Groupers are popular aquaculture species in South-East Asia, but their cultivation is affected by infectious disease outbreaks. Mucosa-associated lymphoid tissues provide a first-line defence against pathogens; however, few studies are available relating to cellular or proteomic responses of mucosal immunity in grouper. Skin, gill and intestine were sampled from brown-marbled grouper Epinephelus fuscoguttatus (Forsskål, 1775) at 4 and 96 hr post-infection (hpi) and 7 days post-infection (dpi) following intraperitoneal infection with Vibrio harveyi, and stained with haematoxylin/eosin and Alcian Blue/periodic acid–Schiff. Skin mucus was analysed by 2D-gel electrophoresis, and proteins modulated by the bacterial infection identified. In the infected fish, significant increases in sacciform cells in skin and increased levels of nucleoside diphosphate kinase in mucus were detected at 4 hpi. At 96 hpi, goblet cells containing acidic mucins significantly increased in the intestine, while those containing mixed mucins increased in skin and gills of infected fish. Proteasome subunit alpha type-I and extracellular Cu/Zn superoxide dismutase levels also increased in mucus. Rodlet and mast cells did not appear to respond to the infection. Mucosal tissues of grouper appeared actively involved in response to Vibrio infection. This information may help future research on improving grouper health, production and vaccine development.  相似文献

Clavibacter michiganensis subsp. michiganensis (Cmm) is a quarantine organism in Europe and in many other countries. It is one of the most severe bacterial pathogens affecting tomato. Screening tomato plants for their resistance level to Cmm requires a large amount of space under quarantine conditions and is therefore costly. This project developed a new inoculation protocol on in vitro tomato plants to facilitate a more economic and higher throughput disease screening. A new method using the PathoScreen system was tested to localize green fluorescent protein-tagged Cmm in planta and to quantify the pathogen based on the percentage of corrected GFP (cGFP%). The system was sensitive in detecting the GFP-tagged Cmm in the shoots, but in the roots a high autofluorescence masked detection and thus sensitivity of the assay. The in vitro protocol was tested on several wild relatives of tomato, which were previously screened in a greenhouse assay. The correlation between wilt symptoms in vitro and wilt symptoms in the greenhouse was overall moderate (= 0.6462). The protocol worked well in differentiating the two parents that were used in the mapping studies. This study shows that the in vitro protocol can be efficiently used for resistance breeding in many tomato genotypes.  相似文献

The optimum dietary protein requirement of the Malaysian mahseer (Tor tambroides) fingerlings was determined in this study. In this completely randomized designed experiment, formulated diets of five levels of dietary protein (30, 35, 40, 45 and 50%) were tested on the T. tambroides fingerlings (initial body weight of 5.85 +/- 0.40 g), reared in aquarium fitted with a biofiltering system. The fingerlings were fed twice daily at 5% of biomass. The fingerling body weight and total length was taken at every two weeks. Mortality was recorded daily. The dietary protein had significant effects on the body weight gain and Specific Growth Rate (SGR) of the fingerlings. The body weight gain and SGR of fingerlings fed with the diet with the dietary protein level of 40% was significantly higher (p<0.05) than that of 30, 35 and 50%. The feed conversion ratio of the 40% dietary protein was the significantly lowest at 2.19 +/- 0.163. The dietary protein level of 40% was the most optimum for T. tambroides fingerlings.  相似文献

Improved rice lines were developed frome three parents with the resistance or tolerance to bacterial leaf blight, blast and drought stress, respectively, using single-, double- and three-way crosses. The improved lines were assessed for agro-morphological and yield traits under non-drought stress (NS) and reproductive-stage drought stress (RS) treatments. The mean comparison of traits measured between parent plants and progenies (improved lines) were similar, and there were significant and non-significant differences among the parents and improved lines (genotypes) under NS and RS. Smilarly, there was significant and non-significant differences in the interaction among both parent varieties and improved lines for NS and RS. Cluster and 3D-model of principal component analysis did not generate categorical clusters according to crossing methods, and there were no exclusive crossing method inclined variations under the treatments. The improved lines were high-yielding, disease resistant, and drought-tolerant compared with their parents. All the crossing methods were good for this crop improvement program without preference to any, despite the number of genes introgressed.  相似文献

