Many species of African nonhuman primates are naturally infected with simian immunodeficiency viruses (SIVs) in the wild and in captivity. In contrast to HIV-infected humans, these natural SIV hosts typically do not develop AIDS, despite chronic infection with a highly replicating virus. In this Review, we discuss the most recent advances on the mechanisms of protection from disease progression in natural SIV hosts, with emphasis on how they differ from pathogenic HIV/SIV infections of humans and rhesus macaques. These mechanisms include: (i) resolution of immune activation after acute infection, (ii) restricted pattern of target cell infection, and (iii) protection from mother-to-infant transmission. We highlight the areas that should be pursued in future studies, focusing on potential applications for the treatment and prevention of HIV infection.  相似文献

Insoluble fiber fractions from 11 fruits and vegetables were investigated for their lignin composition using the derivatization followed by reductive cleavage (DFRC) methodology. To enrich lignin contents and to minimize polysaccharide excess that led to nonanalyzable DFRC chromatograms, the insoluble fibers were degraded by a carbohydrolases mixture. The residues that were found to be representative for the insoluble fiber lignins were analyzed. The investigated fibers differ considerably in their lignin contents and also in their lignin compositions. With the exception of radish fiber, only trace amounts (or none) of the products resulting from p-hydroxyphenyl units were detected. Lignins noticeably differed in the ratio of the DFRC products resulting from syringyl units (S) and guaiacyl (G) units (G/S ratios ranged from approximately 39 to 0.2). The insoluble fiber lignins were classified as G-rich lignins (G/S ratio > 3; carrot, spinach, kiwi, curly kale, radish, and asparagus), S-rich lignins (S/G ratio > 3; rhubarb), or balanced lignins (0.3 < G/S ratio < 3; pear, apple, small radish, and kohlrabi). Information about further structural characteristics, for example, cinnamyl endgroups, was obtained from the analysis of DFRC minor products.  相似文献

In Germany the sweet and sour cherry breeding is concentrated at the Julius Kühn-Institut in Dresden-Pillnitz. The main breeding goals are directed on sweet cherries for fresh market and sour cherries for processing. The selection of new cultivars is focused on fruit quality, high and stable fruit set and tolerance to biotic and abiotic stress. In result of the breeding activities in sweet cherry four new cultivars, ‘Narana’, ‘Areko’, ‘Swing’, ‘Habunt’, were selected. ‘Narana’ is an early ripening cultivar with good fruit characteristics. ‘Areko’ ripens in the middle of the cherry season some days before cultivar ‘Kordia’ and has large, firm fruits. The both cultivars ‘Swing’ and ‘Habunt’ are self-compatible and more recommended for the use in house gardening. In sour cherry breeding four new cultivars, ‘Coralin’, ‘Spinell’, ‘Jachim’, ‘Boas’, were selected. The cultivar ‘Coralin’ shows good characteristics for processing and mechanical harvest and is tolerant to fungus diseases on leaves. With the large and tasty fruits, the cultivar ‘Spinell’ is mainly suitable for fresh consumption. The both other cultivars ‘Jachim’ and ‘Boas’ have a pillar growth habit.  相似文献

Clostridium difficile, associated with a wide spectrum of diseases in humans, as well as in several animal species, is an important cause of colitis in adult horses and foals. The aim of this study was to investigate by toxin gene profile and PCR-ribotyping the molecular characteristics of 14 C. difficile strains isolated from 42 faeces of healthy horses. Both toxin genes, tcdA and tcdB, were present in only 1 isolate (7.1%). Six isolates (42.9%) demonstrated tcdA/tcdB+ genotype, and seven isolates (50.0%) were tcdA−/tcdB−. All strains were binary toxin genes negative (cdtA−/cdtB−). The PCR-positive strains, except for the tcdA+/tcdB+ isolate, tested negative for, in vitro, A and/or B toxins production by EIA. Eleven distinct ribotypes were observed.In conclusion, C. difficile can be present in the normal intestinal flora of healthy adult horses, in addition to foals. These animals could therefore play an important role as potential reservoirs of toxigenic strains.  相似文献

Bile acid adsorption by lignified dietary fiber in the human intestine is proposed as a mechanism for lowering blood cholesterol level and reducing colon cancer risk. In this study, we investigated how the concentration and composition of lignin in fiber influences the in vitro adsorption of primary bile acids (glycocholate, taurocholate, and glycochenodeoxycholate) and a secondary bile acid (deoxycholate). Adsorption studies were performed by incubating nonlignified and artificially lignified maize cell walls (dehydrogenation polymer‐cell walls) with bile acids under conditions imitating the small intestine and distal colon. Artificially lignified cell walls had varying but defined lignin concentrations (4.8–19.0%) and compositions (varying from pure guaiacyl to pure syringyl lignins) but a uniform polysaccharide‐protein matrix. Adsorption of bile acids by cell walls was in a range of 6–31% (4–26 nmol of bile acids/mg of cell walls), with glycochenodeoxycholate showing the highest adsorption rates. Neither lignin concentration nor lignin composition influenced bile acid adsorption, thus disproving a major role of lignin in bile acid adsorption.  相似文献

