The effect of three mutant genes affecting anthocyanin biosynthesis – anthocyaninless of Hoffmann (ah), anthocyaninwithout (aw) and baby lea syndrome (bls) – on tomato germination capacities at 24 ° C was investigated. The study was performed on nine anthocyanin containing lines – Apedice, Apeca, Ailsa Craig, Monfavet 167, Monfavet 168, Por, Piernita, VF 36 and VFNT cherry and 13 isogenic/near isogenic lines (IL/NIL) from them: 4 differing for gene ah, 3 differing for gene aw and 6 differing for gene bls). In the majority of the anthocyaninless IL/NILs germination began earlier than that in the wild type lines. Significant differences were observed in the time to 50% germination between the anthocyanin-containing and the anthocyaninless IL/NILs except for the most rapidly germinating line VFNT cherry. Seed weight, water uptake, effect of testa removal and presence or absence of maternal effects in F1 germination responses were investigated in order to elucidate the causes of the more rapid germination in the anthocyaninless IL/NILs. The increased amount of water uptake by the anthocyaninless lines, the shorter treatment time necessary for their testa removal, the important maternal effects in the genetic variability of the time to 50% germination and the differences in germination capacities between the wild type and the mutant IL/NILs after testa removal, indicated that the seed coat and the endosperm had to be affected by the effects of the three genes in a way that caused enhanced germination. Riemerella columbina (RC) and Riemerella anatipestifer (RA) belong to the genus Riemerella within the family Flavobacteriaceae. While RA is a well-described pathogen of waterfowl and other avian species, only little is known about RC. Previous work reporting the isolation of RC from internal organs of clinically diseased pigeons suggested a potential pathogenic role in this avian species. In this study we examined pharyngeal swabs collected from pigeons and found RC to be widely distributed also among healthy birds. Further characterization of 81 RC-isolates revealed several atypical strains, which differed from all previously described RC-isolates by the lack of aesculin-hydrolysis activity (17 isolates) or by expression of yellow or orange pigmentation (6 isolates). Sequence analysis of the 16S rRNA and outer membrane protein A (ompA) gene supported the affiliation of these strains to the species RC. Aesculin-hydrolysis negative isolates were found to be biochemically indistinguishable from RA. We demonstrated that bacterial fingerprinting using matrix assisted laser desorption/ionisation–time of flight mass spectrometry (MALDI–TOF MS) analysis is useful for the identification and differentiation of RC and RA.  相似文献

ObjectiveTo determine the effect of intravenous ketamine on the minimum alveolar concentration of sevoflurane needed to block autonomic response (MACBAR) to a noxious stimulus in dogs.Study designRandomized, crossover, prospective design.AnimalsEight, healthy, adult male, mixed-breed dogs, weighing 11.2–16.1 kg.MethodsDogs were anesthetized with sevoflurane on two occasions, 1 week apart, and baseline MACBAR (B-MACBAR) was determined on each occasion. MACBAR was defined as the mean of the end-tidal sevoflurane concentrations that prevented and allowed an increase (≥15%) in heart rate or invasive mean arterial pressure in response to a noxious electrical stimulus (50 V, 50 Hz, 10 ms). Dogs then randomly received either a low-dose (LDS) or high-dose series (HDS) of ketamine, and treatment MACBAR (T-MACBAR) was determined. The LDS had an initial loading dose (LD) of 0.5 mg kg?1 and constant rate infusion (CRI) at 6.25 μg kg?1 minute?1, followed, after T-MACBAR determination, by a second LD (1 mg kg?1) and CRI (12.5 μg kg?1 minute?1). The HDS had an initial LD (2 mg kg?1) and CRI (25 μg kg?1 minute?1) followed by a second LD (3 mg kg?1) and CRI (50 μg kg?1 minute?1). Data were analyzed with a mixed-model anova and are presented as LSM ± SEM.ResultsThe B-MACBAR was not significantly different between treatments. Ketamine at 12.5, 25, and 50 μg kg?1 minute?1 decreased sevoflurane MACBAR, and the maximal decrease (22%) occurred at 12.5 μg kg?1 minute?1. The percentage change in MACBAR was not correlated with either the log plasma ketamine or norketamine concentration.Conclusions and clinical relevanceKetamine at clinically relevant doses of 12.5, 25, and 50 μg kg?1 minute?1 decreased sevoflurane MACBAR, although the reduction was neither dose-dependent nor linear.  相似文献

To better understand the effects of partial cutting on arboreal lichen production within woodland caribou (Rangifer tarandus caribou) habitat, lichen was hand picked from 1228 branches on 307 subalpine fir (Abies lasiocarpa) trees in Idaho (ID), and in British Columbia (BC). Lichen biomass from partially cut stands was compared with biomass on trees from adjacent uncut stands at each site. Arboreal lichen biomass did not differ significantly between uncut and partially cut stands. Total number of branches per tree did not differ significantly between uncut and partially cut stands. Live branches had more lichen than dead branches. Species composition of arboreal lichen changed in partially cut stands compared with adjacent uncut stands. The ratio of live to dead branches was substantially different within the BC partial cut.  相似文献