In practice, progeny and individual palm selection are believed to be the most suitable breeding approach for improvement of quantitative traits in oil palm because their phenotypic expressions are strongly influenced by abiotic factors. Therefore progeny selection approach was applied in this study for the selection of high fresh fruit bunch (FFB) and dwarf oil palm planting materials. Cross between Deli dura and Nigerian pisifera resulted into 34 D × P full sib progenies with 1036 seedlings. For six consecutive years, data were collected on yield and yield component traits, while vegetative traits were recorded once. Bi-parental analysis was carried out using analysis of variance, followed by progenies mean comparison, variance components, heritabilities and cluster analysis. Highly significant (P ≤ 0.01) progeny effect was recorded in this study and this had a pronounced effect on the expression of all the quantitative traits. Progenies performance of FFB varied significantly and it ranged from 166.49 to 220.06 kg/palm/year (kg/p/yr) with trial mean of 192.93 kg/p/yr. Palm height after 8 years of field planting ranged from 1.67 to 2.78 m (control cross) with trial mean of 2.12 m. Broad sense heritability (\({\text{h}}_{\text{B}}^{2}\)) was found to be very low (<17.60%) for all the yield traits, however this parameter was high for vegetative traits with palm height having \({\text{h}}_{\text{B}}^{2}\) of 90%. Cluster analysis based on all the quantitative traits grouped all the 34 DP progenies into nine distinct clusters. From this study, five progenies (DP3, DP4, DP5, DP8 and DP24) were identified to be high yielding and dwarf palms compare to trial mean. At density of 140 palm/ha, they will produce FFB of 28.63–30.81 t/ha and average of 29.69 t/ha which is about 27.15% higher in FFB when compared to the current planting material with FFB of 23.35 t/ha. In addition, the selected progenies possessed average annual palm increment of 29.82 cm/yr with range of 26 and 32.5 cm/yr which was 57.33% shorter than the current planting material with palm height increment of 45–75 cm/yr.  相似文献

Seaweeds are valuable sources of biologically active compounds that could be used as ingredients for pharmacological applications. Industrial fermentation using microorganisms provides a wide array of fermented foods and functional compounds with excellent health benefits. These fermentation-derived natural compounds have the potential to be applied as nutraceuticals and functional foods. Therefore, this paper presents an overview of the fermentation of sustainable seaweeds in producing valuable components, including anticoagulant, antioxidant, antimicrobial, anti-inflammatory, and anticancer. Fermentation-derived compounds are expected to receive more attention due to their environmentally friendly processing as well as consumer desire for natural foods.  相似文献

BACKGROUND: Pesticides are developed with carriers to improve their physicochemical properties and, accordingly, the bioefficacy of the applied formulation. For foliar‐applied herbicide, generally less than 0.1% of the active ingredient reaching the target site could reduce pesticide performance. Recently, a carrier of nanoemulsion consisting of oil, surfactant and water, with a particle size of less than 200 nm, has been shown to enhance drug permeability for skin penetration in pharmaceutical delivery systems. In the present work, the aim was to formulate a water‐soluble herbicide, glyphosate isopropylamine (IPA), using a green nanoemulsion system for a biological activity study against the weeds creeping foxglove, slender button weed and buffalo grass. RESULTS: The nanoemulsion formulations displayed a significantly lower spray deposition on creeping foxglove (2.9–3.5 ng cm?2), slender button weed (2.6–2.9 ng cm?2) and buffalo grass (1.8–2.4 ng cm?2) than Roundup® (3.7–5.1 ng cm?2). The visible injury rates of weeds treated with the nanoemulsion formulations were statistically equivalent to those relating to Roundup® at 14 days after treatment, with a control range of 86.67–96.67%. CONCLUSION: It was hypothesised that the significant difference in spray deposition with equal injury rates can be attributed to enhanced bioactivity of the nanoemulsion formulations. A newly isolated white-rot fungus, Armillaria sp. strain F022, was isolated from the decayed wood in a tropical rain forest. Strain F022 was capable of decolorizing a variety of synthetic dyes, including azo, triphenylmethane, and anthraquinone dyes, with an optimal efficiency of decolorization obtained when dyes added after 96 h of culture, with the exception of Brilliant Green. All of the tested dyes were decolorized by the purified laccase in the absence of any redox mediators, but only a few were completely removed, while others were not completely removed even when decolorization time was increased. The laccase, with possible contributions from unknown enzymes, played a role in the decolorization process carried out by Armillaria sp. F022 cultures, and this biosorption contributed a negligible part to the decolorization by cultures. The effect of dye to fungal growth was also investigated. When dyes were added at 0 h of culture, the maximum dry mycelium weight (DMW) values in the medium containing Brilliant Green were 1/6 of that achieved by the control group. For other dyes, the DMW was similar with control. The toxic tolerance of dye for the cell beads was excellent at least up to a concentration of 500 mg/l. The optimum conditions for decolorization of three synthetic dyes are at pH 4 and 40°C.  相似文献
