The small deformation rheological properties of wheat flour doughs in relation to their structure and hydration were studied by dynamic mechanical thermal analysis, differential scanning calorimetry, and electron spin resonance. The effect of salt and triglycerides was also examined and compared with results we obtained previously on starch dispersions. Moisture content was adjusted to 48 or 60% (w/w, wb). Samples contained 0–16% NaCl (g/100 g of flour‐water) and 0–18% triolein or lard (g/100 g of flour‐water). The obtained results suggested that starch has an active role in determining the evolution of dough rheological characteristics during heating. The main factors controlling rheological behavior during thermal treatment are the volume fraction and deformability of starch granules. Gluten changes the viscoelasticity of the continuous phase and competes with starch for water. The addition of sodium chloride to flour dispersions shifted the structural disorganization and rigidity increased during heating to higher temperatures. At >7% NaCl, the reverse effect was observed. The mechanism controlling the effect of salt on dough rheological behavior was explained in terms of effect on water properties and on starch structure and hydration. Triglycerides had a lubricant effect (i.e., lowering G′ modulus) on the wheat flour dough system. These effects are of great importance for production and quality of bakery products.  相似文献

Dry glassy bread, cooked gluten, and gelatinized wheat starch-sucrose mixtures were prepared. Dielectric properties were studied as a function of temperature at different frequencies ranging from 100 to 1,000 kHz. Both bread and starch samples exhibited a tan δ peak at -53°C at 10 kHz, which is associated with a secondary relaxation characterized by an activation energy of 50 kJ/mol. The gluten sample did not show any relaxation in the temperature range studied. The magnitude of the relaxation peak was sensitive to the sucrose concentration of the starch sample. The higher the sucrose content, the greater the amplitude of the peak. Such an effect is attributed to a greater β-relaxation induced by an increasing number of relaxing units or increasing size of the loosely packed regions of the glass. However, it cannot be concluded whether the relaxation is associated with a group present on both starch and sucrose or that the sucrose has an enhancing effect on starch mobility.  相似文献

Abnormal compressions and fusions of vertebral bodies are frequently observed in reared rainbow trout and could result from chronic and unbearable muscle pressures acting on the axial skeleton during intensive growth. Sustained swimming at moderate speeds was shown to induce many positive effects on growth and swimming performances in salmonids, but yet little is known about its effects on vertebral remodeling processes and related hormonal regulation. Rainbow trout were subjected to three different swimming speeds (0, 1.0 and 1.5 body length (BL) s− 1), starting one month after they were first fed (65 mm) and ending when they reached 260 mm in size (market-size of 275 g). At the end of the experiment, 20 trout were sampled in each lot (N = 60) and blood samples were taken. Vertebrae abnormalities were assessed by radiological examinations. Vertebrae from the middle axial region (V32–38) were selected to evaluate bone mineralization (BM) and total bone area (Tt-B.Ar.) on radiographed transverse sections (125 ± 10 μm). Assays were performed to evaluate mineral homeostasis (calcemia and phosphatemia), bone cell activities (alkaline phosphatase, ALP, and tartrate-resistant acid phosphatase, TRAP) and bone regulating hormones (calcitonin, CT and thyroid hormones, THs). Sustained exercise reduced the appearance of fused vertebrae, enhanced vertebral BM and decreased vertebral Tt-B.Ar., while it increased circulating CT and TH levels. No variations were observed on mineral homeostasis and bone cell activities. Increasing the swimming speed up to 1.5 BL s− 1 had positive effects on the vertebral skeleton, and therefore, seems to be a suitable approach to prevent aggravation of vertebral abnormalities in juvenile trout. The changes observed in vertebral features are interpreted as a compromise between the necessity to mobilize vertebral mineral ions in response to various physiological demands and to maintain vertebral strength against mechanical constraints.  相似文献

