The effects of spirulina and its chromophore phycocyanin, both without bound Se or selenium-enriched, were studied on plasma cholesterol, early atherosclerosis, cardiac production of superoxide anions, and NAD(P)H oxidase expression in hamsters. Forty hamsters were divided into 5 groups of 8 and fed an atherogenic diet for 12 weeks. They received by gavage either 7.14 mL/(kg day) phycocyanin (PC), Se-rich phycocyanin (SePC), spirulina (SP) or Se-rich spirulina (SeSP) in water, or water as control. SeSP and SePC supplied 0.4 microg of Se per 100 g body weight. Plasma cholesterol and non-HDL cholesterol concentrations were lower in group consuming SePC. HDL-cholesterol was never affected. SePC significantly increased plasma antioxidant capacity by 42% compared with controls. A sparing effect in liver glutathione peroxidase (87% on average) and superoxide dismutase (56% on average) activity was observed for all the groups compared to controls. Aortic fatty streak area was significantly reduced in the experimental groups, especially by PC (82%) and SePC (85%). Cardiac production of superoxide anion significantly decreased by approximately 46-76% in the four experimental groups and especially in SePC group (76%). The expression of p22phox subunit of NAD(P)H oxidase decreased by 34% after consumption of SePC. The results indicate that chronic consumption of Se-rich spirulina phycocyanin powerfully prevents the development of atherosclerosis. The underlying mechanism is related mainly to inhibiting pro-oxidant factors and at a lesser extent improving the serum lipid profile. 相似文献

After gelatinization in water, starch granules persist in swollen hydrated forms known as ghosts. Three potential mechanisms for ghost formation are tested. Proteins and lipids on the granule surface are found to be a determinant of ghost robustness, but not ghost formation. Proteins inside pre-made maize or wheat starch ghosts are degraded extensively by proteases without any apparent change in ghost properties, making an internal protein cross-linking mechanism unlikely. Waxy maize mutants with a range of amylose contents have ghost integrities that correlate with (low) apparent amylose levels. It is hypothesized that ghost formation is due to cross-linking of polysaccharide chains within swollen granules, most likely involving double helices formed from polymer chains that become free to move following heat-induced granule swelling. The size and robustness of granule ghosts is proposed to be determined by the relative rates of swelling and cross-linking, modulated by surface non-polysaccharide components. 相似文献

Several studies have shown that healthy individuals with fasting plasma glucose (FPG) levels at the high end of the normal range have an increased risk of mortality. To identify genetic determinants that contribute to interindividual variation in FPG, we tested 392,935 single-nucleotide polymorphisms (SNPs) in 654 normoglycemic participants for association with FPG, and we replicated the most strongly associated SNP (rs560887, P = 4 x 10(-7)) in 9353 participants. SNP rs560887 maps to intron 3 of the G6PC2 gene, which encodes glucose-6-phosphatase catalytic subunit-related protein (also known as IGRP), a protein selectively expressed in pancreatic islets. This SNP was associated with FPG (linear regression coefficient beta = -0.06 millimoles per liter per A allele, combined P = 4 x 10(-23)) and with pancreatic beta cell function (Homa-B model, combined P = 3 x 10(-13)) in three populations; however, it was not associated with type 2 diabetes risk. We speculate that G6PC2 regulates FPG by modulating the set point for glucose-stimulated insulin secretion in pancreatic beta cells. 相似文献

the decreasing availability of registered fungicides in specific regions, breeding for resistance against P. horiana will gain importance and will need to involve the appropriate resistance genes for the pathotypes that may be present. As
pathotypes have not been well characterized in this system, the main objective was to build an international collection of
isolates and screen these on a large collection of cultivars to identify different pathotypes. Using a robust and high throughput
bioassay, we tested 36 selected cultivars with 22 individual single-pustule isolates of P. horiana. The isolates originated from three different continents over 4 different collection years and included some isolates from
cultivars previously reported as resistant. In most cases the bioassays resulted in a clear scoring of interaction phenotypes
as susceptible or resistant, while in several cases consistent intermediate phenotypes were found, often on specific cultivars.
Twenty-four of the cultivars gave a differential interaction phenotype profile. All isolates produced a unique profile, infecting
a minimum of 4 and a maximum of 19 differential cultivars. Based on the Person analysis of these profiles, this pathosystem
contains at least seven resistance genes (and seven avirulence genes), demonstrating the highly complex race structure in

