Assessment of Water Quality in Association with Land use in the Tillamook Bay Watershed, Oregon, USA

The water quality in tributaries to Tillamook Bay, Oregon, frequently exceeds standards for fecal coliform bacteria (FCB) and temperature. FCB inputs to the bay have forced periodic closure of the oyster shellfish industry. In addition, impaired water quality may be contributing to reduced salmonid populations in the bay and its tributaries through reduction in the quality of the habitat. Because of these concerns, the Tillamook Bay National Estuary Project (TBNEP) conducted several characterization studies and a long-term water quality monitoring program for the tributary rivers. This paper summarizes data collected to date within these efforts, including storm-based data on FCB and total suspended solids (TSS), and bimonthly data on nutrient concentrations in selected rivers. Monitoring data from 1996 to 2002 are summarized. Results for FCB and TSS are reported by storm, which are classified according to season and precipitation patterns in order to minimize intra-annual variability. There are not indications of large changes in water quality throughout the period of record, but it is too early in the program for trends analysis. Storms that exhibited the highest FCB concentrations tended to be those that occurred during fall and/or those that were preceded by relatively dry conditions and included high rainfall intensity. Implementation of storm-based monitoring and classification of storms according to season effectively reduces the large variability inherent in the FCB monitoring data, thereby facilitating future trends analysis. Continued storm-based monitoring of FCB and TSS, and also continued collection of rainfall and river discharge data, will provide the database that will be needed to determine to what extent on-the-ground remediation actions and best management practices (BMPs) within the Tillamook Basin are having their desired effects. 相似文献

Phytochemicals and antioxidant activity of milled fractions of different wheat varieties 总被引:7,自引:0,他引:7
The health-promoting effects of whole-grain consumption have been attributed in part to their unique phytochemical contents and profiles that complement those found in fruits and vegetables. Wheat is an important component of the human diet; however, little is known about the phytochemical profiles and total antioxidant activities of milled fractions of different wheat varieties. The objectives of this study were to investigate the distribution of phytochemicals (total phenolics, flavonoids, ferulic acid, and carotenoids) and to determine hydrophilic and lipophilic antioxidant activity in milled fractions (endosperm and bran/germ) of three different wheat varieties, two of which were grown in two environments. Grain samples of each of the wheat varieties were milled into endosperm and bran/germ fractions. Each fraction was extracted and analyzed for total phenolics, ferulic acid, flavonoids, carotenoid contents, and hydrophilic and lipophilic antioxidant activities. Total phenolic content of bran/germ fractions (2867-3120 micromol of gallic acid equiv/100 g) was 15-18-fold higher (p < 0.01) than that of respective endosperm fractions. Ferulic acid content ranged from 1005 to 1130 micromol/100 g in bran/germ fractions and from 15 to 21 micromol/100 g in the endosperm fractions. The bran/germ fraction flavonoid content was 740-940 micromol of catechin equiv/100 g. On average, bran/germ fractions of wheat had 4-fold more lutein, 12-fold more zeaxanthin, and 2-fold more beta-cryptoxanthin than the endosperm fractions. Hydrophilic antioxidant activity of bran/germ samples (7.1-16.4 micromol of vitamin C equiv/g) was 13-27-fold higher than that of the respective endosperm samples. Similarly, lipophilic antioxidant activity was 28-89-fold higher in the bran/germ fractions (1785-4669 nmol of vitamin E equiv/g). Hydrophilic antioxidant activity contribution to the total antioxidant activity (hydrophilic + lipophilic) was >80%. In whole-wheat flour, the bran/germ fraction contributed 83% of the total phenolic content, 79% of the total flavonoid content, 51% of the total lutein, 78% of the total zeaxanthin, 42% of the total beta-cryptoxanthin, 85% of the total hydrophilic antioxidant activity, and 94% of the total lipophilic antioxidant activity. Our results showed that different milled fractions of wheat have different profiles of both hydrophilic and lipophilic phytochemicals. Reduction of ferric myoglobin (metmyoglobin, MetMb) to its ferrous form is important for maintaining fresh meat color because only reduced myoglobin can bind oxygen to form the consumer-preferred cherry red color in fresh meat. The objective of this study was to characterize an apparent mitochondria electron transport chain (ETC)-linked pathway for MetMb reduction in vitro. MetMb was reduced in the presence of mitochondria and succinate (p < 0.05); mitochondria or succinate alone did not facilitate MetMb reduction relative to controls (p > 0.05). Flushing samples with oxygen greatly decreased MetMb reduction, while flushing with argon increased MetMb reduction when compared with controls (p < 0.05). ETC inhibitors were used to localize the site where electrons became available for MetMb reduction. MetMb reduction was increased by rotenone addition and decreased by malonic acid (p < 0.05); the reduction was completely abolished by additions of antimycin A or myxothiazol when compared with controls (p < 0.05). These results suggest that electrons become available for MetMb reduction at a site(s) between complex III and IV. Mitochondrial ETC-linked MetMb reduction increased with increased mitochondrial density and succinate concentration (p < 0.05); the greatest MetMb reduction was observed at pH 7.2 and 37 degrees C, and ETC-linked MetMb reducing activity decreased with time postmortem (p < 0.05). These results indicate that ETC-linked MetMb reduction exists but would be minimally active in postmortem muscles. 相似文献

