Summary: A new locomotory disturbance of cattle is described. The condition has occurred sporadically since the mid-1980s. Affected herds had all grazed flood plain pastures in a restricted area of north-western New South Wales. Calves were either born with clinical signs or developed them by 4 months of age. The disease was characterised by a slowly progressive, irreversible, asymmetrical, paresis of the hind limbs. Affected cattle experienced persistent hyper-extension of the hip and stifle joints. Macroscopic and microscopic examination of the nervous and musculoskeletal systems failed to demonstrate abnormalities that would account for the clinical signs. The disorder shares many similarities with bovine spastic paresis. It is suggested that the pathogenesis of the disorder is nervous, and probably involves nigro-striatal, medulla oblongata, and spinal dysfunctional inputs. An in-utero plant poisoning was suspected but no specific plant association was determined.  相似文献

SUMMARY An Australian bovine herpesvirus 1 (BHV1) isolate with a defined (427 base pair) deletion in the protein coding region of the thymidine kinase gene was obtained by standard marker rescue procedures. After selection in the presence of the nucleotide analogue 5iodo-deoxy-uridine the virus was analysed by hybridisation with three differential oligonucleotide probes, restriction endonuclease profile studies and DNA sequence analysis. The virus elicited an immune response in recipient animals after either intramuscular or intravenous administration and produced no significant deleterious side-effects when administered at a dose sufficient to stimulate the host immune response. The safety and immunogenicity of the recombinant BHV1 virus 39B1 were similar to those reported for other registered BHV1 vaccines and the virus would appear to be suitable for the production of a vaccine seed lot and more exhaustive field trials as a prelude to commercial vaccine production and registration.  相似文献

OBJECTIVE: To develop rapid (< 8 hour) tests using polymerase chain reaction (PCR) for the diagnosis of equine herpesvirus 3 (EHV3; equine coital exanthema virus), equine gammaherpesviruses 2 (EHV2) and EHV5, equine adenovirus 1 (EAdV1), EAdV2, equine arteritis virus (EAV), equine rhinitis A virus (ERAV; formerly equine rhinovirus 1) DESIGN: Either single round or second round (seminested) PCRs were developed and validated. METHODS: Oligonucleotide primers were designed that were specific for each virus, PCR conditions were defined and the specificity and sensitivity of the assays were determined. The application of the tests was validated using a number of independent virus isolates for most of the viruses studied. The PCRs were applied directly to clinical samples where samples were available. RESULTS: We developed a single round PCR for the diagnosis of EHV3, a seminested PCR for EHV2 and single round PCRs for EHV5, EAdV1, EAdV2 and RT-PCRs for EAV and ERAV. The PCR primer sets for each virus were designed and shown to be highly specific (did not amplify any recognised non-target template) and sensitive (detection of minimal amounts of virus) and, where multiple virus isolates were available all isolates were detected. CONCLUSION: The development and validation of a comprehensive panel of PCR diagnostic tests, predominantly for viruses causing equine respiratory disease, that can be completed within 8 hours from receipt of clinical samples, provides a major advance in the rapid diagnosis or exclusion diagnosis of these endemic equine virus diseases in Australia.  相似文献

Broad-sense heritability, and genetic and phenotypic correlations were estimated for yield (nut-in-shell; kernel yield; and net present value of crop) per tree in macadamia to 10 years after planting from a trial of 40 cultivars replicated over 4locations. In addition, canopy width at 10years of age was measured and used to calculate yield efficiency as yield per square metre of projected canopy area. Stem girth above the graft union was also measured. There was a low broad-sense heritability (0.06 < H <0.22) and correlation of cultivars across locations (r gloc = 0.14–0.52) for yield per tree. However, the genetic correlation of cumulative yield was high among 3 of the 4locations (g ≥ 0.75), suggesting the higher genetic variance at Rockhampton was the main cause of the genotype by environmental interaction. Heritability was higher for canopy width (H = 0.28) and yield efficiency (H ≈ 0.47) and cultivar performance was highly correlated across locations (r gloc ≈ 0.70) for these traits. There was a strong genetic correlation (r g ≥ 0.90) between cumulative yield per tree of cultivars to 7 years and cumulative yield per tree to 10 years. Genetic correlation among all measures of yield per tree were high (r g ≥ 0.73), but there was no genetic correlation with canopy width. On the other hand, small cultivars tended to have higher yield efficiency (r g <–0.62). There was also a slight negative correlation between nut-in-shell yield per tree and kernel recovery (r g = –0.37). These results suggest the use of family information and index selection may improve the efficiency of selection and breeding programs in macadamia. Eight-year-old lychee (Litchi chinensis Sonn.) trees, cv. 'Bengal,' growing in krasnozem soil were subjected to soil water deficit from one month before flowering until harvest by covering the ground with polyethylene sheeting and withholding irrigation. The ratio of daytime stomatal conductance of unirrigated to irrigated trees decreased 20% during the three months of increasing water deficit. Predawn leaf water potentials of irrigated trees averaged about -0.3 MPa throughout the period, whereas they declined progressively to -0.9 MPa in unirrigated trees. Minimum daytime leaf water potential in the unirrigated trees decreased from -1.0 to -1.1 MPa at the beginning of the drought period to -2.2 to -2.4 MPa after three months, and calculated whole-plant conductance did not change with decreasing availability of water. The calculated soil-root water potential declined to less than -1.0 MPa in unirrigated trees. Capacitance effects on the relationship between leaf water potential and transpiration were significant only at low transpiration rates. Although unirrigated trees reduced soil water content at 0-30 cm depths to an equivalent water potential of -1.0 MPa, fruit shedding was significantly less than in irrigated trees. Water deficit had no effect on the fresh weight of pericarp, but caused increased seed size and decreased fresh weight of flesh, resulting in fruit from unirrigated trees being 16% lower in total fresh weight per fruit than fruit from irrigated trees.  相似文献