Diarrhoea due to enterotoxigenic Escherichia coli with fimbriae F4 (ETEC-F4) is an important problem in neonatal and just weaned piglets and hence for the pig farming industry. There is substantial evidence for a genetic basis for susceptibility to ETEC-F4 since not all piglets suffer from diarrhoea after an ETEC-F4 infection. It is assumed that the wild boar was originally ETEC-F4 resistant and that susceptibility towards ETEC arose after domestication. There are different phenotypes in the pig determined by which of the three existing F4 variants (F4ab, F4ac or F4ad) they are susceptible or resistant for. This suggests that several F4 receptors exist, expressed individually or in combination with each other on the brush border of the piglet’s small intestine. As such, the mucin-type glycoproteins (IMTGP) are described as F4ab/ac receptors, while the intestinal neutral glycospingolipid (IGLad) is proposed as an F4ad receptor. GP74 is a putative F4ab receptor. However, the specific genes that encode for the susceptibility are not yet known. In the past decades, linkage analyses revealed that the loci encoding for the receptor(s) for the two most frequent variants F4ab and F4ac were mapped to the 13th chromosome of the pig (Sus scrofa 13, SSC13). After fine mapping, the region of interest was mapped between two microsatellite markers, Sw207 and S0075, and interesting candidate genes surfaced. Numerous SNP analyses and a few expression studies on the three MUC-genes (MUC4, MUC13 and MUC20) and the transferrin receptor gene (TFRC) as well as on some other positional candidate genes have been performed in order to find the causative mutation for the ETEC-F4ab/ac receptor(s). Swine brucellosis is caused by the biovars 1, 2 and 3 of Brucella suis the identification of which up to now relies on microbiological tests lacking adequate specificity together with time consuming and expensive molecular procedures. Based on sequence variation of the omp2b gene, we have developed a four primer set multiplex PCR assay that was tested for polymorphism analysis of B. suis biovars causing brucellosis in swine. The assay exploits the single nucleotide polymorphisms found in omp2b gene of B. suis reference biovars which are conserved in 43 B. suis field isolates from different geographic origins and hosts. Three specific amplification patterns (S1, S2 and S3) were obtained for reference strains of B. suis biovars 1, 2 and 3, respectively. However, some B. suis field isolates identified as biovars 2 or 3 according AMOS-PCR, PCR-RFLP of omp31 and omp2 genes and classical bacteriological methods, resulted also in S1 patterns, limiting the typing usefulness of the method. 相似文献

The need to avert unacceptable and irreversible environmental change is the most urgent challenge facing society. Landscape ecology has the capacity to help address these challenges by providing spatially-explicit solutions to landscape sustainability problems. However, despite a large body of research, the real impact of landscape ecology on sustainable landscape management and planning is still limited. In this paper, we first outline a typology of landscape sustainability problems which serves to guide landscape ecologists in the problem-solving process. We then outline a formal problem-solving approach, whereby landscape ecologists can better bring about disciplinary integration, a consideration of multiple landscape functions over long time scales, and a focus on decision making. This framework explicitly considers multiple ecological objectives and socio-economic constraints, the spatial allocation of scarce resources to address these objectives, and the timing of the implementation of management actions. It aims to make explicit the problem-solving objectives, management options and the system understanding required to make sustainable landscape planning decisions. We propose that by adopting a more problem-solving approach, landscape ecologists can make a significant contribution towards realising sustainable future landscapes. 相似文献

Low temperature (LT) exposure has been shown to delay postharvest needle abscission and senescence (NAS) in balsam fir and changes in vapor pressure deficit (VPD) are expected to alter these processes. Two and half year-old seedlings were exposed to a LT of 5°C for 15 days while the control (CT) group was maintained at 22°C. Seedlings were then exposed to four different VPD (0.22, 0.87, 1.3 and 1.86?kPa) and observed for relative water content (RWC), xylem pressure potential (XPP), membrane injury index (MII), stem capacitance (SC) and NAS. An interactive effect of LT and VPD was observed in RWC, XPP, MII and NAS. Low VPD (0.22 and 0.87?kPa) resulted in less negative XPP, lower electrolyte leakage, higher SC and ultimately higher NAS than those at high VPD. Maximum NAS was recorded at 0.22?kPa. At 1.86?kPa, LT had 5× lower RWC (13%), 3× more negative XPP (?1.1?MPa), 1.8× higher membrane damage and 35% lower NAS (47 days) than CT. The SC declined with an increase in VPD with no effect of LT. The XPP and RWC of LT seedlings showed a positive relationship with NAS with R2 values of 0.54 and 0.59, respectively. LT offered no benefits to NAS at high VPD environments. 相似文献
