Oxidation products of (-)-epicatechin and (-)-epigallocatechin by treatment with homogenates of 62 plants belonging to 49 families were compared. Forty-six plants were capable of synthesizing theaflavin, a black tea pigment, regardless of whether they contained catechins. Loquat, Japanese pear, and blueberry had activities higher than that of fresh tea leaves after 5 h of treatment; furthermore, these plants oxidized theaflavin to theanaphthoquinone. An additional new metabolite, dehydrotheasinensin, was generated on treatment with fresh tea leaves, eggplant, and unripened Japanese orange. Evidence for the oxidation of epigallocatechin and theaflavin by electron transfer to epicatechin quinone was demonstrated in a time course study using bananas and trapping the quinone intermediates as glutathione conjugates.  相似文献

Ovomucoid, an egg protein comprising approximately 10% egg white, was digested using the enzyme pepsin, and fragments were isolated by anion-exchange and reverse phase HPLC. Four distinct fragments were identified by analysis with SDS-PAGE, including three large fragments with molecular weights of around 24, 18, and 14 kDa. N- and C-terminal and amino acid sequencing analyses identified the fragments as V134-C186 (domain 3), V21-A133, and A1-A133 (domain 1+2). Further separation and sequencing of the fraction composed of small peptides, to determine their exact makeup and location in the protein, remained to be carried out and identified a peptide G51-Y73. All four fragments showed IgE-binding activity, as measured by ELISA, using human sera from egg-allergic individuals. Little change in the digestibility of ovomucoid by trypsin and chymotrypsin was observed following digestion with pepsin, indicating that pepsin-digested ovomucoid retains its trypsin (protease) inhibitor activities. Reduced carboxymethylated ovomucoid was prepared, and digestion with pepsin produced significantly more peptides than did the digestion of the native ovomucoid, indicating that the disulfide bonds play a significant role in the digestive resistance of ovomucoid. The reduction of ovomucoid enhanced its digestibility and lower allergenicity of the protein.  相似文献

The immunomodulatory effect of GMP and its derivates on the cell proliferative response of human macrophagelike cell, U937, and its effect on phagocytic activities via incorporation of fluorescence beads were studied. GMP was found to be a potent immunoenhancer at low concentrations, significantly enhancing the proliferation and phagocytic activities of U937. The modulatory function could be radically altered by enzymatic treatments. Pepsin digestion significantly enhanced the degree of cell proliferation and phagocytic activities, whereas trypsin had no significant effect. The immunoenhancing effects decreased significantly after sialidase treatment; however, more than 70% of activity was retained after treatment. GMP with different carbohydrate chains was shown to possess different modulatory capabilities. Sialic acid-rich GMP fractions showed an enhanced response. These findings indicate that both the carbohydrate chains compositions, including the terminal sialic acids and the polypeptide portions of GMP, are essential for the stimulatory effects of GMP on cell proliferation and phagocytic activities of U937.  相似文献

A feeding trial was conducted to determine the interactive effects of vitamin C (ascorbic acid, AsA) and E (α‐tocopherol, α‐Toc) supplementation with dietary oxidized fish oil (OFO) on the growth performance, whole‐body AsA, and α‐Toc concentrations and fatty acid composition of juvenile sea cucumber. In a 9‐wk feeding trial, juveniles (average weight: 0.6 ± 0.1 g) were cultured in twenty‐four 50‐L tanks (30 juveniles per tank) in triplicate, and fed with eight test diets containing two levels of OFO (8.9 and 156.9 meq/kg) with varying levels of vitamin C (500 and 1000 mg AsA equivalents/kg diet) and E (100 and 200 mg α‐Toc equivalents/kg diet) supplementation, respectively. Body weight gain of sea cucumber was significantly reduced by dietary OFO, while mortality and whole‐body thiobarbituric acid‐reactive substances value were increased significantly. Increasing dietary vitamin C and E levels significantly increased whole‐body α‐Toc and AsA concentrations, respectively. A high level of vitamin E combined with OFO led to consumption of AsA. Even with supplementation of a large dose of vitamin C and/or E in diets, growth performance could not be improved, probably due to the high levels of vitamins in the control diets.  相似文献

