Ilimaquinone (IQ), a metabolite found in marine sponges, has been reported to have a number of biological properties, including potential anticancer activity against colon cancer. However, no clear understanding of the precise mechanism involved is known. The aim of this study was to examine the molecular mechanism by which IQ acts on HCT-116 cells. The anticancer activity of IQ was investigated by means of a cell viability assay followed by the determination of induction of apoptosis by means of the use of acridine orange–ethidium bromide (AO/EB) staining, Annexin V/PI double staining, DNA fragmentation assays, and TUNEL assays. The mitochondrial membrane potential (ΔΨm) was detected using the JC-1 staining technique, and the apoptosis-associated proteins were analyzed using real-time qRT-PCR. A molecular docking study of IQ with apoptosis-associated proteins was also conducted in order to assess the interaction between IQ and them. Our results suggest that IQ significantly suppressed the viability of HCT-116 cells in a dose-dependent manner. Fluorescent microscopy, flow cytometry, DNA fragmentation and the TUNEL assay in treated cells demonstrated apoptotic death mode. As an additional confirmation of apoptosis, the increased level of caspase-3 and caspase-9 expression and the downregulation of Bcl-2 and mitochondrial dysfunction were observed in HCT-116 cells after treatment with IQ, which was accompanied by a decrease in mitochondrial membrane potential (ΔΨm). Overall, the results of our studies demonstrate that IQ could trigger mitochondria-mediated apoptosis as demonstrated by a decrease in ΔΨm, activation of caspase-9/-3, damage of DNA and a decrease in the proportion of Bcl-2 through the mitochondrial-mediated apoptosis pathway. 相似文献

von Willebrand factor A (vWA) genes are well characterized in humans except for few BONZAI genes, but the vWA genes are least explored in plants. Considering the novelty and vital role of vWA genes, this study aimed at characterization of vWA superfamily in rice. Rice genome was found to have 40 vWA genes distributed across all the 12 chromosomes, and 20 of the 40 vWA genes were unique while the remaining shared large fragment similarities with each other, indicating gene duplication. In addition to vWA domain, vWA proteins possess other different motifs or domains, such as ubiquitin interacting motif in protein degradation pathway, and RING finger in protein-protein interaction. Expression analysis of vWA genes in available expression data suggested that they probably function in biotic and abiotic stress responses including hormonal response and signaling. The frequency of transposon elements in the entire 3K rice germplasm was negligible except for 9 vWA genes, indicating the importance of these genes in rice. Structural and functional diversities showed that the vWA genes in a blast-resistant rice variety Tetep had huge variations compared to blast-susceptible rice varieties HP2216 and Nipponbare. qRT-PCR analysis of vWA genes in Magnaporthe oryzae infected rice tissues indicated OsvWA9, OsvWA36, OsvWA37 and OsvWA18 as the optimal candidate genes for disease resistance. Marble goby (Oxyeleotris marmorata Bleeker), with its high demand and price, has a great potential as a profitable commercial aquaculture candidate in Malaysia and Southeast Asia region. Efforts are being made to produce this species in a better controlled culture environment like recirculating aquaculture system (RAS) due to poor growth performance and disease problems shown by conventional cage and outdoor pond culture systems. Quantification of waste excreted by fish is critical to RAS design. This study was conducted to characterize the waste excretion rates of marble goby fed with different diets (live food and minced fish). Ammonia-N (TAN), urea-N, nitrite-N (NO2-N), nitrate-N (NO3-N), total-N (TN), organic-N (ON), feces-N, 5-day biochemical oxygen demand (BOD5) and total suspended solid (TSS) produced from marble goby were determined over a 72-h excretion period. Under given experimental conditions, the results showed that feed type had significant influence on the waste excretion rates, with marble goby fed live tilapia (Oreochromis niloticus) exhibiting significantly (P < 0.05) the lowest amount of waste excretion comparable to that of fish fed live common carp (Cyprinus carpio) and minced scads (Decapterus russellii). This indicates that feeding marble goby with tilapia poses less adverse effects on water quality and is thus a suitable diet for this species. The waste excreted by the fish is composed of nitrogenous excretion (TAN, Urea-N, ON, Feces-N), and productions of dissolved biodegradable organic substances (BOD5) and TSS (TSSfeces + TSSwater). About 58-71% of the nitrogen consumed in food was excreted and its rate depended mainly on the feed type. TAN was the chief end-product of protein metabolism; about 74-84% of the daily total nitrogenous excretion was TAN. Urea-N accounted for 13-21% of the daily total nitrogenous excretion indicating that urea-N is an important nitrogenous excretory end-product in marble goby. The waste excretion data presented in this study can be served as a pre-requisite for designing a RAS for this species. The overall BOD5 and TSS production found in this study also point to the need for including bio-filtration unit and suspended solids removal mechanism in the RAS design. 相似文献

