We investigated the population structure of the quokka (Setonix brachyurus) on the mainland of Western Australia using mark–recapture techniques. Seven previously known local populations and one unconfirmed site supporting the preferred, patchy and discrete, swampy habitat of the quokka were trapped. The quokka is now considered as locally extinct at three sites. The five remaining sites had extremely low numbers, ranging from 1 to 36 individuals. Population density at these sites ranged from 0.07 to 4.3 individuals per hectare. There has been no response to the on-going, 6 year fox control programme occurring in the region despite the quokkas’ high fecundity and this is due to low recruitment levels. The remaining quokka populations in the northern jarrah forest appear to be the terminal remnants of a collapsing metapopulation. 相似文献

The shoot apical meristem (SAM) is a collection of stem cells that resides at the tip of each shoot and provides the cells of the shoot. It is divided into functional regions. The central zone (CZ) at the tip of the meristem is the domain of expression of the CLAVATA3 (CLV3) gene, encoding a putative ligand for a transmembrane receptor kinase, CLAVATA1, active in cells of the rib meristem (RM), located just below the CZ. We show here that CLV3 restricts its own domain of expression (the CZ) by preventing differentiation of peripheral zone cells (PZ), which surround the CZ, into CZ cells and restricts overall SAM size by a separate, long-range effect on cell division rate. 相似文献

The 1918 influenza pandemic resulted in about 20 million deaths. This enormous impact, coupled with renewed interest in emerging infections, makes characterization of the virus involved a priority. Receptor binding, the initial event in virus infection, is a major determinant of virus transmissibility that, for influenza viruses, is mediated by the hemagglutinin (HA) membrane glycoprotein. We have determined the crystal structures of the HA from the 1918 virus and two closely related HAs in complex with receptor analogs. They explain how the 1918 HA, while retaining receptor binding site amino acids characteristic of an avian precursor HA, is able to bind human receptors and how, as a consequence, the virus was able to spread in the human population. 相似文献

Observations of the 1.4- to 2.4-micrometer spectrum of Pluto reveal absorptions of carbon monoxide and nitrogen ices and confirm the presence of solid methane. Frozen nitrogen is more abundant than the other two ices by a factor of about 50; gaseous nitrogen must therefore be the major atmospheric constituent. The absence of carbon dioxide absorptions is one of several differences between the spectra of Pluto and Triton in this region. A key gene involved in plant senescence, mutations of which partially disable chlorophyll catabolism and confer stay-green leaf and cotyledon phenotypes, has been identified in Pisum sativum, Arabidopsis thaliana, and Festuca pratensis by using classical and molecular genetics and comparative genomics. A stay-green locus in F. pratensis is syntenically equivalent to a similar stay-green locus on rice chromosome 9. Functional testing in Arabidopsis of a homolog of the rice candidate gene revealed (i) senescence-associated gene expression and (ii) a stay-green phenotype after RNA interference silencing. Genetic mapping in pea demonstrated cosegregation with the yellow/green cotyledon polymorphism (I/i) first reported by Gregor Mendel in 1866. 相似文献

