Comparison of two fully sequenced genomes of Buchnera aphidicola, the obligate endosymbionts of aphids, reveals the most extreme genome stability to date: no chromosome rearrangements or gene acquisitions have occurred in the past 50 to 70 million years, despite substantial sequence evolution and the inactivation and loss of individual genes. In contrast, the genomes of their closest free-living relatives, Escherichia coli and Salmonella spp., are more than 2000-fold more labile in content and gene order. The genomic stasis of B. aphidicola, likely attributable to the loss of phages, repeated sequences, and recA, indicates that B. aphidicola is no longer a source of ecological innovation for its hosts.  相似文献

The derogation to use a percentage of cheaper non-organic feeds in organic livestock diets for herbivores expired from January 2008. (In Norway, a maximum of 15% conventional feedstuffs per year was allowed until 24 August 2005, 5% in the interim.) This study aimed to assess changes in resource use and financial impacts for organic dairy herds of the 100% organic feeding rule (compared with 85% organic feeds) using a two-stage stochastic programming modelling framework. In this study, the objective function was to maximise expected net income. Two organic dairy farms in the lowlands of southern Norway, both having a milk quota of 100,000 l but with varying farmland availability (37 ha vs. 20 ha), were examined. Furthermore, situations with and without possibilities to buy organic silage were studied. Milk production per cow was highest on the farm with the most strict land constraint. For farmers that fully utilise the non-organic feed allowance, the more expensive organic feeds determine the marginal feed cost. Hence, a removal of the non-organic feed allowance will not influence optimal farm practice (provided that marginal milk production is still profitable). A switch from conventional to organic concentrates as the only adjustment was found on the 37 ha farm and also on the 20 ha farm if there was no possibility to buy some organic silage. On the 20 ha farm with possibilities to buy organic silage, it turned out to be unprofitable to keep all the cows. Fewer cows resulted in several other changes in the model solution such as a lower milk yield per cow, less milk produced in total and more work off-farm. The financial gain from carrying out the optimal management changes was, however, very small. In all cases studied, the introduction of 100% organic feeds resulted in an economic loss of NOK 19,200–23,600 (or 7–12% of the expected net income with 85% organic feeds). Twenty-three lactating dairy cows were switched from a standard diet to a ration of alfalfa/grass silage and 1 kg rapeseed cake supplemented with raw potatoes and fodder beets allowed on a semi-restricted basis. The effects of feed choice on urinary markers for nitrogen metabolism were evaluated by the Stepwise regression technique. Each kilogram dry matter of potatoes and fodder beets lowered milk urea concentration and also the urinary ratio urea/creatinine to a similar extent, but allantoin and fecal results suggested larger hindgut fermentation and hence lower ruminal microbial protein production with potatoes than with fodder beets. The variation for concentration of urinary constituents associated with animal, sampling day and sampling time suggests multiple sampling within a day as preferable for spot sampling. It is concluded that the reluctance to potatoes by some cows may cause initial difficulties when fed to high yielding cows naïve to the feed.  相似文献

Overall, arable soils in Sweden are currently generally close to phosphorus (P) balance, but excessive P accumulation has occurred on animal fur farms, i.e., those rearing mink (Mustela vison) and foxes (Alopex lagopus and Vulpes vulpes). Manure P from these farms has sometimes regarded as sparingly soluble. Laboratory lysimeter topsoil trials with simulated rain demonstrated that potential leaching of P in dissolved reactive form (DRP) can be very high, even for heavy clay (50%–65%) soils. The Swedish/Norwegian soil test P‐AL (soil P extracted with acid NH4 lactate, AL) proved useful as a potential indicator of DRP leaching risk (regression coefficient [R2] = 0.89) from fur farms. The upper 5‐cm soil layer, with 190% higher (median) soil P status than the 5–20 cm layer, was the major source of potential DRP leaching through soil columns at the site, despite having been under grass or green fallow for the past 8 y. In percolate from topsoil lysimeters, DRP concentration increased by 0.29 mg L–1 after the long‐term manure application but only by 0.14 mg L–1 after the single slurry application when compared to no addition of slurry. Therefore, the build‐up to a high soil P status due to the long‐term application of mink manure was more important than a single application of pig slurry at a rate corresponding to 22 kg P ha–1 with respect to soil leachate DRP losses in this lysimeter study. The study stresses the importance of precision farming, in which the amount of slurry‐P applied is based on testing the already existing soil P content.  相似文献

