL8. We further evaluated the EBEC innate immune response in horses that underwent a 4-month-training program. While training had no effect on TLR mRNA expression in EBEC as well as in bronchial biopsies, it increased the production of IFN-β after stimulation with a TLR3 ligand and decreased the secretion of TNF-α and IL-6 after stimulation with a TLR2 and TLR3 ligand. These findings may be implicated in the increased risk for viral and bacterial infections observed in sport horses. Altogether, we report a successful model for the culture of EBEC that can be applied to the investigation of pathophysiologic conditions in longitudinal studies.  相似文献