Diarrhoea due to enterotoxigenic Escherichia coli with fimbriae F4 (ETEC-F4) is an important problem in neonatal and just weaned piglets and hence for the pig farming industry. There is substantial evidence for a genetic basis for susceptibility to ETEC-F4 since not all piglets suffer from diarrhoea after an ETEC-F4 infection. It is assumed that the wild boar was originally ETEC-F4 resistant and that susceptibility towards ETEC arose after domestication. There are different phenotypes in the pig determined by which of the three existing F4 variants (F4ab, F4ac or F4ad) they are susceptible or resistant for. This suggests that several F4 receptors exist, expressed individually or in combination with each other on the brush border of the piglet’s small intestine. As such, the mucin-type glycoproteins (IMTGP) are described as F4ab/ac receptors, while the intestinal neutral glycospingolipid (IGLad) is proposed as an F4ad receptor. GP74 is a putative F4ab receptor. However, the specific genes that encode for the susceptibility are not yet known. In the past decades, linkage analyses revealed that the loci encoding for the receptor(s) for the two most frequent variants F4ab and F4ac were mapped to the 13th chromosome of the pig (Sus scrofa 13, SSC13). After fine mapping, the region of interest was mapped between two microsatellite markers, Sw207 and S0075, and interesting candidate genes surfaced. Numerous SNP analyses and a few expression studies on the three MUC-genes (MUC4, MUC13 and MUC20) and the transferrin receptor gene (TFRC) as well as on some other positional candidate genes have been performed in order to find the causative mutation for the ETEC-F4ab/ac receptor(s). However, until today, the exact mutation causing susceptibility to ETEC-F4 remains unknown.  相似文献

This report describes an uncommon variant of humpy-back syndrome associated with multiple rib fractures and multisystemic vasculitis in several nursing piglets and, for the first time, a skin disease in swine consistent with alopecia areata. After gelatinization in water, starch granules persist in swollen hydrated forms known as ghosts. Three potential mechanisms for ghost formation are tested. Proteins and lipids on the granule surface are found to be a determinant of ghost robustness, but not ghost formation. Proteins inside pre-made maize or wheat starch ghosts are degraded extensively by proteases without any apparent change in ghost properties, making an internal protein cross-linking mechanism unlikely. Waxy maize mutants with a range of amylose contents have ghost integrities that correlate with (low) apparent amylose levels. It is hypothesized that ghost formation is due to cross-linking of polysaccharide chains within swollen granules, most likely involving double helices formed from polymer chains that become free to move following heat-induced granule swelling. The size and robustness of granule ghosts is proposed to be determined by the relative rates of swelling and cross-linking, modulated by surface non-polysaccharide components.  相似文献

Human immunodeficiency virus type 1 (HIV-1), the cause of human acquired immunodeficiency syndrome (AIDS), is a zoonotic infection of staggering proportions and social impact. Yet uncertainty persists regarding its natural reservoir. The virus most closely related to HIV-1 is a simian immunodeficiency virus (SIV) thus far identified only in captive members of the chimpanzee subspecies Pan troglodytes troglodytes. Here we report the detection of SIVcpz antibodies and nucleic acids in fecal samples from wild-living P. t. troglodytes apes in southern Cameroon, where prevalence rates in some communities reached 29 to 35%. By sequence analysis of endemic SIVcpz strains, we could trace the origins of pandemic (group M) and nonpandemic (group N) HIV-1 to distinct, geographically isolated chimpanzee communities. These findings establish P. t. troglodytes as a natural reservoir of HIV-1.  相似文献

Swine brucellosis is caused by the biovars 1, 2 and 3 of Brucella suis the identification of which up to now relies on microbiological tests lacking adequate specificity together with time consuming and expensive molecular procedures. Based on sequence variation of the omp2b gene, we have developed a four primer set multiplex PCR assay that was tested for polymorphism analysis of B. suis biovars causing brucellosis in swine. The assay exploits the single nucleotide polymorphisms found in omp2b gene of B. suis reference biovars which are conserved in 43 B. suis field isolates from different geographic origins and hosts. Three specific amplification patterns (S1, S2 and S3) were obtained for reference strains of B. suis biovars 1, 2 and 3, respectively. However, some B. suis field isolates identified as biovars 2 or 3 according AMOS-PCR, PCR-RFLP of omp31 and omp2 genes and classical bacteriological methods, resulted also in S1 patterns, limiting the typing usefulness of the method.  相似文献

Experiments were carried out in plasticized wooden channels fed by a small creek in the Reserve des Laurentides, 80 km north of Quebec city. Channels were naturally colonized by invertebrates for 65 d before treatment. Treated channels were acidified in August with dilute H2SO4 only, or with acid plus a solution of Al sulfate (final concentration of 0.19 mg L?1). The control channel received creek water only (pH 6.3 to 6.9). The addition of Al had no effect on invertebrate density and biomass. After 73 d of acidification, invertebrate densities were only one third the number found in the control channel. Invertebrate hiomass was not different within channels, although biomass was generally higher in the two acidified channels. Difference in densities between acidified and non-acidified channels was attributed to lack of colonization and not to an increase in drift. Microtendipes, a large and resistant larva of Chironomidae constituted a large fraction of the hiomass, largely outweighing numerous very small larvae. Effects on the density were attributed to the direct effect of low pH and not to indirect action through food limitations.  相似文献