The emergence of egg allergy has had both industrial and clinical implications. In industrialized countries, egg allergy accounts for one of the most prevalent food hypersensitivities, especially in children. Atopic dermatitis represents the most common clinical manifestation in infancy; however, the range of clinical signs is broad and encompasses life-threatening anaphylaxis. The dominant egg allergens are proteins and are mainly present in the egg white, for example, ovalbumin, ovomucoid, ovotransferrin, and lysozyme. However, egg yolk also displays low-level allergenicity, for example, alpha-livetin. Strict avoidance of the offending food remains the most common recommendation for egg-allergic individuals. Nevertheless, the omnipresence of egg-derived components in prepackaged or prepared foods makes it difficult. Therefore, more efficient preventive approaches are investigated to protect consumers from inadvertent exposure and ensuing adverse reactions. On the one hand, commercial kits have become readily available that allow for the detection of egg contaminants at trace levels. On the other hand, attempts to produce hypoallergenic egg-containing products through food-processing techniques have met with promising results, but the approach is limited due to its potentially undesirable effects on the unique functional and sensory attributes of egg proteins. Therefore, the development of preventive or curative strategies for egg allergy remains strongly warranted. Pilot studies have suggested that oral immunotherapy (IT) with raw or cooked preparations of egg may represent a safe alternative, immediately available to allergic subjects, but remains applicable to only nonanaphylactic patients. Due to the limitations of conventional IT, novel forms of immunotherapy are sought based on information obtained from the molecular characterization of major egg allergens. In the past decade, promising approaches to the treatment and prevention of egg allergy have been explored and include, among others, the production of hypoallergenic recombinant egg proteins, the development of customized peptides, and bacterial-mediated immunotherapy. Nonspecific approaches have also been evaluated, and preliminary trials with the use of probiotic bacteria have yielded encouraging results. The current understanding of egg allergens offers novel approaches toward the making of food products safe for human consumption and the development of efficient immunotherapeutic strategies.  相似文献

Rice sheath blight disease caused by Rhizoctonia solani Kuhn results in significant yield and quality losses in rice growing areas worldwide. The glycolytic pathway is important in the resistance response to R. solani infection in rice. This study examined one of the regulatory steps in this pathway catalyzed by pyrophosphate-fructose-6-phosphate-phosphotransferase (PFP) and 6-phosphofructokinase (PFK). PFP and PFK activity in R. solani-infected rice plants increased. The mRNA expression of PFP/PFK isozymes showed that PFK 1, 2, 4 and 5 in the resistant line at 1 dpi were high as compared to the gradual increase observed in the expression of all PFP isozymes. Also PFK 1, PFK 3, PFK 4, PFK 5, PFP 2 and PFP 5 were adaptive to sheath blight disease infection and linked to defence response while, the expressions of PFK 2, PFP 1, PFP 3 and PFP 4 although adaptive, were not specific to R. solani infection. These observations provide evidence that (a) both PFP and PFK have isozymes that play an adaptive role after R. solani infection but while those of PFK are expressed at higher levels within a short time after infection those of PFP are expressed gradually, (b) the adaptive activation of PFP in R. solani-infected rice plants is correlated with the paired expression of its α- and β-subunits as shown by PFP 2 and PFP 5, and (c) the expression of some α-subunits is not specific to R. solani infection as shown by PFP 1, PFP 3 and PFP 4.  相似文献

We characterized 14 anonymous nuclear loci from Pinus thunbergii Parl., an important pine species native to Japan. One hundred and twenty-six single nucleotide polymorphisms (SNPs) were identified from these loci, giving a frequency of 1 SNP per 51 bp. Nucleotide diversity (θ) ranged from 1.06 × 10-3 to 11.87 × 10-3, with an average of 4.99 × 10-3. Only one locus (mK45) deviated significantly from the Hardy-Weinberg equilibrium. Thirteen of 14 loci were applicable in other pine species. These loci will be useful for nucleotide variation studies and will provide material for SNP-based marker development in P. thunbergii and related species.  相似文献

The aim of this study was to determine the efficacy of several alternative postharvest treatments to sulfur dioxide (SO2) in maintaining quality and reducing fungal decay during cold storage of ‘Red Globe’ table grapes. Grapes were packaged in perforated polyethylene (PPE) or modified atmosphere packaging (MAP) bags (ZOEpac or Antimicrobial) with or without different grades of ethanol vapor-generating sachets (Antimold®30, Antimold®60 or Antimold®80) or a SO2-generating pad, and the grapes were kept at 0 °C and 90-95% relative humidity for 4 months. Packaging of grapes with a SO2 pad in PPE or ZOEpac bags provided better control of fungal decay and stem browning than PPE or ZOEpac bags alone, PPE or ZOEpac bags with Antimold sachets or Antimicrobial bags alone. The PPE bag containing the Antimold®80 sachet was as effective as the SO2 treatments in reducing the incidence of fungal decay in naturally infected and artificially inoculated grapes for 1 month. Ethanol vapor released by the Antimold sachets enhanced berry color, but caused stem browning depending on ethanol vapor concentrations in the headspace of the bags. The ZOEpac-210 bags alone resulted in significantly lower weight loss and incidence of stem browning, compared to other treatments; however, they did not reduce fungal decay or develop an appreciable level of modified atmosphere.  相似文献

N-Chloroacetyl-N-phenylglycine esters were shown to exert various degrees of growth inhibiting activity against young shoots of annual grasses. The activity against the rice plant and barnyard-grass was determined for 58 derivatives where either the aromatic substituent or ester moiety was modified. The structure-activity relationships were analyzed using physicochemical parameters of the molecule such as log P, σ, and Es, and regression analysis. By comparing the correlations derived for the rice plant and barnyard-grass, the selectivity in herbicidal activity is discussed.  相似文献
