Morbillivirus infections have been responsible for mass mortalities in several species of marine mammals. Nevertheless, relatively little is known on the pathogenesis of the disease and the immune response to the agent, especially in cetaceans, hindering the treatment of individuals and the development of appropriate vaccines, given the difficulty of performing experimental work in marine mammals. The reconstitution of severe combined immunodeficient (SCID) mice, which do not have the ability to reject grafts, with lymphocytes from different species has been used with increasing success as a surrogate species model to study the immune system. We injected NOD/SCID mice with lymphocytes from different species of cetaceans and further vaccinated those mice with a commercial canine distemper virus (CDV) vaccine to develop a practical model to study cetacean immune response to a morbillivirus. Reconstitution was detected in 10/20 mice reconstituted with harbor porpoise spleen, 6/10 mice reconstituted with harbor porpoise lymph node cells, 8/10 mice reconstituted with fresh beluga PBMCs and none of the mice reconstituted with neonate bottlenose dolphin spleen or thymus cells when assessed 42-63 days after reconstitution. While a humoral immune response was detected in none of the reconstituted mice, a cell-mediated immune response to the CDV vaccine was detected in 6/15 (40%) and 2/18 (11%) of the SCID mice after reconstitution with cetacean immune cells after a single or booster vaccination, respectively, for a combined total of 8/33 (24%). This represents the first demonstration of successful reconstitution of SCID mice with marine mammal cells, and to the authors' knowledge, the first direct demonstration of a primary antigen-specific cell-mediated immune response in reconstituted SCID mice. This model will be useful for further research on the physiology of the marine mammal immune system and its response to infectious agents and vaccines, with possible important outcomes in conservation issues.  相似文献

Soluble oligomers are common to most amyloids and may represent the primary toxic species of amyloids, like the Abeta peptide in Alzheimer's disease (AD). Here we show that all of the soluble oligomers tested display a common conformation-dependent structure that is unique to soluble oligomers regardless of sequence. The in vitro toxicity of soluble oligomers is inhibited by oligomer-specific antibody. Soluble oligomers have a unique distribution in human AD brain that is distinct from fibrillar amyloid. These results indicate that different types of soluble amyloid oligomers have a common structure and suggest they share a common mechanism of toxicity.  相似文献

When high pressure (250 to 300 kilobars) was applied suddenly (shock-loading) to gabbro, the plagioclase was transformed to a noncrystalline phase (maskelynite) by a solid-state reaction at a low temperature, while the proxene remained crystalline. The shock-loaded gabbro resembles meteorites of the shergottite class; this suggests that the latter formed as a result of shock. The shock-loading of gabbro at 600 to 800 kilobars raised the temperature above the melting range of the plagioclase.  相似文献

Over a 5-yr period, spring-calving cows were used in a carry-over design experiment to evaluate effects of calf age at weaning on cow and calf performance and production economics. Weaning management groups were early (n = 60, calf age 150 d, EW), traditional (n = 60, calf age 210 d, NW), and late (n = 60, calf age 270 d, LW). Cow body condition score (BCS) and weights at the last weaning date were different (P < .05) for EW (5.8, 583 kg), NW (5.5, 560 kg), and LW (5.2, 541 kg) management groups. Pregnancy rates among groups were similar. Days on feed for groups differed (P = .001) and was 247 for EW, 204 for NW, and 164 d for LW steers. Average daily gain in the feedlot differed (P = .01) among groups and averaged 1.5 kg for LW, 1.4 kg for NW, and 1.3 kg for EW steers. Dry matter intake while steers were in the feedlot was greater (P = .001) for LW than for NW and EW calves. Hot carcass weight was greater (P = .01) for EW (328 kg) and NW (332 kg) calves than for LW (321 kg) steers, and fat depth was greater (P = .05) for EW and NW steers than for LW steers. When carcass data for the NW and LW steers were adjusted to the fat depth of EW steers, carcass characteristics among groups were similar. Net income per steer at slaughter for the feedlot phase was greater (P < .001) for the EW ($75.36) and NW ($62.16) steers than for the LW ($10.09) steers. Again, when carcass data for the NW and LW steers were adjusted to the same fat depth of the EW steers, net income differences among groups were reduced. Replacement heifers were developed in a drylot and costs were higher (P < .001) for the EW than for NW and LW heifers. Annual cow costs were greater (P < .10) for the LW ($443.45) than for the EW ($410.09) and NW ($421.35) groups. Break-even for each system on a steer financial basis was not different between the NW and LW groups, and both the NW and LW groups had lower (P = .08) break-evens than the EW group. Age of the calf at weaning affects cow weight and BCS. Net income in each system is influenced by cow costs, month of the year that steer calves are purchased into the feedlot and finished steers are sold, month of the year cull cows are marketed, and replacement heifer development costs.  相似文献

Seasonal weight loss (SWL) is the most pressing constraint in ruminant production systems in tropical climates. SWL is controlled using supplementation, which is costly and difficult to implement in extensive systems, or using breeds adapted to tropical hot dry climates, like the Damara and Dorper. Albeit 15 years in Australia, little is known on how these sheep compare to Australian Merino. Here, the responses of the Damara, Dorper and Merino breeds to nutritional stress were compared. Seventy-two 6-month-old ram lambs, 24 from each breed, were allocated to growth (gaining 100 g/day) or restricted diets (losing 100 g/day, 85 % of maintenance needs). Animals were weighed and carcass and meat characteristics determined. Results point out to the existence of important differences between the three genotypes, in particular between the Merino and the Southern African breeds. Additionally, Merino ram lambs seem to have been more influenced by SWL than the other two, with consequences on meat characteristics.  相似文献

The silverleaf whitefly Bemisia tabaci (Genn.) biotype B (Hemiptera: Aleyrodidae) is an economically important pest of tomatoes Solanum lycopersicum (L.), causing irregular ripening on fruits and transmitting several plant pathogenic geminiviruses. The management of this pest is commonly based on repetitive spraying with synthetic pesticides, causing serious environmental damages and increase of resistance by insect population. In the present study, essential oils from the leaves of Artemisia camphorata Vill., Ageratum conyzoides L., Foeniculum vulgare Mill., Lippia alba (Mill.) N. E. Br., Plectranthus neochilus Schltr., and Tagetes erecta L. were investigated for their possible repellent and oviposition-deterrent effects against B. tabaci biotype B on tomato. In a multi-choice assay, P. neochilus essential oil was the most active repellent and oviposition deterrent. Essential oils of A. conyzoides and T. erecta significantly deterred the female B. tabaci biotype B from laying eggs on treated tomato leaflets compared with the control. (E)-Caryophyllene (30.67 %) and the monoterpenes α-pinene (15.02 %) and α-thujene (11.70 %) were identified as the major constituents of the essential oil of P. neochilus. Our findings demonstrated the potential of essential oil of P. neochilus and other oils in the reduction of settlement and oviposition of B. tabaci biotype B on tomato.  相似文献