Robust measurements of porous-medium tortuosity are one of the many components needed for accurate characterization and prediction of fluid flow and contaminant transport in the subsurface. A gas-phase diffusive tracer-test method is evaluated for the in-situ measurement of tortuosity in the vadose zone. This technique presents an alternative to employing widely-used correlations to estimate tortuosity. A small-scale field study was conducted using a single well and a non-reactive gas-phase tracer (sulfur hexafluoride; SF6). Gas samples were collected from the injection point periodically after tracer injection into the soil matrix. An effective radius of influence of 50 cm was determined for the tests. An analytical solution was calibrated to the measured temporal concentration distribution, producing a fitted value for tortuosity. The value obtained from the tracer tests was compared to values estimated with several widely-used correlations. The value obtained from the tracer tests was generally larger than the values estimated with the correlations, which spanned a relatively wide range. The tracer-test method provides a means by which to determine in-situ measurements of tortuosity, allowing for better characterization of contaminant transport in the vadose zone. 相似文献

In this study three different approaches were employed to identify key odorants in Sauvignon blanc wines. First, the concentrations of the odorants were compared to their respective aroma detection thresholds. The resulting odor activity values (OAV) were transformed into a normalized and weighted measure that allows the aroma profiles of different wines to be compared and the contribution of a single aroma in a complex mixture to be evaluated. Based on their OAV, 3-mercaptohexanol and 3-mercaptohexyl acetate were the two most important aroma compounds in many Marlborough Sauvignon blanc wines. Due to limitations with the OAV approach, the study was extended to include aroma extract dilution analysis (AEDA), which revealed that β-damascenone, together with the varietal thiols, esters, and higher alcohols, are key odorants in Sauvignon blanc wines. The final approach undertaken was aroma reconstitution and omission tests using a deodorized wine base and the creation of a model Marlborough Sauvignon blanc. Single compounds and groups of compounds were omitted from the model to study their impact on the sensory properties of the model wine. Reconstitution and omission confirmed that varietal thiols, esters, terpenes, and β-damascenone are all important contributors to Sauvignon blanc aroma. Bahia grass (Paspalum notatum), johnson grass (Sorghum halpense) and switchgrass (Panicum virginatum) werecompared for their ability to accumulate 137Csand 90Sr from three different contaminated soilsin the presence and absence of either sphagnum peator poultry litter amendments. Thegreatest increases in plant biomass, and radionuclideaccumulation were observed with poultry litter foreach of the three grass species. Johnson grass hadgreater aboveground plant biomass, activity of137Cs and 90Sr in plant tissue, %accumulation of 137Cs or 90Sr from soil andbioconcentration ratio in each soil amendment, at eachharvest compared to bahia and switchgrass. Thegreatest accumulation of 137Cs and 90Sr wasmeasured in johnson grass grown in soil that wasamended with poultry litter. In tropical ecosystems, access to nitrogen (N) and phosphorus (P) limits the decomposition rate of leaf-litter. Leaf-litter ants are abundant in this microhabitat and present a wide variety of diets. Our aim was to study the response of various ant trophic groups to an increased nutrient availability which boosts the decomposition of their habitat and selectively affects the abundance of their prey.A 6-month nutrient addition experiment (CN, CNP) was performed in a tropical montane forest of the Ecuadorian Andes. The density of ants, of other predators (e.g. arachnids, beetles) and of their potential prey (mesofauna, ranging from 0.1 to 2 mm) was measured in treatments and control plots.The litter volume in fertilized plots decreased significantly. Collembola and total mesofauna density were enhanced by the CNP addition. Ants responded differentially according to their trophic group: despite increased prey availability, predatory species in general and collembolan hunters in particular were negatively affected by both treatments. Other ant trophic group densities did not change. By contrast, the density of Dermaptera increased with the treatments. A complementary isotopic approach allowed us to trace carbon fluxes through the food web.Our results suggest that the nutrient input enhanced the litter decomposition rate, leading to reduction of habitat size. They also suggest that predatory ants in tropical leaf-litter food webs are limited by habitat size rather than by prey availability, and that these ants are more affected by habitat loss than their prey, other ant trophic groups and other macrofauna taxa. 相似文献

