The aims of the study were to standardise an immunohistochemical (IHC) method for the detection of progesterone receptors (PR) on formalin-fixed, paraffin wax-embedded tissue sections of feline mammary gland tumours and dysplasias, comparing the results with those obtained using the radiolabelled ligand dextran coated charcoal (DCC) assay applied to frozen tissue samples from the same cases. Also, to define the immunohistochemical distribution of PR in the different cellular compartments of the lesions and to compare the oestrogen receptor (ER) and PR status of the feline mammary lesions. Proliferative mammary lesions collected from 34 cats were studied; 25 malignant tumours and 9 benign tumours and dysplasias. PR protein was present at a concentration of 5 fmol mg(-1) (positivity threshold) in 37.5 per cent of malignant tumours and 66.7 per cent of benign tumours and dysplasias while immunoreative products to PR antibody were found in the nuclei of tumour cells in 38.5 per cent and 66.7 per cent of the cases, respectively. Concordance between DCC-PR and IHC-PR was 88.5 per cent (P<0.001). The specificity (true negatives) and sensitivity (true positives) of the IHC method were 89.4 per cent and 87.5 per cent respectively. The presence of PR was linked to the absence of ovariectomy (P<0.02). Oestrogen receptors, detected by either method, were also detected in half the cases in which PR had been detected. In malignant tumours, the most prevalent groups were the ER + PR + and ER-PR + groups.  相似文献

The immunohistochemical expression of the smooth muscle-specific protein calponin was studied to assess the contribution of myoepithelial cells to the histogenesis of spindle cells of complex and mixed tumors of the mammary gland of the dog and the origin of cartilage and bone in mixed tumors. Formalin-fixed tissues from 55 benign and malignant tumors (49 also containing surrounding normal mammary gland) were evaluated. Periacinar and periductal myoepithelial cells of all the 49 normal mammary glands were diffusely stained by the anti-human calponin monoclonal antibody. Calponin was found in 53 (98%) of the tumors studied, reacting with the myoepithelium-like cells of 86% of benign tumors and their remnants in 85% of malignant tumors. Five different types of calponin-immunoreactive myoepithelial cells were identified: hypertrophic myoepithelial cells. fusiform cells, stellate myoepithelial cells, rounded (myoepithelial) cells, and chondroblasts. Differences in staining intensity and staining pattern among these five types of cells suggested a transition of myoepithelial cells to chondroblasts. Stromal myofibroblasts also showed calponin immunoreactivity, but they did not react with a cytokeratin 14 monoclonal antibody, which recognizes myoepithelial cells in mammary gland. Calponin appears to be a very sensitive marker of normal and neoplastic myoepithelium in the canine mammary gland, and its identification in different cell types of complex and mixed tumors of the mammary gland of the dog suggests a major histogenetic role for myoepithelial cells.  相似文献

Spontaneous basaloid adenomas occurred in four out of 354 dogs with mammary tumors. Affected dogs were pure-bred, intact females between 6 and 8 years of age. Three dogs were nuliparous, two had pseudopregnancies, and none had received contraceptive steroids. The tumors were multiple (three cases) or unique, less than 1 cm in diameter, well delineated, and composed of uniform cords and clusters of monomorphic epithelial cells with focal signs of squamous or glandular differentiation. A basal cell immunophenotype (cytokeratins 5 and 14 positive) without either glandular epithelial (cytokeratins 8, 18, and 19 negative) or myoepithelial (calponin negative) differentiation was observed in the majority of tumor cells. No recurrence or metastasis was recorded after follow-up periods between 3 and 24 months. In spite of the hormone-dependent nature of this tumor in female Beagles given experimental contraceptive steroids, spontaneous basaloid adenomas lacked estrogen receptor alpha and progesterone receptors.  相似文献

