Accurate diagnosis of canine filariosis is essential for choosing correct therapeutic approach. Therefore, reliable methods for discriminating among the different filarial infections in dogs are needed. The authors report simple and highly specific molecular methods that identify the three most common filarial nematodes of European dogs: Dirofilaria immitis, D. repens and Acanthocheilonema (syn. Dipetalonema) reconditum, based on (1) PCR amplifications of mitochondrial DNA (12S rDNA and coxI) with general filarial primers followed by digestion with restriction enzymes that generates band polymorphisms clearly discriminating the three species and (2) PCR amplifications with species-specific primers to support the restriction analysis, in particular in the case of multiple infections.  相似文献

Two hybrid poplar maternal half-sib families, resulting from controlled crosses of the female parent Populus deltoides 'S9-2' with P. nigra 'Ghoy' and P. trichocarpa 'V24', were grown at three sites: northern Italy, central France and southern England. Juvenile stem growth traits (height, circumference and volume) and sylleptic branchiness (number of branches, density of branches per unit of stem height, percentage of the stem carrying branches and distance of the highest sylleptic branch to the top of the stem) were measured on 1-year-old shoots. Our general objectives were to determine the degree to which the expression of stem growth and syllepsis and the relationships between them are affected by environmental conditions and to evaluate the efficiency of indirect selection for stem growth using branching traits as secondary criteria. The performance of both families differed significantly within and between sites. Pronounced heterosis was observed and highly significant genotype x environment interactions were found for all traits across the sites. Syllepsis showed more marked genetic variation and plasticity than stem growth traits. Relationships between sylleptic branchiness and stem growth depended on environmental conditions. Heritability values at the individual level ranged between 0.09 and 0.59, but genetic gain in stem volume was not significantly improved when selection was based on sylleptic branch characteristics. However, despite strong phenotypic plasticity among sites, genotypic ranking among the sites was relatively stable.  相似文献

Strigolactones are a class of bioactive natural metabolites produced by plant roots and released into the rhizosphere. They were discovered as signals indispensable for the induction of germination of seeds of root parasitic weeds, but since then, interestingly, many other biological, physiological and ecological roles have been described. This has suddenly provoked huge scientific interest in these compounds within different research fields. In this short perspective the attention is focused mainly on the ongoing and future research aimed at deciphering the biological roles of strigolactones that could positively affect, more or less directly, the management of parasitic weeds.  相似文献

The authors report the efficacy of an injectable, moxidectin sustained release (SR) formulation for the prevention of canine heartworm infection in endemic areas in northern and central Italy. Three field trials were carried out on a total of 324 dogs. Two hundred forty-three dogs were treated with moxidectin SR 6 months apart and 81 dogs (positive controls) with moxidectin tablets given monthly for 5 consecutive months during the risk season each year throughout the study. Results of testing for microfilariae and circulating adult female antigens were negative for all the experimentally treated dogs at the 6, 7, 11 and 19 months after the last injection. No adverse reactions to moxidectin SR were observed but a moderate pain at palpation and swelling (5-6 cm) at the injection site after the first treatment. In the study areas, prevalence of Dirofilaria immitis infection calculated by testing dogs which had no preventive treatment in the previous transmission season ranged from 33 to 63%. This study confirms the efficacy and safety of injectable, moxidectin SR formulation in the prevention of heartworm infection in dogs and demonstrates that the prophylactic efficacy lasts for the full season and strongly suggests that the product gives 1-year protection.  相似文献

Thirty 6-trifluoromethylpyrazolo[3,4-d]pyrimidines were synthesised and tested for antifungal activity in vitro against ten phytopathogenic fungi of different taxonomic classes. Six of the compounds had noteworthy activity against Sclerotinia minor, Corticium solani and Phoma betae. Numerous canine papillomaviruses (CPVs) have been identified (CPV1–23). CPV1, 2, and 6 have been associated with inverted papillomas (IPs). We retrieved 19 IPs from 3 histopathology archives, and evaluated and scored koilocytes, inclusion bodies, giant keratohyalin granules, cytoplasmic pallor, ballooning degeneration, and parakeratosis. IHC targeting major capsid proteins of PV was performed, and CPV genotyping was achieved by PCR testing. Tissue localization of CPV DNA and RNA was studied by chromogenic and RNAscope in situ hybridization (DNA-CISH, RNA-ISH, respectively). IPs were localized to the limbs (50%), trunk (30%), and head (20%), mainly as single nodules (16 of 19). In 15 of 19 cases, immunopositivity was detected within the nuclei in corneal and subcorneal epidermal layers. PCR revealed CPV1 in 11 IPs and CPV2 DNA in 3 IPs. Overall, 14 of 17 cases were positive by both DNA-CISH and RNA-ISH, in accord with PCR results. A histologic score >5 was always obtained in cases in which the viral etiology was demonstrated by IHC, DNA-CISH, and RNA-ISH. IHC and molecular approaches were useful to ascertain the viral etiology of IPs. Although IHC is the first choice for diagnostic purposes, ISH testing allows identification of PV type and the infection phase. RNA-ISH seems a promising tool to deepen our understanding of the pathogenesis of different PV types in animal species.  相似文献

The algal polysaccharide agar has long been used as a food binder due to its structure, rheological behaviour, stability and interactions – properties that help to generate firm, round, disk‐shaped pellets that may be used in recirculating sea urchin‐rearing systems. Three algae‐based diets (Ulva lactuca, Gracilaria gracilis, Cystoseira sp.) containing 3% and 6% agar were tested on the sea urchin Paracentrotus lividus in order to examine the effect of varying percentages of agar on pellet stability in water and sea urchin gonad growth. The kinetics of water absorption and solute leaching of pellets were measured by immersing quadruplicate samples of the pellets in water for 1, 2, 3, 4, 5 and 6 days. Our results show that the pellets had good water stability, were readily consumed by sea urchins and the presence of agar did not hamper sea urchin gonad growth. Animals fed Ulva‐containing pellets reached a more advanced gametogenic stage with respect to animals fed Cystoseira‐ and Gracilaria‐containing pellets. Moreover, the presence of agarase activity in the digestive system indicated that agar may be an energy source. Pellets are relatively low cost and easy to prepare and store. They may represent a useful resource for rearing sea urchins under intensive conditions.  相似文献

A simple and sensitive method for the confirmation of perfluidone residues in rice grains is described. The procedure involves extraction, purification on an anionexchange resin, chlorination, methylation and gas-chromatographic analysis with electron capture detection. The recoveries from fortified rice grains varied from 84.0 to 92.4% in the range 0.01–0.20 mg/kg. The sensitivity was such that perfluidone could be detected down to a level of 0.005 mg/kg. The method can be used for confirmation of perfluidone quantitatively as well as qualitatively.  相似文献

Vegetative propagation techniques such as grafting can be used, in conjunction with field studies, to decouple the relative effects of age and size on tree metabolism and growth. Despite interest in this approach, little attention has been paid to the best metrics for assessing the growth performance of grafted plants over time. Based on an analysis of the grafting literature and our own data, we show that the choice of metrics to assess tree growth can entirely change the conclusions reached about the relative importance of age versus size. We recommend that absolute as well as relative rates of growth are calculated and that scion size be standardized as much as possible at the start of the experiment. Once proper metrics are chosen, all of the available published evidence is largely concordant with two concepts: (1) age-mediated controls of tree growth are likely to be important during the first few years of a tree's life (before phase change); and (2) after the first few years of a tree's life, size-mediated factors largely prevail over age-mediated factors in determining tree growth rates. We found no support for theories invoking age-mediated sink limitations in old trees.  相似文献