This report describes 2 cases of spontaneous aortic dissecting hematoma in young Border Collie and Border Collie crossbred dogs. Histology was performed in one of the cases involving an unusual splitting of the elastin present within the wall of the aorta, consistent with elastin dysplasia as described in Marfan syndrome in humans. The first case involved a young purebred Border Collie that died suddenly and the second case involved a Border Collie crossbred dog that died after a 1-month history of seizures. Gross lesions included pericardial tamponade with dissection of the ascending aorta in the former case and thoracic cavity hemorrhage, mediastinal hematoma, and aortic dissection in the latter. Histologic lesions in the case of the Border Collie crossbred dog included a dissecting hematoma of the ascending aorta with elastin dysplasia and right axillary arterial intimal proliferation.  相似文献

Changes in pathotype frequencies due to sexual reproduction during summer were assessed in barley plots inoculated at the start of the growing season with powdery mildew isolates of the pathotype GL1 (Va22Vh) which belonged to a clonal lineage frequent in Northern France in 1992–1996 and was absent or rare in the local population at the time of the study. The capacity for recombination among isolates belonging to the pathotype GL1 was confirmed by performing crosses in the greenhouse. The field experiment was repeated over two years, with two field plots inoculated with either a single isolate (one mating type) or four isolates (two mating types). Following artificial inoculation, the frequency of GL1 increased to between 40% and 80% of the total conidia population at the end of the asexual epidemics before summer. When only one isolate (one mating type) of the pathotype GL1 was present, the frequency of the GL1 pathotype decreased to 4–5% of the ascospore population, following sexual recombination between the inoculated isolate and the local natural population. When isolates of both mating types of GL1 were inoculated, however, the GL1 pathotype remained dominant (28%) in the ascospore population. A pathotype dominant at the end of the summer might possibly have over-summered through asexual reproduction, or alternatively it may originate from sexual reproduction. The observed GL1 frequencies and numbers of virulences per isolate (complexity) observed on the field plots were compared to those calculated with a model assuming random or non-random mating.  相似文献

Venturia nashicola is the causal agent of scab, a fungal disease affecting Asian pears. The Japanese pear cv. ‘Kousui’ is highly susceptible to the race 1 of this fungus whereas the cv. ‘Kinchaku’ and the non-host European pear cv. ‘Flemish Beauty’ are resistant. The aim of this work is to investigate the role of polygalacturonase-inhibiting proteins (PGIPs) of pear during the interactions with V. nashicola leading to susceptibility or resistance. PGIP protein was detected from immature fruit of Kousui and Kinchaku. It showed a molecular mass of 42 kDa that shifted to 35 kDa after chemical deglycosylation. The gene pgip was amplified by PCR using genomic DNA and/or cDNA from young leaves of Kousui, Kinchaku, and European pear cvs. Flemish Beauty, ‘Bartlett’, and an Asian wild pear strain ‘Mamenashi 12’, then sequenced after sub-cloning. Some conserved variations were identified in the sequence indicating that gene family also exists in pgip of Japanese pear and confirmed by Southern blot analysis. The expression of PGIP was studied in scab-inoculated leaves of the susceptible Kousui and the resistant Kinchaku and Flemish Beauty. pgip Gene and its encoding protein were highly and rapidly activated in these resistant plants. In addition, PGIP extracts derived from Kinchaku and Flemish Beauty partially inhibited the activity of polygalacturonase (PG) from V. nashicola suggesting a possible role of PGIP in limiting fungal growth frequently observed in these resistant cultivars.  相似文献

OBJECTIVE: To determine whether antibodies against Sarcocystis neurona could be detected in CSF from clinically normal neonatal (2 to 7 days old) and young (2 to 3 months old) foals. DESIGN: Prospective study. ANIMALS: 15 clinically normal neonatal Thoroughbred foals. PROCEDURE: Serum and CSF samples were obtained from foals at 2 to 7 days of age and tested for antibodies against S. neurona by means of western blotting. Serum samples from the mares were also tested for antibodies against S. neurona. Additional CSF and blood samples were obtained from 5 foals between 13 and 41 days after birth and between 62 and 90 days after birth. RESULTS: Antibodies against S. neurona were detected in serum from 13 mares and their foals; antibodies against S. neurona were detected in CSF from 12 of these 13 foals. Degree of immunoreactivity in serum and CSF decreased over time, and antibodies against S. neurona were no longer detected in CSF from 2 foals 83 and 84 days after birth. However, antibodies could still be detected in CSF from the other 3 foals between 62 and 90 days after birth. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicate that antibodies against S. neurona can be detected in CSF from clinically normal neonatal (2 to 7 days old) foals born to seropositive mares. This suggests that western blotting of CSF cannot be reliably used to diagnose equine protozoal myeloencephalitis in foals < 3 months of age born to seropositive mares.  相似文献
