Over a period of three months in 1985, paramyxovirus type 1 infection was demonstrated for the first time in Canada, in six flocks of pigeons in Ontario, Alberta, and British Columbia. The paramyxovirus type 1 isolates did not cause clinical disease when serially passaged four times in four-to six-week-old chickens, and isolates were classified as lentogenic before and after such serial passage. Further cases of paramyxovirus type 1 clinical disease have not been reported since the last of these six outbreaks in August 1985.  相似文献

In vitro cultured shoots of three Chinese poplar species (Populus tomentosa, P. alba cv. Pyramidalis andP. euphratica) and an improved poplar (FS-51), as a control, were exposed to the stress of low osmotic potential, salinity and high pH, and the shoot growth was measured.P. tomentosa andP. euphratica could grow even under the high osmotic stress (0.5m mannitol; −1.7 MPa).P. euphratica shoots which were previously cultured on the medium containing 0.7m mannitol (−2.4 MPa) recovered its growth after being transplanted to the medium without mannitol. The shoot growth ofP. euphratica was not so reduced even on the medium containing 100 mM NaCl. However, growth of the other poplars markedly reduced with 10 mM NaCl. Medium pH (pH 5.8–10) also did not affect the shoot growth ofP. euphratica. These poplars have different tolerance to the stresses in cultured shootin vitro. P. euphratica was extremely tolerant to the stresses which are possible to damage plants in dry land. A part of this study was presented at the 104th Annual Meeting of the Japanese Forestry Society. 1H-Nuclear magnetic resonance (NMR) microscopy was used to study freezing behavior in wintering leaf buds of Momi fir (Abies firma Sieb. et Zucc.) and Japanese red pine (Pinus densiflora Sieb. et Zucc.). The images acquired predominantly reflected the density of mobile (i.e., non-ice) protons mainly from unfrozen water. By comparing images taken at various subfreezing temperatures, we determined which tissues produced the high and low temperature exotherms detected by differential thermal analyses. Typical extra-organ freezing was successfully imaged in leaf buds of A. firma. The bud scales readily froze at -7 degrees C, but shoot primordia remained supercooled to -14 degrees C in December buds and to -21 degrees C in March buds. The size of supercooled shoot primordia was reduced with decreasing temperature, indicating a gradual decrease in water content of the shoot primordia. In contrast, the signal from shoot primordia of P. densiflora disappeared between -7 and -14 degrees C, corresponding to the high temperature exotherm at -8 degrees C, indicating extracellular freezing of the shoot primordia. The xylem and bark tissues readily froze at -7 degrees C in A. firma and between -7 and -14 degrees C in P. densiflora. We conclude that NMR microscopy can noninvasively provide more spatially specific information about freezing behavior in leaf buds than traditional methods such as differential thermal analysis. In particular, it allows the organized and harmonized freezing behaviors in complex organs to be visualized directly thereby revealing the diversity of mechanisms involved in freezing behaviors.  相似文献

We estimated fine root biomass in a Japanese cedar (Cryptomeria japonica) plantation using a min-irhizotron technique. Since data obtained from minirhizo-trons are limited to the length and diameter of fine roots observed on minirhizotron tubes, data conversion is necessary to determine the fine root biomass per unit soil volume or unit stand area. We first examined the regression between diameter squared and weight per unit length of fine roots in soil core samples, and calculated the fine root biomass on minirhizotron tubes from their length and diameter. Then we determined conversion factors based on the ratio of the fine root biomass in soil core samples to that on minirhizotron tubes. We examined calculation methods, using a single conversion factor for total fine root biomass in the soil for depths of 0–40cm (Cal1), or using four conversion factors for fine roots in the soil at 10-cm intervals (Cal2). Cal1 overestimated fine root biomass in the lower soil or underestimated that in the upper soil, while fine root biomass calculated using Cal2 better matched that in soil core samples. These results suggest that minirhizotron data should be converted separately for different soil depths to better estimate fine root biomass.  相似文献