Uranium is a radiotoxic and chemotoxic heavy metal. Uptake and accumulation of U has been studied in plants native to uranium mine sites, but not in cultivated plants which are commonly consumed by humans. The objective of this study was better understanding of U uptake and accumulation in cultivated plants and whether different contents of uranium (U) in the substrate affect its concentration in plants and their dry matter mass. Two substrate variants for growing plants, which differed in the uranium content: solid wastes (tailings) and tailings mixed with sand (w/w 1:1). Large amounts of solid wastes (tailings) resulting from the exploitation and treatment of uranium ore from the closed uranium mine Gabrovnica–Kalna, on southeast of Serbia, contained generally 15.33 mg U/kg. In the experiment, three plant species (corn NSSC 231, sunflower N.S. Dukat, and green peas Smederevska Palanka) were grown in pots on the four substrate variants during 40 days. Substrate was suffused by drinking water (DW) and “uranium water” (UW), which issue out from the mine, contained 0,053 mg U/ dm3. Obtained results show that when UW was added to tailing concentration of U in plants increased. When the content of U in the substrate was lowered by adding sand, the concentration of U in plants decreased, though was significantly higher in comparison to the variants to which DW was added. Dry matter mass was higher in variants where UW was used. Concentration of U was significantly higher in root than in above-ground parts.  相似文献

Valuable information about possible types of linkages, reaction mechanisms, and sequences for oxidative coupling of phenolic compounds in planta is available from in vitro model systems. Ferulate oligomers were generated in a system using ethyl ferulate, peroxidase, and hydrogen peroxide under various conditions. A molar ferulate/H2O2 ratio of 1:1, an ethanol level of 30% in an aqueous sodium phosphate buffer (pH 6.0), and a reaction time of 10 min were considered to be ideal to produce maximal proportions of ferulate trimers and tetramers from ethyl ferulate as starting material. The dominant trimer and tetramer were each isolated from the reaction mixture and identified as 8-O-4/8-5(cyclic)-dehydrotriferulic acid triethyl ester and 8-5(cyclic)/4-O-5/8-5(cyclic)-dehydrotetraferulic acid tetraethyl ester. The structure of the 8-O-4/8-5(cyclic)-dehydrotriferulic acid triethyl ester revealed that a third ferulate unit is bound to a preformed 8-O-4-diferulate dimer, a surprising reaction sequence considering the dominance of 8-5-coupled dimers among dehydrodiferulates in H2O2/peroxidase-based model reactions. As 4-O-5-coupling is not favored in the dimerization process of ferulates, the main tetramer isolated in this study is probably formed by 4-O-5-coupling of two preformed 8-5(cyclic)-diferulates, a logical step in analogy with reactions occurring in lignin biosynthesis.  相似文献

A series of experiments were conducted to obtain an efficient larval rearing protocol for Megabalanus azoricus. The first part of this study investigates the effect of microalgae‐based diets on survival and larval development. Mono and mixed‐diets were tested at 20 ± 1°C, in a sequence of 11‐day feeding experiments. The second part presents a preliminary study on the influence of a biofilm on recruitment and use of oyster spat collectors in a mass rearing system. A photographic record of larval development and a brief reference to the diagnostic features that enable quick larval staging are also presented, along with morphometric measurements. Of the microalgae tested (Chaetoceros sp., Chloromonas sp., Dunaliella sp., T‐Isochrysis sp. and Skeletonema sp.) the mixed‐diet Skeletonema sp. with T‐Isocrysis sp. showed the highest survival percentages: total survival ranged from 79.7 to 85.7% and 69.7–80.0% of nauplii were in stage VI after 11 days of rearing. Cypris were also present, but only represented 5.3% of the survivors at most. In the mass rearing system juveniles were found settled in the collectors after 25 days, at 20 ± 1°C. However recruitment was less than 1%. Preliminary results showed no settlement preference towards collectors with biofilm. Nevertheless, this study provides the first record of M. azoricus settlement under laboratorial conditions and represent a starting point for future larval rearing studies.  相似文献

Arabinogalactans and galactomannans from coffee beverages are part of the dietary fiber complex. Chemical structures and fermentability of soluble dietary fiber obtained from a standard filter coffee beverage (Coffea arabica, origin Colombia, medium roasted) by human intestinal bacteria were investigated. One cup (150 mL) of filter coffee contained approximately 0.5 g of soluble dietary fiber (enzymatic-gravimetric methodology), 62% of which were polysaccharides. The remainder was composed of Maillard reaction products and other nonidentified substances. Galactomannans and type II arabinogalactans were present in almost equal proportions. Coffee dietary fiber was readily fermented by human fecal slurries, resulting in the production of short-chain fatty acids (SCFA). After 24 h of fermentation, 85% of total carbohydrates were degraded. In general, arabinosyl units from the polysaccharide fraction were degraded at a slower rate than mannosyl and galactosyl units. In the process of depolymerization arabinogalactans were debranched and the ratio of (1-->3)-linked to (1-->6)-linked galactosyl residues decreased. Structural units composed of (1-->5)-linked arabinosyl residues were least degradable, whereas terminally linked arabinosyl residues were easily utilized. The impact of coffee fiber on numerically dominant population groups of the intestinal microbiota was investigated by fluorescence in situ hybridization combined with flow cytometry (FISH-FC). After 24 h of fermentation, an increase of about 60% of species belonging to the Bacteroides-Prevotella group was observed. The growth of bifidobacteria and lactobacilli was not stimulated.  相似文献