A 12-year-old, entire, nulliparous crossbreed female dog was presented with a history of vulval bleeding, bulging of the perineum and faecal tenesmus. A firm, non-painful perineal mass, measuring 9.11x5.4 cm, with erythema was detected. Abdominal radiography showed compression and elevation of the rectal ampulla. A dose of 10 mg/kg aglepristone was administered subcutaneously on days 1, 2, 8, 15, 28 and 35. An incision biopsy was taken on day 15 and immunohistochemical analysis showed that the majority of neoplastic cells expressed progesterone receptors. Both the cutaneous erythema and the faecal tenesmus had resolved by day 28. A 50 per cent reduction in size was observed by day 60 (surgical excision). This study shows that benign tumours of the vagina of the dog that contain progesterone receptors can be reduced in size in a palliative or neoadjuvant setting using the progesterone receptor antagonist aglepristone.  相似文献

An in vitro study was conducted to determine the anthelminthic activity of hydro-methanolic extracts of Larrea tridentata on sheathed and exsheathed larvae of Haemonchus contortus. Larvae of the parasite were incubated at 20–25 °C in hydro-methanolic extracts at concentrations of 12.5, 25, 50, 100, and 200 mg/mL for 24, 48, or 72 h. Ivermectin and water were the positive and negative controls, respectively. Total phenolic compounds of leaves of L. tridentata were 97.88?±?10.45 mg/g of dry matter. Other compounds detected in this shrub by HPLC-mass spectrometry were sesamin, galocatechin, peonidin 3-O rutinoside, methyl galangin, epigallocatechin 7-O-glucuronide, and epigalocatechin. Mortality rate of sheathed and exsheathed H. contortus was low (16–34%) with doses ≤?100 mg/mL of the extracts. At 200 mg/ml, the hydro-methanolic extracts of L. tridentata killed 32.1 and 68.4% of sheathed and exsheathed larvae, respectively, regardless of incubation time. The effective concentration of the L. tridentata extract for 50% larvae mortality (EC50) after 24 h of incubation was 36 mg/mL (CI?=?6–94). Microscopic observations revealed damage to the cuticle of this parasite exposed to extracts of L. tridentata. These in vitro results provided evidence that L. tridentata extracts possess anti-Haemonchus contortus properties, particularly during the exsheathed stage of this nematode. It would be necessary to assess the safety of this shrub in vivo and also to carry out in vivo efficacy studies.

The effects of five salmon aquaculture anti-sea louse chemotherapeutant active ingredients [azamethiphos (AZ), hydrogen peroxide (HP), emamectin benzoate (EB), cypermethrin (CP) and deltamethrin (DM)] were examined in five life stages (egg, Stage I, Stage III, Stage V and juvenile) of Pacific spot prawn (Pandalus platyceros) for two exposure durations (1-h and 3-h). Endpoints affected by exposures to chemotherapeutants included morbidity in all life stages, egg hatch success, hatchling fitness, latency to moult between successive stages and survival to the juvenile stage. Across life stages, 1-h and 3-h median lethal effect concentrations on morbidity ranged from 73 to 809 mg/L HP; 10–236 µg/L AZ; 321 to >1200 µg/L EB; 0.1 to >5 µg/L CP; and 12 to >1000 ng/L DM. Across endpoints, DM was the most toxic, followed by CP, AZ and EB, followed by HP, which was generally observed to be the least toxic regardless of endpoint examined. The embryo and juvenile life stages were generally more resilient to effects than any larval life stage. Longer duration (3-h) exposures were generally more potent than shorter (1-h) exposures, with up to a threefold greater magnitude of effect when these exposure time frames were compared. Considering predicted environmental concentrations, the estimated greatest risk to ELS spot prawns was observed to be posed by DM, followed by CP, AZ and then EB, with a variable relative risk of HP across different endpoints/life stages.  相似文献