Earthworms secrete granules of calcium carbonate. These are potentially important in soil biogeochemical cycles and are routinely recorded in archaeological studies of Quaternary soils. Production rates of calcium carbonate granules by the earthworm Lumbricus terrestris L. were determined over 27 days in a range of soils with differing chemical properties (pH, organic matter content, water holding capacity, bulk composition, cation exchange capacity and exchangeable cations). Production rate varied between soils, lay in the range (0–4.3 mg) earthworm?1 d?1 with an average rate of (0.8 mg) earthworm?1 d?1 and was significantly correlated (r = 0.68, P ≤ 0.01) with soil pH. In a second experiment lasting 315 days earthworms repeatedly (over periods of 39–57 days) produced comparable masses of granules. Converting individual earthworm granule production rates into fluxes expressed on a per hectare of land per year basis depends heavily on estimates of earthworm numbers. Using values of 10–20 L. terrestris m?2 suggests a rate of . Data obtained from flow-through dissolution experiments suggest that at near neutral pH, granule geometric surface area-normalised dissolution rates are similar to those for other biogenic and inorganic calcites. Fits of the data to the dissolution relationship r = k(1 ? Ω)n where r = dissolution rate, k = a rate constant, Ω = relative saturation and n = the reaction order gave values of k = 1.72 × 10?10 mol cm?2 s?1 and n = 1.8 for the geometric surface area-normalised rates and k = 3.51 × 10?13 mol cm?2 s?1 and n = 1.8 for the BET surface area-normalised rates. In 196 day leaching column experiments trends in granule dissolution rate referenced to soil chemistry corresponded to predictions made by the SLIM model for dissolution of limestone in soil. If soil solution approaches saturation with respect to calcium carbonate, granule dissolution will slow or even stop and granules be preserved indefinitely. Soils are structurally heterogeneous across a wide range of spatio-temporal scales. Consequently, external environmental conditions do not have a uniform effect throughout the soil, resulting in a large diversity of micro-habitats. It has been suggested that soil function can be studied without explicit consideration of such fine detail, but recent research has indicated that the micro-scale distribution of organisms may be of importance for a mechanistic understanding of many soil functions. Current techniques still lack the adequate sensitivity and resolution for data collection at the micro-scale, and the question ‘How important are various soil processes acting at different scales for ecological function?’ is therefore challenging to answer. The nano-scale secondary ion mass spectrometer (NanoSIMS) represents the latest generation of ion microprobes, which link high-resolution microscopy with isotopic analysis. The main advantage of NanoSIMS over other secondary ion mass spectrometers is its ability to operate at high mass resolution, whilst maintaining both excellent signal transmission and spatial resolution (down to 50 nm). NanoSIMS has been used previously in studies focussing on presolar materials from meteorites, in material science, biology, geology and mineralogy. Recently, the potential of NanoSIMS as a new tool in the study of biophysical interfaces in soils has been demonstrated. This paper describes the principles of NanoSIMS and discusses the potential of this tool to contribute to the field of biogeochemistry and soil ecology. Practical considerations (sample size and preparation, simultaneous collection of isotopes, mass resolution, isobaric interference and quantification of the isotopes of interest) are discussed. Adequate sample preparation, avoiding bias due to artefacts, and identification of regions-of-interest will be critical concerns if NanoSIMS is used as a new tool in biogeochemistry and soil ecology. Finally, we review the areas of research most likely to benefit from the high spatial and high mass resolution attainable with this new approach. 相似文献

The effect of nutritional restriction and rehabilitation imposed between 5 and 35 d and 35 and 166 d of age, respectively, on body and tissue growth were investigated in 96 Yorkshire pigs from 12 litters. One-half of the pigs were allowed to suckle continuously until weaning (fully fed, FF); the remainder were removed from the sow for 16 h of each 24 h period (1600 to 0800 h), during which time they received only water (restricted feeding, RF). All pigs were fed ad libitum from 35 d of age. Although significantly lower body and tissue (liver, kidney and gastrocnemius muscle) weights were observed in the RF pigs at 35 and 70 d of age, by 166 d, because of compensatory growth between 35 and 166 d, the difference in body and muscle weights between the two groups of pigs (RF and FF) was reduced to 5%, but the difference in kidney and liver weight remained as high as 15 and 9%, respectively. Total DNA, RNA and protein in the tissues examined were also lower in the RF pigs at 35 and 70 d of age, but increased after ad libitum feeding to more than 90% of the corresponding value in the FF pigs by the time they reached 166 d of age. The results of these measurements and the comparison of the protein: DNA ratios of the tissues indicated that growth retardation in the RF pigs was associated with reduced cell proliferation in liver and kidney and nuclear proliferation in muscle rather than any decrease in cell (or fiber) volume. Percentage of fat was lower while percentage of protein was higher (on a dry matter basis) in the RF pigs than in the FF pigs at 35 d of age. No significant difference in fat and protein content in the carcasses between the RF and FF pigs at 70 or 166 d of age or 90 kg body weight was noted. OBJECTIVE: To determine anesthetic techniques and the drugs used to provide anesthesia and analgesia to reptiles. DESIGN: Mail-out questionnaire. SAMPLE POPULATION: 367 members of the Association of Reptile and Amphibian Veterinarians. PROCEDURE: 1,091 members listed in the 2002 directory of the Association of Reptile and Amphibian Veterinarians were asked to complete a questionnaire regarding anesthesia and analgesia. RESULTS: 367 of 1,091 (33.6%) individuals completed the questionnaire; 88.8% used inhalants (particularly isoflurane) for anesthesia, and ketamine, propofol, and butorphanol were the most commonly used injectable agents. Intubation, fluids, and having a dedicated anesthetist were most commonly used for patient support, and pulse oximetry and Doppler ultrasonography were most commonly used for monitoring. Respiratory depression, difficulty monitoring anesthetic depth, prolonged recovery, and hypothermia were the most frequent complications. Nearly all respondents believed that reptiles feel pain, but analgesics were used infrequently for many reasons. CONCLUSIONS AND CLINICAL RELEVANCE: Providing anesthesia in reptiles is difficult, especially regarding anesthetic depth and vital parameters, and methods of support are used less frequently than in domestic species. Provision of analgesia is uncommon. Research regarding pain and its assessment, response to analgesics, and drug pharmacokinetics is needed. Dissemination of this information to practitioners needs to be improved for enhancement of the standard of care for reptiles. 相似文献
