The QB binding niche of photosystem II is also the binding site for many different herbicides. In order to understand the mode of binding of the herbicides, a 3-dimensional model of the binding niche was constructed. The model was based upon a comparison of the known structure of the QB binding niche in purple bacteria with sequence and mutant data of the D-1 protein of photosystem II. Plastoquinone builds up hydrogen bonds to phenylalanine 265 backbone amide nitrogen, to serine 264 hydroxyl, and to histidine 215 delta-1 nitrogen. In addition to these hydrogen bond donors and acceptors, herbicides can build up hydrogen bonds to backbone carbonyl of alamine 263 and to serine 268 hydroxyl. This is supported by binding data of inhibitors in Chlamydomonas reinhardtii chloroplasts of wild type and of five D-1 protein mutants (Ser264 Ala, Ala251 Val, Phe255Tyr, Val2191le, Leu275Phe).  相似文献

Treating the outer surfaces of isolated cuticles of Seville orange (Citrus aurantium L.) and pear (Pyrus communis L. cv. Bartlett) leaves with a number of nonionic (polyoxyethylene) surfactants increased their permeability to water by factors ranging from 4.1 to 14.7 and from 7.2 to 152.4, respectively. However, sodium dodecylsulphate, an anionic surfactant, had little effect on water permeability. In both species the major aliphatic constituents were n-alkanes, 1-alkanols and n-alkyl esters. None of the surfactant treatments altered the amounts or composition of waxes in the cuticles used for transport measurements. The reasons for the apparent absence of solubilization or dissolution of cuticular waxes by nonionic surfactants are discussed and a hypothesis is presented for the mechanism by which activator adjuvants may increase the permeability of plant cuticles.  相似文献

Growth of subarctic Scots pine (Pinus sylvestris L.) trees was investigated by a combination of process-based models and dendroecological approaches. Tree ring width indices were strongly autocorrelated and correlated with simulated photosynthetic production of the previous year and with organic matter N mineralization of the current year. An autoregressive model, with photosynthesis and N mineralization as external inputs, explained growth of the trees well. However, relationships for the period 1950-1992 differed significantly from relationships for the period 1876-1949; the slope of the regression of tree ring width index and photosynthesis was lower for the 1950-1992 period. Also, the autocorrelation structure of the data changed. First-order autocorrelation decreased and second-order autocorrelation increased from the earlier to the later period. This means that growth is becoming less sensitive to variations in photosynthetic production, whereas the relationships between growth and N mineralization are remaining fairly constant. We postulate that, although photosynthesis has increased in response to increasing CO2 concentrations, tree growth rate cannot parallel the increase in photosynthesis because potential growth rate is limited directly by temperature.  相似文献

Despite the steadily increasing area under protected agriculture there is a current lack of knowledge about the effects of the 30% black shading screen on microclimate and crop water requirements. Meteorological and lysimeter measurements inside a screenhouse planted with sweet pepper were compared to external reference data. Irrigation water use efficiency (IWUE) was calculated from yield records Y and water meter readings I applied. Shading reduced mean global radiation R G by more than 40%, and the screen transmissivity τ screen was shown to vary with solar elevation angle β. Wind speed inside the screenhouse u in was reduced by more than 50%. Crop water requirements ET c were 38% lower than estimates for an open field crop, suggesting a significant water saving potential when using screenhouses. However, the screen did not significantly modify maximum temperature T max and daily vapor pressure deficit. The FAO-Penman–Monteith approach based on meteorological measurements in the screenhouse accurately predicted daily crop evapotranspiration, and was in close agreement with lysimeter measurements. IWUE was relatively high (10.7 kg m−3 in 2004 and 13.5 kg m−3 in 2005), but additional research is required to quantify the effect of shading on yield as well as to determine the water saving potential of other commonly used screens. The AcrA/AcrB/TolC complex spans the inner and outer membranes of Escherichia coli and serves as its major drug-resistance pump. Driven by the proton motive force, it mediates the efflux of bile salts, detergents, organic solvents, and many structurally unrelated antibiotics. Here, we report a crystallographic structure of trimeric AcrB determined at 2.9 and 3.0 angstrom resolution in space groups that allow asymmetry of the monomers. This structure reveals three different monomer conformations representing consecutive states in a transport cycle. The structural data imply an alternating access mechanism and a novel peristaltic mode of drug transport by this type of transporter.  相似文献

Tropospheric ozone and black carbon (BC) contribute to both degraded air quality and global warming. We considered ~400 emission control measures to reduce these pollutants by using current technology and experience. We identified 14 measures targeting methane and BC emissions that reduce projected global mean warming ~0.5°C by 2050. This strategy avoids 0.7 to 4.7 million annual premature deaths from outdoor air pollution and increases annual crop yields by 30 to 135 million metric tons due to ozone reductions in 2030 and beyond. Benefits of methane emissions reductions are valued at $700 to $5000 per metric ton, which is well above typical marginal abatement costs (less than $250). The selected controls target different sources and influence climate on shorter time scales than those of carbon dioxide-reduction measures. Implementing both substantially reduces the risks of crossing the 2°C threshold.  相似文献

Excessive nutrient loading is a major threat to aquatic ecosystems worldwide that leads to profound changes in aquatic biodiversity and biogeochemical processes. Systematic quantitative assessment of functional ecosystem measures for river networks is, however, lacking, especially at continental scales. Here, we narrow this gap by means of a pan-European field experiment on a fundamental ecosystem process--leaf-litter breakdown--in 100 streams across a greater than 1000-fold nutrient gradient. Dramatically slowed breakdown at both extremes of the gradient indicated strong nutrient limitation in unaffected systems, potential for strong stimulation in moderately altered systems, and inhibition in highly polluted streams. This large-scale response pattern emphasizes the need to complement established structural approaches (such as water chemistry, hydrogeomorphology, and biological diversity metrics) with functional measures (such as litter-breakdown rate, whole-system metabolism, and nutrient spiraling) for assessing ecosystem health.  相似文献

Entanglement is the essential feature of quantum mechanics. Notably, observers of two or more entangled particles will find correlations in their measurement results that cannot be explained by classical statistics. To make it a useful resource, particularly for scalable long-distance quantum communication, the heralded generation of entanglement between distant massive quantum systems is necessary. We report on the creation and analysis of heralded entanglement between spins of two single rubidium-87 atoms trapped independently 20 meters apart. Our results illustrate the viability of an integral resource for quantum information science, as well as for fundamental tests of quantum mechanics.  相似文献