The lipid peroxidation product malondialdehyde is mostly bound to proteins in foods as an N-2-propenal derivative that is released as N-epsilon-(2-propenal)lysine by digestive enzymes. N-2-Propenals have been identified as the major forms of malondialdehyde in urine. To determine whether available lysine can be released from the N-2-propenals of lysine in vivo, two preparations containing N-epsilon-(2-propenal)lysine and N-alpha-(2-propenal)lysine or N,N'-di-(2-propenal)lysine were synthesized using radioactively labeled lysine and were administered to rats by gastric intubation and intraperitoneal injection. Both preparations were absorbed from the digestive tract, although not as efficiently as free lysine, but most of the radioactivity was excreted in urine. The radioactive label was also readily excreted after intraperitoneal injection. It is concluded that the N-2-propenals of lysine are fairly stable in vivo, so that, although they are absorbed from the gut, most of the absorbed material is not metabolized and is readily excreted as nonavailable lysine.  相似文献

A kinetic analysis of the anaerobic digestion process of wastewater derived from the production of protein isolates from extracted sunflower flour was carried out. The digestion was conducted in a laboratory-scale fluidized bed reactor with saponite (magnesium silicate) as support for the mediating bacteria at psychrophilic temperature (15-19 degrees C). Soluble chemical oxygen demand (COD(s)) removal efficiencies in the range of 95.9-69.0% were achieved in the reactor at organic loading rates (OLR) of between 0.57 and 2.49 g total COD (COD(t))/L d, hydraulic retention times (HRT) of between 20.0 and 4.5 days, and average feed total COD concentration of 11.3 g/L. The yield coefficient of methane production was 0.32 L of methane (at STP) per gram of COD(t) removed. The total volatile fatty acid (TVFA) levels and the TVFA/alkalinity ratio were lower than the suggested limits for digester failure for OLR and HRT up to 2.26 g COD(t)/L d and 5.0 days, respectively. The specific rate of substrate uptake, r (g COD(s)/g VSS d), correlated with the concentration of biodegradable substrate, S (g COD(s)/L), through an equation of the Michaelis-Menten type. The maximum substrate utilization rate, k, and the Michaelis constant, K(s)(), were found to be 0.125 g COD(s)/g VSS d and 124 mg COD(s)/L, respectively. This proposed model predicted the behavior of the reactor very accurately showing deviations lower than 10% between the experimental and theoretical values of substrate uptake rates. A mass (COD(t)) balance around the reactor allowed the COD equivalent of methane volume (W(CH)4) to be obtained, which gave a value of 2.89 g COD(t)/L CH(4), which was virtually coincident with the theoretical value of 2.86 g COD(t)/L CH(4).  相似文献

A study has been conducted into the genetic variation analysis of the tetraploid asparagus landrace ‘Morado de Huetor’ and its relationship with current commercial cultivars using Random Amplified Polymorphic DNA (RAPD) molecular markers. The presence of different ploidic levels in the ‘Morado de Huetor’ landrace has been also studied using cytogenetic analysis. Ten decamer oligonucleotides were used to obtain RAPD markers and to characterise 52 individuals of ‘Morado de Huetor’, 7 of the tetraploid cultivar ‘Purple Passion’, and 55 of 5 diploid hybrid commercial cultivars. Jaccard similarity index was calculated and a cluster analysis using UPGMA (Unweighted Pair-Group Method using Arithmetic Average) was performed. Tetraploid asparagus, ‘Morado de Huetor’ and ‘Purple Passion’, were well differentiated from the diploid ones showing a higher genetic variability. This result suggests a different origin for tetraploid and diploid asparagus varieties. Within the diploid cultivars a variety distribution was obtained. A specific monomorphic band (OPB20883) was found in tetraploid varieties. Within the diploid cultivars, two bands (OPB20830, OPC15705) can be used to distinguish between the hybrid cultivars used in this study. Tri-, penta-, hexa-, and octoploid plants were found in ‘Morado de Huetor’. The origin of these ploidic levels is discussed and their use in the development of new varieties is proposed. In conclusion, ‘Morado de Huetor’ is a genetic resource that could be used to increase the narrow genetic background reported in diploid asparagus cultivars.  相似文献