Artemisia annua L. is an aromatic-antibacterial herb that destroys malarial parasites, lowers fever, and checks bleeding, and from which
the secondary compound of interest is artemisinin. It has become increasingly popular as an effective and safe alternative
therapy against malaria, and its derivatives are effective against multidrug resistant Plasmodium falciparum strains. Nitrogen is required by the plants in the largest quantity and is most limiting where biomass production is desired.
On the other hand, gibberellic acid plays an important role in flowering, growth, and development and also in other physiological
and biochemical processes. The feasibility of foliar GA3 (75 mg·L−1) alone or with varying levels of soil applied nitrogen (40, 80, and 120 mg·kg−1 soil) was tested on A. annua in the present study. The application of GA3 proved effective in alleviating the growth, photosynthesis, and enzyme activities of A. annua. However, N levels combined with GA3 showed better responses, and further improvement in these parameters was observed. Furthermore, the most important task we

The dietary folic acid requirement of fingerling Catla catla (3.4 ± 0.17 g; 7.6 ± 0.41 cm) was evaluated by feeding casein–gelatin‐based isonitrogenous (350 g/kg crude protein) and isocaloric (16.72 kJ/g GE) diets containing different concentrations of folic acid (0, 0.2, 0.4, 0.6, 0.8, 1.0, 2.0 mg/kg) to triplicate groups to apparent satiation at 08:00, 12:30 and 17:30 hr for 16 weeks. Absolute weight gain (AWG; 40.07 g/fish), specific growth rate (SGR; 2.25%), feed conversion ratio (FCR; 1.53), protein retention efficiency (PRE; 31.42%) and protein gain (PG; 6.74) improved significantly (p < .05) with increasing folic acid levels up to 0.4 mg/kg diet and then reached a plateau. However, maximum liver folic acid concentration increased up to 0.6 mg/kg diet. Dietary folic acid levels also significantly affected (p < .05) body composition of fish. No significant change (p > .05) in haematological parameters except in fish fed folic acid‐free diet was noted. Antioxidant and immune parameters increased with increasing concentration of dietary folic acid up to 0.4 mg/kg diet. Broken‐line regression analysis of AWG, FCR, PRE, PG, HCT and liver folic acid concentrations of fingerling C. catla against dietary folic acid levels indicated optimum growth, FCR, PRE, PG, HCT and liver folic acid saturation ranging between 0.22 and 0.56 mg/kg diet, respectively. 相似文献

Response Surface Methodology (RSM) via Central Composite Design (CCD) was used to optimize the alkaline hydrolysis of paddy straw to improve ferulic acid extraction. The parameters involved, namely temperature (°C), concentration of NaOH (M) and duration of extraction (h) are screened by Full Factorial Studies (FFD). The result obtained from FFD was 0.518% (5.18 mg/g). The interactions between each parameters involved were studied using Central Composite Design (CCD). Upon optimization, the percentage of ferulic acid improved from 0.518% to 0.817% (8.17 mg/g). The optimum conditions obtained from this study are 125 °C, 3.90 M, 2.30 h for temperature, concentration of NaOH and extraction time, respectively. This result shows a significant improvement than the previous published work. Details of the experimental design, statistical analysis and interpretation are discussed below. 相似文献

Endurance horses are subjected to heightened stress even after several conditioning protocols. Hence, the goal of this study was to examine the effect of age and performance on the hematology, biochemistry, and physical parameters after an endurance race. Eighteen horses aged 6-15 years were grouped into 80-km race category. All the horses were physically examined, and blood samples were collected postrace. After physical examination, the poor (n = 9) and good (n = 9) performance horses were identified. Potassium concentration was significantly higher (P < .05) in the good performance horses; chloride concentration was significantly higher (P < .05) in the poor performance horses. The mean blood glucose concentration of the poor performance horses in all the age-groups was significantly higher (P < .05). The mean blood lactate concentration was significantly lower (P < .05) in the poor performance horses. There was significant effect of performance on heart rate (P < .031), capillary refill time (P < .013), and gut motility (P < .05). Univariate Fisher exact test results were significantly higher for skin recoil (P < .03), mucous membrane (P < .03), and gait (P < .04) in the performance categories. In conclusion, the influence of age was seen in the poor performance category in relation to lactate, but age did not influence other physical, hematological, and biochemical parameters of the endurance horses. Thus, further studies are required to determine whether physical, hematological, and biochemical parameters during training could be used to estimate performance in endurance horses based on age. 相似文献

Flesh characteristics comprising chemical composition (protein, fat and moisture percentages and pH) as well as a sensory attribute (colour) were measured in 1951 fillet samples from two generations of two populations (a selection line, selected for increased harvest weight, and a control line, selected for average breeding values for harvest weight) of the genetically improved farmed tilapia (GIFT) strain of Nile tilapia. These data were jointly analysed with 5331 harvest weight records from three generations to estimate genetic parameters and correlated selection responses. Multiple trait animal models were used in the analyses. The potential for correlated responses was evaluated by estimating heritabilities and genetic correlations between body traits (weight, length, depth and width) and fillet traits (weight and yield) with the above mentioned flesh characteristics. The heritabilities for protein%, fat% and colour were low (0.06–0.11), whereas for moisture% and pH they were moderate (0.15–0.20). Genetic correlations among some flesh characteristics (moisture% with protein%, fat% and pH and with fat%) as well as between body and fillet traits with flesh characteristics, were significantly different from zero but low (?0.34 to 0.31). Correlated responses were evaluated by comparing the least squares means between the Selection and Control lines. Our results indicate that selection for high growth increased harvest weight in the Selection line relative to the Control line but it did not change the flesh characteristics of the GIFT strain. 相似文献