The organic layer is an important carbon pool in the forest ecosystem but has sometimes been underestimated or ignored. An equation estimating the carbon mass of the organic layer from its dry weight was developed using a dataset collected from various forests in Japan. The equation (logC = –0.393 + 1.031logW, where C is the carbon mass of the organic layer per unit area and W is the dry weight of organic layer per unit area) could be used to estimate the carbon mass of the organic layer with a small error (R 2 = 0.986, n = 67). The factor that converts organic matter to carbon was also examined. The factor of 1.89 ± 0.23 was suitable for the organic layer.  相似文献

ABSTRACT Apple latent spherical virus (ALSV) expressing yellow and cyan fluorescent proteins (ALSV-YFP and ALSV-CFP) was used to investigate the distribution of identical virus populations in coinfected plants. In Chenopodium quinoa plants inoculated with a mixture of ALSV-YFP and ALSV-CFP, fluorescence from YFP and CFP was always distributed separately in both inoculated and upper uninoculated leaves. Inoculation of each ALSV-YFP and ALSV-CFP to different leaves of a C. quinoa plant resulted in the separate distribution of each virus population among different upper leaves. When C. quinoa leaves were first inoculated with ALSV-CFP and then ALSV-YFP was reinoculated into the same leaves at various times after the first inoculation, ALSV-YFP infected only tissues where ALSV-CFP infection had not been established. The spatial separation was also found in Nicotiana benthamiana leaves coinoculated with Bean yellow mosaic virus (BYMV)-YFP and BYMV-CFP. In contrast, both YFP and CFP fluorescence signals were observed in the same tissues of N. benthamiana leaves mixed infected with ALSV-YFP and BYMV-CFP. YFP fluorescence from ALSV-YFP in mixed-infected leaves was brighter and longer than in leaves infected with ALSV-YFP singly.  相似文献

A 4-year-old male Shiba dog initially presented with pain of an undetermined origin and hypersensitivity to touch. Seven days later, the dog developed ataxia, hind-leg weakness and knuckling. The dog died on 20 days after presentation. Postmortem examination revealed a mass in the body of thoracic vertebra. Histopathologically, the mass consisted of granulomatous inflammation, including fungal organisms that were immunohistochemically positive for Candida albicans. Similar granulomatous lesions were observed in the systemic lymph nodes, kidneys, pancreas, spleen, prostate gland, thyroid glands and heart. This case was diagnosed as systemic candidiasis with spondylitis.  相似文献

The alphavbeta3 and alphavbeta5 integrins are known as transmembrane receptors capable of binding to the RGD amino acid peptide sequence. In mouse early gonadogenesis, some proteins containing the RGD sequence are deposited into extracellular space and participate in morphogenesis. We analyzed the expression patterns of the alphavbeta3 and alphavbeta5 integrins in mouse developing gonads (10.5-13.5 days post coitum) using whole-mount in situ hybridization. The alphav integrin mRNA was homogenously expressed in developing gonadal regions. On the other hand, the beta3 integrin mRNA was found only in large and round cells (presumptive germ cells), whereas beta5 integrin was localized in gonadal somatic cells, with the exception of coelomic epithelial cells. The beta3 integrin-expressed cells were determined to be primordial germ cells because the number of these cells was drastically reduced in busulfan-treated gonads. In this study, we demonstrated that the alphavbeta3 and alphavbeta5 integrins are widely localized in the mouse developing gonads and discussed their presumptive functions on mouse gonadogenesis.  相似文献

Severe yellowing of leaves was observed on red raspberry in Akita Prefecture. When inoculated with sap from symptomatic raspberry leaves, Chenopodium quinoa plants developed chlorotic ringspot and mottling that are typical of raspberry bushy dwarf virus (RBDV) infection. In western blot analysis, an antibody to the coat protein (CP) of RBDV reacted against ca. 30-kDa protein specific to the diseased trees. In RT-PCR testing for RBDV, single DNA fragments were amplified from total RNA samples of the diseased trees. The nucleotide sequences of the DNA fragments covering the entire CP region revealed 87–97?% identities with those of RBDV isolates.  相似文献