Metagenomic analyses can provide extensive information on the structure, composition, and predicted gene functions of diverse environmental microbial assemblages. Each environment presents its own unique challenges to metagenomic investigation and requires a specifically designed approach to accommodate physicochemical and biotic factors unique to each environment that can pose technical hurdles and/or bias the metagenomic analyses. In particular, soils harbor an exceptional diversity of prokaryotes that are largely undescribed beyond the level of ribotype and are a potentially vast resource for natural product discovery. The successful application of a soil metagenomic approach depends on selecting the appropriate DNA extraction, purification, and if necessary, cloning methods for the intended downstream analyses. The most important technical considerations in a metagenomic study include obtaining a sufficient yield of high-purity DNA representing the targeted microorganisms within an environmental sample or enrichment and (if required) constructing a metagenomic library in a suitable vector and host. Size does matter in the context of the average insert size within a clone library or the sequence read length for a high-throughput sequencing approach. It is also imperative to select the appropriate metagenomic screening strategy to address the specific question(s) of interest, which should drive the selection of methods used in the earlier stages of a metagenomic project (e.g., DNA size, to clone or not to clone). Here, we present both the promising and problematic nature of soil metagenomics and discuss the factors that should be considered when selecting soil sampling, DNA extraction, purification, and cloning methods to implement based on the ultimate study objectives. 相似文献

Estimating the amount of water on leaf surfaces is an increasing concern for remote sensing and hydrology. Measuring the magnitude and spatial extent of leaf wetness events will provide useful information for water and energy balance modeling and remote sensing. As part of the Soil Moisture Experiments 2005 (SMEX05), the temporal and spatial characterization of leaf wetness over a heterogeneous agricultural domain was investigated. Leaf wetness sensors and physical measurements were collected from 15 June to 3 July 2005 in and around the Walnut Creek Watershed near Ames, Iowa, USA. Comparison of the results of the in situ leaf wetness sensor measurements and the physical sampling revealed a moderate correlation for both corn (Zea mays L.) and soybeans (Glycine max Merr.). Regression equations were developed to estimate leaf wetness quantity from these leaf wetness sensors and combined with a vegetation leaf area index map to produce a spatial leaf wetness product hourly during the experiment with an error of approximately 0.05 kg/(m2 LAI). Using this strategy, future efforts in spatial hydrologic modeling and remote sensing would be able to incorporate quantitative estimates of leaf wetness amount in watershed scale studies using only in situ measurements. 相似文献

In managed landscapes, habitat structure is frequently manipulated through the creation of features such as tracks, hedges, and waterways. If predator and prey activity are concentrated around these features, levels of predation may be elevated in these landscapes. This issue is of particular importance when habitat structures are used to attract species of conservation concern. For example, the installation of linear waterways in wet grasslands is a common form of habitat management to benefit breeding waders and wader nests and foraging chicks tend to be aggregated around wet features. If predator activity is also focused around these features, and if their linearity increases the probability of prey being located, then the conservation benefits of this management technique may be eliminated. We explore predator movement in relation to the structure and complexity of linear wet features within a lowland wet grassland landscape. We examine patterns of nest and chick predation in lapwing (Vanellus vanellus) at the whole-site, between-field and within-field scales. Mammalian predators were responsible for the majority of nest predation. However, we found no evidence that mammalian predators used linear wet features disproportionately within the landscape, or that wet feature distribution influenced the probability of nest or chick predation. At the whole-site scale, nest predation rates were significantly higher in areas with greater predator presence and lowest where the number of breeding neighbours was high. Thus, predation levels were influenced by large-scale patterns of predator presence and lapwing density but not by the use of linear wet features as a habitat management tool. Managing predator impacts is therefore likely to require empirical assessments of local predator distribution and abundance in order to target measures effectively. 相似文献