Plant availability and risk for leaching and/or runoff losses of phosphorus (P) from soils depend among others on P concentration in the soil solution. Water-soluble P in soil measures soil solution P concentration. The aim of this study was to understand the effect of wheat residue char (biochar) addition on water-soluble P concentration in a wide range of biochar-amended soils. Eleven agricultural fields representing dominant soil texture classes of Swedish agricultural lands were chosen. Concentrations of water-soluble P in the soils and in biochar were measured prior to biochar incorporation to soils in the laboratory. Experiments with three dominant soil textures—silt loam, clay loam, and an intermediate loam soil with different rates of biochar addition (i.e., 0.5, 1, 2, and 4 %; w/w) showed that the highest concentration of water-soluble P was achieved at an application rate of 1 %. At higher application rates, P concentrations decreased which coincided with a pH increase of 0.3–0.7 units. When the 11 soils were amended with 1 % (w/w) biochar, water-soluble P concentrations increased in most of the soils ranging from 11 to 253 %. However, much of the water-soluble P added through the biochar was retained (33–100 %). We concluded that wheat residue char can act as a source of soluble P, and low and high additions of biochar can have different effects on soil solution P concentration due to possible reactions with Ca and Mg added with biochar.  相似文献

Florfenicol (FFC) as a chloramphenicol’s derivative is a special broad-spectrum antibiotic that was used in veterinary clinics. In the present study, we investigated the effect of different doses of FFC on the humoral immune response of broiler chickens to Newcastle disease virus (NDV) vaccine under the impact of E. coli infection. In addition, the expression of the interferon-inducible genes (IRF7, 2′-5′OAS and Mx1) were analyzed in the spleen tissue of these chickens using quantitative real-time PCR (qRT-PCR). The non-treated group with FFC and non-infected with E. coli had the highest immune responses against NDV compared with the FFC treated groups. In the case of E. coli infection, the group treated with FFC (30 mg/Kg BWt) showed lower NDV HI and IgG ELISA Ab levels compared to the group treated with FFC (60 mg/Kg BWt). A dose dependent up-regulation was observed in the level of the interferon-alpha pathway related genes (IRF7 and 2′-5′OAS) in the FFC treated groups compared to the non-treated group. At the slaughter time, the numbers of adipocyte in the bone marrow were significantly higher with moderate atrophy of the hematopoietic lineages in the FFC treated birds compared to the non-treated birds. These results indicated that this FFC dosage dependent increase in the humoral immune responses against NDV vaccine could be attributed to its efficient therapeutic effect on the E. coli infection. However, the increase in the FFC dosage can negatively but temporarily affect the chicken body weights. Additionally, it can exert up regulation effect on the chicken innate immune response with moderate hypoplasia of the bone marrow cells.  相似文献

Abstract

The transmission of pathogens is a common consequence of animal food production. Marine salmon farms and their processing facilities can serve as sources of virulent fish pathogens; our study is the first to confirm the broadcast of a live fish pathogen from a farmed salmon processing facility into the marine waters of Canada's Pacific coast. We found live salmon lice Lepeophtheirus salmonis, mucus, and fish tissue in effluent from the processing facility. Sea lice transmitted from this source may pose a threat to wild salmon populations, and the release of untreated offal, including blood water, is of considerable concern. Further research is needed to quantify the extent to which processing facilities release sea lice and to determine whether more virulent fish pathogens are present in effluent. These data underscore the need for fish farming nations to develop mandatory biosecurity programs to ensure that farmed salmon processing facilities will prevent the broadcast of infectious fish pathogens into wild fish habitat.

Samples collected from 791 wild animals (Canada geese, roe deer, hares, moose, wild boar and gulls) shot during hunting were examined for verocytotoxin-producing Escherichia coli (VTEC) O157, and thermophilic Campylobacter and Salmonella species. With the exception of one positive isolate from a wild boar, VTEC O157 was not isolated from any of the animals. Salmonella species were isolated only from the gulls, of which 4 per cent were estimated to be positive. Thermophilic Campylobacter species were commonly isolated from all the species except deer.  相似文献
